The synthetic biology toolkit for baker’s yeast,Saccharomyces cerevisiae,includes extensive genome engineering toolkits and parts ***,with the increasing complexity of engineering tasks and versatile applications of this model eukaryote,there is a continued interest to expand and diversify the rational engineering capabilities in this chassis by FAIR(findable,accessible,interoperable,and reproducible)*** this study,we designed and characterised 41 synthetic guide RNA sequences to expand the CRISPR-based genome engineering capabilities for easy and efficient replacement of genomically encoded ***,we characterize in high temporal resolution 20 native promoters and 18 terminators using fluorescein and LUdOX CL-X as references for GFP expression and Od600 measurements,***,all data and reported analysis is provided in a publicly accessible jupyter notebook providing a tool for researchers with low-coding skills to further explore the generated data as well as a template for researchers to write their own *** expect the data,parts,and databases associated with this study to support a FAIR-compliant resource for further advancing the engineering of yeasts.

Renal tumor surgery places patients at increased risk for chronic kidney disease(CKd). Accurate quantification of kidney function changes before and after surgery is essential to determine the magnitude of decline attributable to an index procedure. Current literature, however, highlights heterogeneity and inconsistencies in measurement techniques thereby contributing to ambiguity amongst studies. Further efforts are necessary to standardize reporting of kidney function outcomes related to renal surgery.

Comparative susceptibility and immune responses of Asian and European honey bees to the American foulbrood pathogen, Paenibacillus larvae

American foulbrood (AFB) disease is caused by Paenibacillus larvae. Currently, this pathogen is widespread in the European honey bee— Apis mellifera. However, little is known about infectivity and pathogenicity of P. lan'ae in the Asiatic cavity-nesting honey bees, Apis cerana. Moreover, comparative knowledge of P. larvae infectivity and pathogenicity between both honey bee species is scarce. In this study, we examined susceptibility, larval mortality, survival rate and expression of genes encoding antimicrobial peptides (AMPs) including defensin, apidaecin, abaecin, and hymenoptaecin in A. mellifera and A. cerana when infected with P. larvae. Our results showed similar effects of P. larvae on the survival rate and patterns of AMP gene expression in both honey bee species when bee larvae are infected with spores at the median lethal concentration (LC5 0 ) for A. mellifera. All AMPs of infected bee larvae showed significant upregulation compared with noninfected bee larvae in both honey bee species. However, larvae of A. cerana were more susceptible than A. mellifera when the same larval ages and spore concentration of P. larvae were used. It also appears that A. cerana showed higher levels of AMP expression than A. mellifera. This research provides the first evidence of survival rate, LC50 and immune response profiles of Asian honey bees, A. cerana, when infected by P. larvae in comparison with the European honey bee, A. mellifera.

The binary polyhydrides of heavy rare earth lutetium that shares a similar valence electron configuration to lanthanum have been experimentally discovered to be *** lutetium polyhydrides were successfully synthesized at high pressure and high temperature conditions using a diamond anvil cell in combinations with the in-situ high pressure laser heating *** resistance measurements as a function of temperature were performed at the same pressure of synthesis in order to study the transitions of superconductivity(SC).The superconducting transition with a maximum onset temperature(Tc)71 K was observed at pressure of 218 GPa in the *** Tcdecreased to 65 K when pressure was at 181 *** the evolution of SC at applied magnetic fields,the upper critical field at zero temperatureμ0Hc2(0)was obtained to be~36 *** in-situ high pressure X-ray diffraction experiments imply that the high TcSC should arise from the Lu4H23phase with Pm3n symmetry that forms a new type of hydrogen cage framework different from those reported for previous light rare earth polyhydride superconductors.

Metabolic engineering efforts toward rewiring metabolism of cells to produce new compounds often require the utilization of non-native enzymatic machinery that is capable of producing a broad range of chemical *** encompass one of the largest classes of chemically diverse natural *** thousands of known polyketides,modular polyketide synthases(PKSs)share a particularly attractive biosynthetic logic for generating chemical *** engineering of modular PKSs could open access to the deliberate production of both existing and novel *** this review,we discuss PKS engineering efforts applied at both the protein and cellular level for the generation of a diverse range of chemical structures,and we examine future applications of PKSs in the production of medicines,fuels and other industrially relevant chemicals.

dear Editor, Plant cells are encased by cell walls--rigid yet highly dynamic composite structures, which consist of cellulose, hemicelluloses, pectic polysaccharides, glycoproteins, and the polyphenol lignin （Somerville et al., 2004）. Biosynthesis of the wall polysaccharides is facilitated by donor- and acceptor- substrate-specific glycosyltransferases catalyzing the transfer of the sugar moiety from a nucleotide sugar to form a specific glycosidic linkage. Glycosyltransferases have been grouped into families based on primary structure, one of which is fam- ily GT77 （***/GT77_***）.

Non-alcoholic steatohepatitis(NASH),an advanced form of non-alcoholic fatty liver disease(NAFLd),has emerged as the leading cause of liver failure and related ***,no medication is specifically approved to treat NAFLd or *** we report that oral administration of honey vesiclelike nanoparticles(H-VLNs)to naturally aged mice protects the liver from NASH development.H-VLNs are dominantly taken up by Kupffer cells in the liver and suppress hepatic chronic inflammation and further development of fibrosis and nodule formation in aged *** their reported antiinflammasome function,H-VLNs are found to inhibit the transcriptional activities of C-JUN and nuclear factor-kappa B(NF-κB).MicroRNAs miR5119 and miR5108 and phenolic compound luteolin in H-VLNs are identified in suppressing both the C-JUN and NF-kB ***,oral intake of H-VLNs represents a promising new user-friendly modality to prevent the development of NASH.

despite tremendous efforts to fight cancer,it remains a major public health problem and a leading cause of death *** increased knowledge of cancer pathways and improved technological platforms,precision therapeutics that specifically target aberrant cancer pathways have improved patient ***,a primary cause of unsuccessful cancer therapy remains cancer drug *** this review,we summarize the broad classes of resistance to cancer therapy,particularly pharmacokinetics,the tumor microenvironment,and drug resistance ***,we describe how bacterial-mediated cancer therapy,a bygone mode of treatment,has been revitalized by synthetic biology and is uniquely suited to address the primary resistance mechanisms that confound traditional *** genetic engineering,we discuss how bacteria can be potent anticancer agents given their tumor targeting potential,anti-tumor activity,safety,and coordinated delivery of anti-cancer drugs.
