Osteomyelitis is a d.vastating d.sease caused.by microbial infection of bone. While the frequency of infection following elective orthoped.c surgery is low, rates of reinfection are d.sturbingly high. Staphylococcus aureus is responsible for the majority of chronic osteomyelitis cases and.is often consid.red.to be incurable d.e to bacterial persistence d.ep within bone. Unfortunately, there is no consensus on clinical classifications of osteomyelitis and.the ensuing treatment algorithm. Given the high patient morbid.ty,mortality, and.economic burd.n caused.by osteomyelitis, it is important to elucid.te mechanisms of bone infection to inform novel strategies for prevention and.curative treatment. Recent d.scoveries in this field.have id.ntified.three d.stinct reservoirs of bacterial biofilm includ.ng: Staphylococcal abscess communities in the local soft tissue and.bone marrow, glycocalyx formation on implant hard.are and.necrotic tissue, and.colonization of the osteocyte-lacuno canalicular network(OLCN) of cortical bone. In contrast, *** intracellular persistence in bone cells has not been substantiated.in vivo, which challenges this mod. of chronic osteomyelitis. There have also been major ad.ances in our und.rstand.ng of the immune proteome against S. aureus, from clinical stud.es of serum antibod.es and.med.a enriched.for newly synthesized.antibod.es(MENSA), which may provid. new opportunities for osteomyelitis d.agnosis, prognosis, and.vaccine d.velopment. Finally, novel therapies such as antimicrobial implant coatings and.antibiotic impregnated.3d.printed.scaffold. represent promising strategies for preventing and.managing this d.vastating d.sease. Here, we review these recent ad.ances and.highlight translational opportunities toward. a cure.

CBASP Ad.pted.to Child.Play Therapy Structure to Prevent Early-Onset Persistent d.pressive d.sord.r

The current paper is a theoretical proposal that interfaces the Cognitive Behavioral Analysis System of Psychotherapy (CBASP) and.its emphasis on interpersonal consequences with the structured.ord.r of a Play Therapy Mod.l for troubled.3 - 8-year-old.child.en. This proposal is not a research paper or a review of literature;instead. it is a treatment proposal that is novel and.untested. CBASP psychotherapy, an empirically valid.ted.treatment, was d.veloped.originally to treat the persistently d.pressed.ad.lt. CBASP’s major focus of interpersonal consequation will be interfaced.with a Play Therapy structured.mod.l to rectify the malad.ptive preoperational functioning of five interpersonal types of problem-child.en. The types are classified.interpersonally using d.J. Kiesler’s Interpersonal Message Inventory (IMI). Kiesler’s IMI is employed.in this proposal as an ongoing assessment mod.lity, a source of information to make treatment strategy consequation d.cisions, and.third.y as an evaluative outcome variable. The troubled.child.types d.scribed.herein frequently become cand.d.tes for early-onset Persistent d.pressive d.sord.r (Pd.) unless rescued.by successful treatment. The origins of early-onset Pd. arise in d.sfunctional household. where toxic interpersonal relationships pred.minate: where “survival from abuse,” not growth, d.scribes the child.s mod.l d.velopmental experiences. These child.en are often exposed.to either serious traumas (e.g., sexual abuse, loss of a parent, physical abuse, physical or emotional neglect) or psychological insults (e.g., continuous, and.chronic verbal and.nonverbal abuse). The result, in the most serious cases, is a maturational stunting at the preoperational stage of d.velopment which, as noted.above, if not successfully resolved. thrusts the child.into early-onset Pd..

A Tiling Lemma and.Its Application to the Ratio Test for Convergence of Series

We prove that any collection which tiles the positive integers must contain one of two types of sub-collections. We then use this result to prove a variation of the Ratio Test for convergence of series. This version of the Ratio Test shows the convergence of certain series for which the Root Test (which is known to be more powerful than the conventional Ratio Test) fails. This version of the Ratio Test is also used.to prove a version of the Banach Contraction Principle for self-maps of a complete metric space.

The Role of RNA Structure in Posttranscriptional Regulation of Gene Expression

As more information is gathered.on the mechanisms of transcription and.translation, it is becoming apparent that these processes are highly regulated. The formation of mRNA second.ry and.tertiary structures is one such regulatory process that until recently it has not been analysed.in d.pth. Formation of these mRNA structures has the potential to enhance and.inhibit alternative splicing of transcripts, and.regulate rates and.amount of translation. As this regulatory mechanism potentially impacts at both the transcriptional and.translational level, while also potentially utilising the vast array of non-cod.ng RNAs, it warrants further investigation. Currently, a variety of high- throughput sequencing techniques includ.ng parallel analysis of RNA structure （PARS）, fragmentation sequencing （FragSeq） and.selective 2-hyd.oxyl acylation analysed.by primer extension （SHAPE） lead.the way in the genome-wid. id.ntification and.analysis of mRNA structure formation. These new sequencing techniques highlight the d.versity and.complexity of the transcriptome, and.d.monstrate another regulatory mechanism that could.become a target for new therapeutic approaches.
