Brain antigens in functionally distinct antigen-presenting cell populations in cervical lymph nodes in MS and EAE

Drainage of central nervous system(CNS) antigens to the brain-draining cervical lymph nodes(CLN) is likely crucial in the initiation and control of autoimmune responses during multiple sclerosis(MS).We demonstrate neuronal antigens within CLN of MS *** monkeys and mice with experimental autoimmune encephalomyelitis(EAE) and in mouse models with non-inflammatory CNS damage,the type and extent of CNS damage was associated with the frequencies of CNS antigens within the cervical lymph *** addition,CNS antigens drained to the spinal-cord-draining lumbar lymph *** human MS CLN,neuronal antigens were present in pro-inflammatory antigen-presenting cells(APC),whereas the majority of myelincontaining cells were *** may reflect a different origin of the cells or different drainage ***,neuronal antigen-containing cells in human CLN did not express the lymph node homing receptor CCR7,whereas myelin antigen-containing cells in situ and in vitro ***,CLN from EAE-affected CCR7-deficient mice contained equal amounts of myelin and neuronal antigens as wild-type *** conclude that the type and frequencies of CNS antigens within the CLN are determined by the type and extent of CNS ***,the presence of myelin and neuronal antigens in functionally distinct APC populations within MS CLN suggests that differential immune responses can be evoked.

ELI-Beamlines(ELI-BL),one of the three pillars of the Extreme Light Infrastructure endeavour,will be in a unique position to perform research in high-energy-density-physics(hEDP),plasma physics and ultra-high intensity(UhI)ð>10^(22) W=cm^(2)) lasereplasma *** the need for hED laboratory physics was identified and the P3(plasma physics platform)installation under construction in ELI-BL will be an *** ELI-BL 10 PW laser makes possible fundamental research topics from high-field physics to new extreme states of matter such as radiation-dominated ones,high-pressure quantum ones,warm dense matter(WDM)and ultra-relativistic *** is of fundamental importance for research in the field of laboratory astrophysics and inertial confinement fusion(ICF).Reaching such extreme states of matter now and in the future will depend on the use of plasma optics for amplifying and focusing laser *** article will present the relevant technological infrastructure being built in ELI-BL for hEDP and UhI,and gives a brief overview of some research under way in the field of UhI,laboratory astrophysics,ICF,WDM,and plasma optics.

目的:探讨嗅鞘细胞(olfactory ensheathing cells,OEC)移植在大鼠损伤脊髓后是否有独特的迁移和轴突生长导向特性。方法:将表达绿色荧光蛋白基因的OEC注入到C4脊髓损伤大鼠距离损伤部位头端1mm及尾端1mm背柱白质处,注射后1、3、12、24h... 详细信息

目的:探讨嗅鞘细胞(olfactory ensheathing cells,OEC)移植在大鼠损伤脊髓后是否有独特的迁移和轴突生长导向特性。方法:将表达绿色荧光蛋白基因的OEC注入到C4脊髓损伤大鼠距离损伤部位头端1mm及尾端1mm背柱白质处,注射后1、3、12、24h及3、7、28d灌注固定取材,冰冻切片和免疫组化分析。用骨髓基质细胞和成纤维细胞移植到同样的损伤部位做为对照进行同样的分析。另将OEC注射到距离损伤部位头尾侧1mm处脊髓灰质内、小剂量注射在白质内、在损伤前3d或损伤后9d注射、注射到未损伤脊髓白质中,观察细胞迁移情况。结果:OEC在细胞注射后1h内即形成由注射压力造成的从注射部位向损伤处的被动性延伸带,并且不断地从注射部位向损伤处延伸扩散,在形态学方面好象起到"桥接"损伤处的作用。对照组骨髓基质细胞和成纤维细胞注射后也迅速形成细胞"桥接"带,并扩散至损伤空腔。将OEC注射到脊髓灰质内或小剂量注射或注射到未损伤的脊髓白质内、在脊髓损伤3d前或9d后注射细胞均没有见到细胞带延伸进入损伤部位。OEC在注射部位、细胞带以及损伤部位有增殖现象。28d后,共焦免疫酶标法证实细胞带取代了脊髓原有星形胶质细胞,但是下行或上行长束轴突没有优先延伸至该细胞带,也没能起到维持皮层脊髓轴突的桥接作用。结论:OEC在植入大鼠损伤脊髓后没有独特的迁移特性,与骨髓基质细胞或成纤维细胞相比没有明显促进轴突生长的作用,也没有支持皮层脊髓轴突在脊髓损伤处形成桥接的作用。

The cryptic lifestyle of most fungi necessitates molecular identification of the guild in environmental *** the past decades,rapid development and affordability of molecular tools have tremendously improved insights of the fungal diversity in all ecosystems and ***,in spite of the progress of molecular methods,knowledge about functional properties of the fungal taxa is vague and interpretation of environmental studies in an ecologically meaningful manner remains *** order to facilitate functional assignments and ecological interpretation of environmental studies we introduce a user friendly traits and character database FungalTraits operating at genus and species hypothesis *** the information from previous efforts such as FUNGuild and FunFun together with involvement of expert knowledge,we reannotated 10,210 and 151 fungal and Stramenopila genera,*** resulted in a stand-alone spreadsheet dataset covering 17 lifestyle related traits of fungal and Stramenopila genera,designed for rapid functional assignments of environmental *** order to assign the trait states to fungal species hypotheses,the scientific community of experts manually categorised and assigned available trait information to 697,413 fungal ITS *** the basis of those sequences we were able to summarise trait and host information into 92,623 fungal species hypotheses at 1%dissimilarity threshold.

目的:观察程序性细胞死亡配体1(programmed death-ligand 1,PD-L1)抑制剂阿替利珠单抗在中国高发实体瘤,包括食管癌(esophageal cancer,EC)、胃癌(gastric cancer,GC)、肝细胞癌(hepatocellular carcinoma,hCC)、鼻咽癌(nasopharyngeal cancer,NPC)和非小细胞肺癌(non-small cell lung cancer,NSCLC)患者中的药代动力学(pharmacokinetics,PK)、疗效和安全性数据。方法:本研究为开放标签的Ⅰ期临床试验,于2016年8月4日至2019年4月15日在中国6个研究中心进行。入组患者年龄≥18岁,患有经组织学证实的无法治愈或转移性的实体瘤,且既往抗肿瘤治疗失败。PK阶段研究了阿替利珠单抗单药治疗的PK和安全性;扩展阶段研究了阿替利珠单抗单药治疗(入组EC、GC、hCC、NPC患者)和联合化疗(入组NSCLC患者)的安全性和有效性。结果:共入组120例患者(PK阶段20例;扩展期每队列20例)。阿替利珠单抗单药组患者(n=100)中有42例(42.0%)为PD-L1阳性,9例(9.0%)为微卫星高度不稳定性。阿替利珠单抗的清除率为0.219 L/d,重复给药6~9周(2~3个周期)后达到稳态。EC、GC、hCC、NPC和NSCLC的客观缓解率(objective response rate,ORR)分别为10.0%、15.0%、10.0%、5.0%和40.0%。在PD-L1阳性的肿瘤患者中,阿替利珠单抗的ORR为11.9%,阿替利珠单抗联合吉西他滨和顺铂的ORR为46.2%。2例GC患者在假性进展后获得了持久的肿瘤缩小。阿替利珠单抗单药组最常见的治疗相关不良事件是疲劳、贫血、发热和白细胞计数减少,联合组最常见的治疗相关AE是贫血、白细胞计数减少和食欲下降。本试验没有发现新的安全信号。结论:中国患者应用阿替利珠单抗的PK、疗效和安全性与之前研究中入组的全球患者的数据相似。

背景术后恶心呕吐（postoperativenauseaandvomiting，PONV）是一种常见的术后并发症。研究证实神经激肽．1（neurokinin一1，NK，）受体拮抗剂可以安全有效地用于预防和治疗人类PONV。Rolapitant（罗纳吡坦）为一种吸收迅速、半衰期相当长（达180小时）的强效选择性NK、受体拮抗剂，潜在的药物相互作用低。我们对Rolapitant用于预防PONV高危人群的量效关系以及术后5天的预防迟发性IK）NV的效果进行了研究。方法这项关于Rolapitant的随机、多中心、双盲、有效剂量范围的研究设有安慰剂组和阳性对照组。纳入619例行开腹手术的成年妇女，基于PONV或晕动病史进行分层，分为6个研究组：Rolapitant口服剂量5mg组、20mg组、70nag组或200mg组；静注昂丹司琼4mg组或安慰剂组。主要研究终点：不考虑补救用药，拔管后24小时的无呕吐发生率。结果Rolapitant20mg、70mg或200mg组患者的术后24小时无呕吐发生率高于安慰剂组。Rolapitant剂量与主要转归呈线性相关。与安慰剂组相比，Rolapitant70mg和200mg组的呕吐次数明显更低（P≤0．001，对数秩和检验）。尽管术后24小时无呕吐次数在Rolapitant组和昂丹司琼对照组没有明显差异，但Rolapitant200mg组和70mg组的术后72小时和120小时无呕吐发生率更高（不考虑补救用药）。结论Rolapitant降低术后呕吐的疗效优于安慰剂，呈剂量依赖性地降低呕吐发生率。而且Rolapitant与安慰剂在不良反应方面没有差异。
