Tight j.nction disruption: Helicobacter pylori and dysregulation of the gastric mucosal barrier

Long-term chronic infection with Helicobacter pylori(H. pylori) is a risk factor for gastric cancer development. In the multi-step process that leads to gastric cancer,tight j.nction dysfunction is thought to occur and serve as a risk factor by permitting the permeation of luminal contents across an otherwise tight mucosa. Mechanisms that regulate tight j.nction function and structure in the normal stomach,or dysfunction in the infected stomach,however,are largely unknown. Although conventional tight j.nction components are expressed in gastric epithelial cells,claudins regulate paracellular permeability and are likely the target of inflammation or H. pylori itself. There are 27 different claudin molecules,each with unique properties that render the mucosa an intact barrier that is permselective in a way that is consistent with cell physiology. Understanding the architecture of tight j.nctions in the normal stomach and then changes that occur during infection is important but challenging,because most of the reports that catalog claudin expression in gastric cancer pathogenesis are contradictory. Furthermore,the role of H. pylori virulence factors,such as cytotoxin-associated gene A and vacoulating cytotoxin,in regulating tight j.nction dysfunction during infection is inconsistent in different gastric cell lines and in vivo,likely because non-gastric epithelial cell cultures were initially used to unravel the details of their effects on the stomach. Hampering further study,as well,is the relative lack of cultured cell models that have tight j.nction claudins that are consistent with native tissues. This summary will review the current state of knowledge about gastric tight j.nctions,normally and in H. pylori infection,and make predictions about the consequences of claudin reorganization during H. pylori infection.

A linear regression model (LRM) for groundwater chemistry in and around the Vaniyambadi industrial area, Tamil Nadu, India

A linear regression model in conj.nction with cluster analysis was applied to the groundwater quality parameters for the Vaniyambadi industrial area, Tamil Nadu, India. These physico-chemical parameters were collected from 25 wells by intensive groundwater sampling conducted during j.nuary 2010. All the maj.r ions, pH and electrical conductivity were analyzed. The abundances of cations were in the order of Na j.r clusters representing the influence of tanneries and geological formations in the study area, which confirmed the results of maj.r ion chemistry.

Adenosine-treated bioprinted muscle constructs prolong cell survival and improve tissue formation

It is crucial to maintaining the viability of biofabricated human-sized tissues to ensure their successful survival and function after *** is a purine nucleoside that has the function to suppress cellular metabolism and has been previously proposed as a method to prolong cell viability under *** this study,we optimized the dose concentration of adenosine for incorporation into bioprinted constructs to preserve long-term cell viability in *** results showed that muscle cells(C2C12)containing 6,7,or 8 mM adenosine maintained high cell viability for 20 days under hypoxic conditions(0.1%O2),whereas cells without adenosine treatment showed 100%cell death after 11 *** 20 days under hypoxic conditions,muscle cells treated with adenosine proliferated and differentiated when transferred to normoxic *** these adenosine concentrations,6 mM was picked as the optimized adenosine concentration for further investigations due to its most effective results on improving cell *** bioprinted muscle constructs containing adenosine(6 mM)maintained high cell viability for 11 days under hypoxic conditions,while the control constructs without adenosine had no live *** in vivo validation,the bioprinted constructs with adenosine implanted under the dorsal subcutaneous space in mice,showed the enhanced formation of muscle tissue with minimal central necrosis and apoptosis,when compared to the constructs without *** positive in vitro and in vivo results demonstrate that the use of adenosine is an effective approach to preventing the challenge of hypoxia-induced necrosis in bioprinted tissues for clinical translation.

Impact of source tissue and ex vivo expansion on the characterization of goat mesenchymal stem cells

Background： There is considerable interest in using goats as models for genetically engineering dairy animals and also for using stem cells as therapeutics for bone and cartilage repair. Mesenchymal stem cells （MSCs） have been isolated and characterized from various species, but are poorly characterized in goats. Results： Goat MSCs isolated from bone marrow （BM-MSCs） and adipose tissue （ASCs） have the ability to undergo osteogenic, adipogenic and chondrogenic differentiation. Cytochemical staining and gene expression analysis show that ASCs have a greater capacity for adipogenic differentiation compared to BM-MSCs and fibroblasts. Different methods of inducing adipogenesis also affect the extent and profile of adipogenic differentiation in MSCs. Goat fibroblasts were not capable of osteogenesis, hence distinguishing them from the MSCs. Goat MSCs and fibroblasts express CD90, CD105, CD73 but not CD45, and exhibit cytoplasmic localization of OCT4 protein. Goat MSCs can be stably transfected by Nucleofection, but, as evidenced by colony-forming efficiency （CFE）, yield significantly different levels of progenitor cells that are robust enough to proliferate into colonies of integrants following G418 selection. BM-MSCs expanded over increasing passages in vitro maintained karyotypic stability up to 20 passages in culture, exhibited an increase in adipogenic differentiation and CFE, but showed altered morphology and amenability to genetic modification by selection. Conclusions： Our findings provide characterization information on goat MSCs, and show that there can be significant differences between MSCs isolated from different tissues and from within the same tissue. Fibroblasts do not exhibit trilineage differentiation potential at the same capacity as MSCs, making it a more reliable method for distinguishing MSCs from fibroblasts, compared to cell surface marker expression.

Current Status of Deltabaculoviruses, Cypoviruses and Chloriridoviruses Pathogenic for Mosquitoes

There are a variety of viral pathogens that cause disease in mosquitoes with most belonging to three maj.r groups. The most common viruses of mosquitoes are the baculoviruses (DBVs) (Baculoviridae: Deltabaculovirus), cytoplasmic polyhedrosis viruses (CPVs) (Reoviridae: Cypovirus) and the iridoviruses (MIVs) (Iridoviridae: Chloriridovirus). Baculoviruses and iridoviruses are DNA viruses while cypoviruses are the main RNA viruses in mosquitoes. This review presents an overview of the current status and recent advancements in understanding the biology and molecular features of mosquito pathogenic viruses.

This paper is the first attempt to evaluate huff-n-puff air inj.ction in a shale oil reservoir using a simulation *** mechanisms and physical processes of huff-n-puff air inj.ction in a shale oil reservoir are investigated through investigating production performance,thermal behavior,reservoir pressure and fluid saturation *** flooding is used as the basic case for a comparative *** simulation study suggests that thermal drive is the main recovery mechanism for huff-n-puff air inj.ction in the shale oil reservoir,but not for simple air *** synergic recovery mechanism of air flooding in conventional light oil reservoirs can be replicated in shale oil reservoirs by using air huff-npuff inj.ction *** huff-n-puff time is better for performing the synergic recovery mechanism of air inj.ction.O2 diffusion plays an important role in huff-n-puff air inj.ction in shale oil *** transmissibility as well as reservoir pressure maintenance ability in huff-n-puff air inj.ction is more pronounced than the simple air flooding after primary depletion *** obvious gas override is exhibited in both air flooding and air huff-n-puff inj.ction scenarios in shale ***-puff air inj.ction has great potential to develop shale oil *** results from this work may stimulate further investigations.
