Overexpression of OsPAP10a, A Root-Associated Acid Phosphatase, Increased Extracellular Organic Phosphorus Utilization in Rice

Phosphorus （P） deficiency is a major limitation for plant growth and development. Among the wide set of responses to cope with low soil P, plants increase their level of intracellular and secreted acid phosphatases （APases）, which helps to catalyze inorganic phosphate （Pi） hydrolysis from organophosphates, in this study we characterized the rice （Oryza sativa） purple acid phosphatase 10a （OsPAPIOa）. OsPAPIOa belongs to group la of purple acid phosphatases （PAPs）, and clusters with the principal secreted PAPs in a variety of plant species including Arabidopsis. The transcript abundance of OsPAPIOa is specifically induced by Pi deficiency and is controlled by OsPHR2, the central transcription factor controlling Pi homeostasis. In gel activity assays of root and shoot protein extracts, it was revealed that OsPAPIOa is a major acid phosphatase isoform induced by Pi starvation. Constitutive overexpression of OsPAPIOa results in a significant increase of phosphatase activity in both shoot and root protein extracts. In vivo root 5-bromo.4-chloro-3-indolyl-phosphate （BCIP） assays and activity measurements on external media showed that OsPAPIOa is a root-associated APase. Furthermore, overexpression of OsPAPIOa significantly improved ATP hydrolysis and utilization compared with wild type plants. These results indicate that OsPAPIOa can potentially be used for crop breeding to improve the efficiency of P use.

Defining the Mitochondrial Stress Response in Arabidopsis thaliana

To obtain a global overview of how mitochondria respond to stress, we aimed to define the plant mitochondrial stress response （MSR）. By combining a set of 1196 Arabidopsis thaliana genes that putatively encode mitochondrial proteins with 16 microarray experiments on stress-related conditions, 45 nuclear encoded genes were defined as widely stress-responsive. Using green fluorescent protein （GFP） fusion assays, the mitochondrial targeting of a large number of these proteins was tested, confirming in total 26 proteins as mitochondrially targeted. Several of these proteins were observed to be dual targeted to mitochondria and plastids, including the small heat shock proteins sHSP23.5 and sHSP23.6. In addition to the well defined stress components of mitochondria, such as alternative oxidases, nicotinamide adenine dinudeotide （NAD（P）H） dehydrogenases, and heat shock proteins, a variety of other proteins, many with unknown function, were identified. The mitochondrial carrier protein family was over-represented in the stress-responsive genes, suggesting that stress induces altered needs for metabolite transport across the mitochondrial inner membrane. Although the genes encoding many of these proteins contain common cis-acting regulatory elements, it was apparent that a number of distinct regulatory processes or signals likely triggered the MSR. Therefore, these genes provide new model systems to study mitochondrial retrograde regulation, in addition to the widely used alternative oxidase model. Additionally, as changes in proteins responsive to stress did not correlate well with changes at a transcript level, it suggests that post-transcriptional mechanisms also play an important role in defining the MSR.

Cotton is a major crop that provides the most important renewable textile fibers in the *** of the taxonomy and evolution of cotton species have received wide attentions,not only due to cotton’s economic value but also due to the fact that Gossypium is an ideal model system to study the origin,evolution,and cultivation of polyploid *** studies suggested the involvement of mitochondrial genome editing sites and copy number as well as mitochondrial functions in cotton fiber ***,with only a few mitogenomes assembled in the cotton genus Gossypium,our knowledge about their roles in cotton evolution and speciation is still *** close this gap,here we assembled 20 mitogenomes from 15 cotton species spanning all the cotton clades(A–G,K,and AD genomes)and 5 cotton relatives using short and long sequencing *** analyses uncovered a high level of mitochondrial gene sequence conservation,abundant sequence repeats and many insertions of foreign sequences,as well as extensive structural variations in cotton *** sequence repeats and foreign sequences caused significant mitogenome size inflation in Gossypium and its close relative Kokia in general,while there is no significant difference between the lint and fuzz cotton mitogenomes in terms of gene content,RNA editing,and gene expression ***,we further revealed the specific presence and expression of two novel mitochondrial open reading frames(ORFs)in lint-fiber cotton ***,these structural features and novel ORFs help us gain valuable insights into the history of cotton evolution and polyploidization and the origin of species producing long lint fibers from a mitogenomic perspective.

Using Circular Economics Strategies to Manage Tourism on Remote Islands:A SMART Approach

Circular economics theory offers destination managers of remote islands a solution-oriented approach to environmental sustainability. This paper examines the practical benefits of this theory that will support the development of a circular economy regarding sustainable island tourism (SIT). The aim of this paper is to inform discussions regarding the feasibility of applying a practice theory that emphasises changes in the behaviours across three segments, visitors, residents, and enterprises using SMART technologies. To achieve this objective, a SMART indicator system (SIS) was used to identify current practices that would support a circular economy (CE). The key innovation of this study is the creation of a proposed framework that may inform best practices for SIT in the future. The findings highlight the potential opportunity of evidenced-based planning to deliver a circular economy strategy (CES) for remote islands in the future. These insights can potentially impact the way that local authorities and destination managers approach the planning of tourism for six islands along the west coast of Ireland.

Coordinated regulation of the mitochondrial retrograde response by circadian clock regulators and ANAC017

Mitochondrial retrograde signaling(MRS)supports photosynthetic function under a variety of *** of mitochondrial dysfunction with myxothiazol(a specific inhibitor of the mitochondrial bc1 complex)or antimycin A(an inhibitor of the mitochondrial bc1 complex and cyclic electron transport in the chloroplast under light conditions)in the light and dark revealed diurnal control of *** was evidenced by(1)significantly enhanced binding of ANAC017 to promoters in the light compared with the dark in Arabidopsis plants treated with myxothiazol(but not antimycin A),(2)overlap in the experimentally determined binding sites for ANAC017 and circadian clock regulators in the promoters of ANAC013 and AOX1a,(3)a diurnal expression pattern for ANAC017 and transcription factors it regulates,(4)altered expression of ANAC017-regulated genes in circadian clock mutants with and without myxothiazol treatment,and(5)a decrease in the magnitude of LHY and CCA1 expression in an ANAC017-overexpressing line and protein–protein interaction between ANAC017 and *** study also shows a large difference in transcriptome responses to antimycin A and myxothiazol in the dark:these responses are ANAC017 independent,observed in shoots and roots,similar to biotic challenge and salicylic acid responses,and involve ERF and ZAT transcription *** suggests that antimycin A treatment stimulates a second MRS pathway that is mediated or converges with salicylic acid signaling and provides a merging point with chloroplast retrograde signaling.

At12Cys-1 （At5g64400） and At12Cys-2 （At5g09570） are two closely related isogenes that encode small, twin cysteine proteins, typically located in mitochondria. At12Cys-2 transcript is induced in a variety of mutants with disrupted mitochondrial proteins, but an increase in At12Cys protein is only detected in mutants with reduced mitochondrial complex I abundance. Induction of At12Cys protein in mutants that lack mitochondrial complex I is accompanied by At12Cys protein located in mitochondria, chloroplasts, and the cytosoh Biochemical analyses revealed that even single gene deletions, i.e., At12cys-1 orAtl2cys-2, have an effect on mitochondrial and chloroplast functions. However, only double mutants, i.e., At12cys-1：At12cys.2, affect the abundance of protein and mRNA transcripts encoding translation elongation factors as well as rRNA abundance. Blue native PAGE showed that At12Cys co-migrated with mitochondrial supercomplex I ＋ lU. Likewise, deletion of both At12cys-1 and At12cys-2 genes, but not single gene deletions, results in enhanced tolerance to drought and light stress and increased anti-oxidant capacity. The induction and multiple localization of At12Cys upon a reduction in complex I abundance provides a mechanism to specifically signal mitochondrial dysfunction to the cytosol and then beyond to other organelles in the cell.
