何首乌肝损伤风险的发现、评价与防控策略研究

近年来,中草药相关的不良反应/事件频繁被报道,尤其针对新发现“有毒”中药的安全性问题,往往难以做出科学的回答并制定有效解决方案。中药由于自身的复杂性及缺乏系统性研究,使得我们对中草药相关肝损伤的成因机制了解有限。因此,破解中草药安全性难题亟需在中药毒性认知及安全风险防控方面实现突破。笔者团队以何首乌肝损伤作为典型范例,基于首创中药药源性肝损伤因果关系评价“整合证据链法”,证实了何首乌致肝损伤的客观真实性。进一步基于团队创新性建立的病证毒理学方法,科学揭示了何首乌肝损伤的物质基础及成因机制,也提出并证实了中药免疫特异质肝损伤“三因致毒”机制假说。在此基础上,阐明了何首乌特异质肝损伤易感人群的病证特征,并筛选发现了多种生物标志物,包括基因标志物HLA-b.35:01、免疫学及代谢组标志物。最后,探索创建了基于易感人群识别、易感物质控制以及个性化用药的“人-药-用”三维风险防控的安全用药策略和方法,为全面揭示“有毒”中药毒副反应科学内涵并建立科学有效的风险防控对策提供了基础。

针对荧光分子检测普遍灵敏度低和检测范围窄的问题,制备了具有等离子激元共振特性的重掺杂半导体纳米结构Cu_(2-x)S和典型的稀土掺杂上转换发光纳米颗粒NaYF_(4)∶Yb.Er,通过三相界面自组装方法获得了Cu_(2-x)S/NaYF_(4)∶Yb.Er薄膜基底。结合有限元模拟,计算了不同摆放情况下Cu_(2-x)S周围的局域电场分布,研究了在实际薄膜中Cu_(2-x)S纳米盘之间产生的等离激元耦合对上转换发光性能以及对拉曼信号增强的影响。结果表明,Cu_(2-x)S等离激元层与NaYF_(4)∶Yb.Er发光层的耦合,不仅得到了上转换3个数量级的提高,还实现了分子检测10^(-7)mol·L^(-1)的检测极限,并且获得了10^(-3)~10^(-7)mol·L^(-1)的宽线性响应,从而达到高灵敏度的定性和定量双功能的精确检测。

b.CKGROUND： Polyhydroxyalkanoates are a good sub.titute for synthetic plastic b.cause they are highly b.ocompatib.e, ecofriendly, and b.odegradab.e. b.cteria in freshwater b.dies such as rivers, tub. wells, and canals are exposed to alternating high and low concentrations of sub.trates that induce PHA production. METHODS： Fresh water samples were collected for isolation of b.cterial strains. Screening of PHA in b.cterial cells was performed with Sudan and Nile Red staining. Extracted PHA was characterized b. FTIR. RESULTS： In this study, nine b.cterial isolates were selected for PHA production on the b.sis ofphenotypic screening. Their ab.lity to accumulate PHAs was determined using different monosaccharides and disaccharides. Two b.cterial isolates b.cillus cereus T1 （KY746353） and b.cillus cereus R3 （KY746354） produced PHAs. Optimal growth of the b.cterial strain （T1） was ob.erved in the presence of glucose, followed b. maximum production of PHAs （63% PHAs） during the logarithmic phase of growth. b. cereus R3 （KY746354） accumulated 60% PHAs b. dry cell weight. CONCLUSION： PHA accumulation was relatively less with fructose, b.t b.th strains showed increased production （up to 50%） with sucrose. The polymer produced was characterized b. Fourier-transform infrared spectroscopy （FTIR）, which showed that the compound contains short-chain PHAs.

基于蛇优化算法的电力系统无功优化

无功优化在电力系统中具有重要的意义,可以提高系统的功率因数,减少有功损耗并改善电压稳定性。蛇优化算法具有全局搜索能力和快速收敛的特点,故将该算法作为一种新的优化方法运用到无功优化中,建立以机端电压、变压器变比和无功补偿量为系统控制变量,以网络损耗最小为目标的无功优化模型。通过采用57节点系统,将蛇优化算法与粒子群算法、改进粒子群算法分别进行比较验证。实验结果表明,蛇优化算法能够有效地优化电力系统的无功功率,提高系统的性能和稳定性。

代偿性多汗(compensatory hyperhidrosis,CH)是胸腔镜下胸交感神经切断术(endoscopic thoracic sympathotomy,ETS)治疗手汗症术后最常见的副作用。极重度CH严重干扰患者的生活、工作、学习和社交等日常活动,使患者生活质量大幅降低。防治CH是当前手汗症微创外科亟待解决的热点与难点问题。本文报道1例术后极重度CH 8年的29岁男性患者,2023年12月11日及2024年1月3日于福建医科大学附属第一医院施行分期单侧胸腔镜下R4~R10扩大胸交感神经切断术(expanded sympathotomy,ES),术后随访3个月,患者临床症状及生活质量显著改善,无手汗复发以及其他并发症,疗效满意。
