Evaluation of the Hypothalamic Kisspeptin System during the Attainment of Puberty in Gilts

Background: Kisspeptin has been demonstrated to affect the attainment of puberty in multiple species, presumably through its actions on GnRH and LH. It has previously been found there is an increase in expression of hypothalamic in the rat and the monkey coincident with puberty. Whether a similar phenomenon occurs in the pig remains to be determined. The objectives of the current experiments were to determine 1) Plasma concentrations of estradiol-17β;2) Anterior pituitary gland concentrations of LH, IGF-I, kisspeptin, and anterior pituitary gland expression of LH-β and GnRH receptor;3) Mediobasal hypothalamus expression of Kiss1 and concentrations of kisspeptin during the peripubertal period in the gilt. Methods: Two experiments were performed, each with 25 crossbred gilts (151 d, 105 kg). Gilts were relocated and exposed to a mature boar beginning on d 1 and continuing throughout the experiments to naturally induce puberty. Gilts that stood immobile within 24 hours of slaughter were considered to have attained puberty. Plasma samples were collected on d 1, 3, and 7 of the first experiment and d 2, 4, 6, and 8 of the second experiment. Gilts were slaughtered on d 7 (experiment 1) or d 8 (experiment 2), when mediobasal hypothalamus (MBH), anterior pituitary glands (AP), and blood were collected. Relative expression of MBH Kiss1 and β-actin and AP GnRH receptor, LH-β, was determined using real-time reverse transcriptase PCR. Fold changes in relative expression were determined using the Relative Expression Software Tool. Hypothalamic and AP content of kisspeptin were determined by RIA and differences were determined using the GLIMMIX procedure of SAS. Results: Relative expression of Kiss1 was increased (P = 0.005) 2.2 fold in the gilts that had attained puberty in the first experiment, however, those that had attained puberty in the second were not different (P > 0.05) from gilts that had not. Volcano video data characterized and classified using computer vision and machine learning algorithms

Can we omit systematic biopsies in patients undergoing MRI fusion-targeted prostate biopsies?

Corneal epithelial defects following vitreoretinal surgery:incidence and outcomes from the DISCOVER study

AIM:To investigate the incidence,risk factors,clinical course,and outcomes of corneal epithelial defects(CED)following vitreoretinal surgery in a prospective study ***:This was a post-hoc analysis of all participants in DISCOVER intraoperative optical coherence tomography *** with CED 1 d after surgery without intraoperative corneal debridement was defined as the postoperative CED *** who underwent intraoperative debridement were defined as intraoperative debridement *** were matched 2:1 with controls(eyes without postoperative CED)for comparative *** primary outcomes were the incidence of CED on postoperative day one and the incidence of required intraoperative *** outcomes included time to defect closure,delayed healing(>2 wk),visual acuity(VA)and presence of scarring at one year and cornea ***:This study included 856 eyes that underwent vitreoretinal *** corneal debridement was performed to 61(7.1%)subjects and postoperative CED developed spontaneously in 94(11.0%)*** factors associated with postoperative CED included prolonged surgical duration(P=0.003),diabetes mellitus(P=0.04),postoperative ocular hypotension(Pdefect closure time(>2 wk)was associated with corneal scar formation at the end of the 1 y in all epithelial defect subjects(Pdergoing vitrectomy was 1.8%.CONCLUSION:Prolonged duration of surgery is the strongest factor associated with both intraoperative debridement and spontaneous postoperative *** defect closure is associated with a greater risk of corneal scarring at one *** overall rate of corneal scarring following vitrectomy is low at<2%.

Advanced bioactive glue tethering Lubricin/PRG4 to promote integrated healing of avascular meniscus tears

Meniscus injuries are extremely common with approximately one million patients undergoing surgical treatment annually in the U.S. alone, but no regenerative therapy exist. Previously, we showed that controlled applications of connective tissue growth factor (CTGF) and transforming growth factor beta 3 (TGFβ3) via fibrin-based bio-glue facilitate meniscus healing by inducing recruitment and stepwise differentiation of synovial mesenchymal stem/progenitor cells. Here, we first explored the potential of genipin, a natural crosslinker, to enhance fibrin-based glue’s mechanical and degradation properties. In parallel, we identified the harmful effects of lubricin on meniscus healing and investigated the mechanism of lubricin deposition on the injured meniscus surface. We found that the pre-deposition of hyaluronic acid (HA) on the torn meniscus surface mediates lubricin deposition. Then we implemented chemical modifications with heparin conjugation and CD44 on our bioactive glue to achieve strong initial bonding and integration of lubricin pre-coated meniscal tissues. Our data suggested that heparin conjugation significantly enhances lubricin-coated meniscal tissues. Similarly, CD44, exhibiting a strong binding affinity to lubricin and hyaluronic acid (HA), further improved the integrated healing of HA/lubricin pre-coated meniscus injuries. These findings may represent an important foundation for developing a translational bio-active glue guiding the regenerative healing of meniscus injuries.
