The unique ORF8 protein from SARS-CoV-2 binds to human dendritic cells and induces a hyper-inflammatory cytokine storm

The novel coronavirus pandemic,first reported in December 2019,was caused by the severe acute respiratory syndrome coronavirus 2(SARS-CoV-2).SARS-CoV-2 infection leads to a strong immune response and activation of antigen-presenting cells,which can elicit acute respiratory distress syndrome(ARDS)characterized by the rapid onset of widespread inflammation,the so-called cytokine *** response to viral infections,monocytes are recruited into the lung and subsequently differentiate into dendritic cells(DCs).DCs are critical players in the development of acute lung inflammation that causes ***,we focus on the interaction of a specific SARS-CoV-2 open reading frame protein,ORF8,with *** show that ORF8 binds to DCs,causes pre-maturation of differentiating DCs,and induces the secretion of multiple proinflammatory cytokines by these *** addition,we identified DC-SIGN as a possible interaction partner of ORF8 on *** of ORF8 leads to reduced production of IL-1β,IL-6,IL-12p70,TNF-α,MCP-1(also named CCL2),and IL-10 by ***,a neutralizing antibody blocking the ORF8-mediated cytokine and chemokine response could be an improved therapeutic strategy against SARS-CoV-2.

In this paper,we develop an entropy-conservative discontinuous Galerkin(DG)method for the shallow water(SW)equation with random *** of the most popular methods for uncertainty quantifcation is the generalized Polynomial Chaos(gPC)approach which we consider in the following *** apply the stochastic Galerkin(SG)method to the stochastic SW *** the SG approach in the stochastic hyperbolic SW system yields a purely deterministic system that is not necessarily hyperbolic *** lack of the hyperbolicity leads to ill-posedness and stability issues in numerical *** transforming the system using Roe variables,the hyperbolicity can be ensured and an entropy-entropy fux pair is known from a recent investigation by Gerster and Herty(***.27(3):639–671,2020).We use this pair and determine a corresponding entropy fux ***,we construct entropy conservative numerical twopoint fuxes for this augmented *** applying these new numerical fuxes in a nodal DG spectral element method(DGSEM)with fux diferencing ansatz,we obtain a provable entropy conservative(dissipative)*** numerical experiments,we validate our theoretical fndings.

Assessment of bone turnover and bone quality in type 2 diabetic bone disease： current concepts and future directions

Substantial evidence exists that in addition to the well-known complications of diabetes, increased fracture risk is an important morbidity. This risk is probably due to altered bone properties in diabetes. Circulating biochemical markers of bone turnover have been found to be decreased in type 2 diabetes （T2D） and may be predictive of fractures independently of bone mineral density （BMD）. Serum sclerostin levels have been found to be increased in T2D and appear to be predictive of fracture risk independent of BMD. Bone imaging technologies, including trabecular bone score （TBS） and quantitative CT testing have revealed differences in diabetic bone as compared to non-diabetic individuals. Specifically, high resolution peripheral quantitative CT （HRpQCT） imaging has demonstrated increased cortical porosity in diabetic postmenopausal women. Other factors such as bone marrow fat saturation and advanced glycation endproduct （AGE） accumulation might also relate to bone cell function and fracture risk in diabetes. These data have increased our understanding of how T2D adversely impacts both bone metabolism and fracture risk.

Enhancing the performance of porous silicon biosensors:the interplay of nanostructure design and microfluidic integration

This work presents the development and design of aptasensor employing porous silicon(PSi)Fabry‒Pérot thin films that are suitable for use as optical transducers for the detection of lactoferrin(LF),which is a protein biomarker secreted at elevated levels during gastrointestinal(GI)inflammatory disorders such as inflammatory bowel disease and chronic *** overcome the primary limitation associated with PSi biosensors—namely,their relatively poor sensitivity due to issues related to complex mass transfer phenomena and reaction kinetics—we employed two strategic approaches:First,we sought to optimize the porous nanostructure with respect to factors including layer thickness,pore diameter,and capture probe ***,we leveraged convection properties by integrating the resulting biosensor into a 3D-printed microfluidic system that also had one of two different micromixer architectures(i.e.,staggered herringbone micromixers or microimpellers)*** demonstrated that tailoring the PSi aptasensor significantly improved its performance,achieving a limit of detection(LOD)of 50 nM—which is>1 order of magnitude lower than that achieved using previously-developed biosensors of this ***,integration into microfluidic systems that incorporated passive and active micromixers further enhanced the aptasensor’s sensitivity,achieving an additional reduction in the LOD by yet another order of *** advancements demonstrate the potential of combining PSi-based optical transducers with microfluidic technology to create sensitive label-free biosensing platforms for the detection of GI inflammatory biomarkers.

Pitt, a Novel Tetratricopeptide Repeat Protein Involved in Light-Dependent Chlorophyll Biosynthesis and Thylakoid Membrane Biogenesis in Synechocystis sp. PCC 6803

Biogenesis of photosynthetic pigment/protein complexes is a highly regulated process that requires various assisting factors. Here, we report on the molecular analysis of the Pitt gene （sir1644） from the cyanobacterium Synechocystis sp. PCC 6803 （Synechocystis 6803） that encodes a membrane-bound tetratricopeptide repeat （TPR） protein of formerly unknown function. Targeted inactivation of Pitt affected photosynthetic performance and light-dependent chlorophyll synthesis. Yeast two-hybrid analyses and native PAGE strongly suggest a complex formation between Pitt and the light-dependent protochlorophyllide oxidoreductase （POR）. Consistently, POR levels are approximately threefold reduced in the pitt insertion mutant. The membrane sublocalization of Pitt was found to be dependent on the presence of the periplasmic photosystem Ⅱ （PSⅡ） biogenesis factor PratA, supporting the idea that Pitt is involved in the early steps of photosynthetic pigment/protein complex formation.

Seasonal changes in the parasite prevalence of a small Malagasy lemur species (Lepilemur edwardsi)

Parasitic infections can impact theﬁtness of individuals and can have inﬂuence on animals’population *** individuals’parasite prevalence often changes depending on external or seasonal changes,for example,rainfall and ambient temperatures,but also on internal changes,for example,changes in body *** this study we aimed to identify the environmental factors that may inﬂuence the intestinal parasite and ectoparasite prevalence of the folivorous Malagasy primate species,Lepilemur edwardsi,living in a seasonal dry deciduous *** living in this habitat have to adapt to seasonal changes of ambient temperature,with almost no precipitation during the dry season and hence strongﬂuctuations of resource availability throughout the *** sampled the feces and ectoparasites of *** throughout the *** parasite prevalence increased from the wet to the dry season and was highest in the late dry season,which might be due to the accompanying decrease in diet ***,ectoparasite prevalence decreased in the dry season,presumably due to the prevailing unfavorable environmental conditions for the development of ectoparasites(i.e.,mites and ticks).Paired with the higher resting metabolism and stress level of *** during the late dry season,it seems that this species may struggle when dry seasons intensify in its habitat.

The impact of frailty on in-hospital complications in elderly patients with acute coronary syndrome

Previous encapsulation response enhances within individual protection against fungal parasite in the mealworm beetle Tenebrio mofitor

Immune defenses of insects show either broad reactions or specificity and durability of induced protection against attacking parasites and pathogens. In this study, we tested whether encapsulation response against nylon monofilament increases between two attempts of activation of immune system in mealworm beetles Tenebrio molitor, and whether previous exposure to nylon monofilament may also increase protection against an entomopathogenic fungus. We found that survival of beetles subjected to immune activation by nylon implant and subsequent fungal exposure a week later was significantly higher than survival of beetles which had been subjected to fimgal infection only. This result suggests that previous immune activation by the nylon implant may be considered as broad spectrum ＂immune priming＂ which helps to fight not only the same intruder but also other parasites.

In the past few years, stem cells have become the focus of research by regenerative medicine professionals and tissue engineers. Embryonic stem cells, although capable of differentiating into cell lineages of all three germ layers, are limited in their utilization due to ethical issues. In contrast, the autologous harvest and subsequent transplantation of adult stem cells from bone marrow, adipose tissue or blood have been experimentally utilized in the treatment of a wide variety of diseases ranging from myocardial infarction to Alzheimer's disease. The physiologic consequences of stem cell transplantation and its impact on functional recovery have been studied in countless animal models and select clinical trials. Unfortunately, the bench to bedside translation of this research has been slow. Nonetheless, stem cell therapy has received the attention of spinal surgeons due to its potential benefits in the treatment of neural damage, muscle trauma, disk degeneration and its potential contribution to bone fusion.
