Comprehensive Assessment of Anxiolytic Properties in 4-HPAA Derivatives: Bridging in Vivo Validation and Molecular Docking Analyses

Anxiety is a significant mental health issue that substantially affects an individual’s quality of life. Feelings of uneasiness, irritability, and sleep disturbances characterize it. 4-Hydroxyphenyl acetic acid (4-HPAA) is identified in brain cells as a physiological byproduct of tyramine. This study hypothesizes that 4-HPAA may regulate anxiety due to its anxiolytic properties, acting as a modulator of the GABAergic system, which plays a crucial role in the pathophysiology of anxiety disorders. Our study aims to enhance the anxiolytic effects of 4-HPAA through chemical modification to improve its pharmacokinetic properties. Three derivatives, namely Isopropyl-4-hydroxy-[phenyl] acetate (IHPA), Isopropyl-4-hydroxy-[phenyl] acetate (MPAA), and 4-methoxyphenyl acetate (MPHA), have been synthesized from 4-HPAA. This assessment will use well-established animal models, specifically the Elevated Plus-Maze (EPM) and Zero Maze (EZM) tests, selected for their validity in replicating anxiety-like symptoms in animals. Chronic caffeine administration via drinking water (0.3 g/l for 14 days) was employed to induce an anxiety state for testing purposes. IHPA and MPAA demonstrated significant anxiolyticactivity when tested in the EPM and EZM experiments. Molecular docking simulations using AutoDock Vina indicated that 4-HPAA derivatives had docking scores ranging from −5.8 to −4.8 kcal/mol, compared to the standard anxiolytic medication Diazepam, which scored −7.1 kcal/mol. These scores suggest a potential for 4-HPAA derivatives to interact effectively with the Gamma-aminobutyric acid (GABA_A) receptor. In conclusion, our in vivo and in silico analyses indicate a promising anxiolytic potential for 4-HPAA derivatives.

A Generalized Accelerated Failure Time Model to Predict Restoration Time from Power Outages

Major disasters such as wildfire, tornado, hurricane, tropical storm, and flooding cause disruptions in infrastructure systems such as power and water supply, wastewater management, telecommunication, and transportation facilities. Disruptions in electricity infrastructure have negative impacts on sectors throughout a region, including education, medical services,financial services, and recreation. In this study, we introduced a novel approach to investigate the factors that can be associated with longer restoration time of power service after a hurricane. Considering restoration time as the dependent variable and using a comprehensive set of county-level data, we estimated a generalized accelerated failure time(GAFT) model that accounts for spatial dependence among observations for time to event data. The model fit improved by 12% after considering the effects of spatial correlation in time to event data. Using the GAFT model and Hurricane Irma's impact on Florida as a case study, we examined:(1) differences in electric power outages and restoration rates among different types of power companies—investor-owned power companies, rural and municipal cooperatives;(2) the relationship between the duration of power outage and power system variables;and(3) the relationship between the duration of power outage and socioeconomic attributes. The findings of this study indicate that counties with a higher percentage of customers served by investor-owned electric companies and lower median household income faced power outage for a longer time. This study identified the key factors to predict restoration time of hurricane-induced power outages, allowing disaster management agencies to adopt strategies required for restoration process.

Mammalian sirtuins are seven members belonging to the silent information regulator 2 family, a group of Class Ⅲ histone/protein deacetylases. sirtuins(sIRT 1-7) have different subcellular localization and function and they regulate cellular protein function through various posttranslational modifications. sIRT1 and 3, the most studied sirtuins, use the product of cellular metabolism nicotinamide adenine dinucleotide as a cofactor to post-translationally deacetylate cellular proteins and consequently link the metabolic status of the cell to protein function. sirtuins have been shown to play a key role in the development and rescue of various metabolic diseases including non-alcoholic fatty liver disease(NAFLD). NAFLD is currently the most chronic liver disease due mainly to high-calorie consumption and lower physical activity. No pharmacological approach is available to treat NAFLD, the current recommended treatment are lifestyle modification such as weight loss through calorie restriction and exercise. Recent studies have shown downregulation of sirtuins in human as well as animal models of NAFLD indicating an important role of sirtuins in the dynamic pathophysiology of NAFLD. In this review, we highlight the recent knowledge on sirtuins, their role in NAFLD and their unique potential role as novel therapeutic target for NAFLD treatment.

spatiotemporal distribution and predictors of tuberculosis incidence in Morocco

Background:Tuberculosis(TB)is a major health problem in *** study aims at examining trends in TB in Morocco and identifying TB spatial clusters and TB-associated ***:Country-level surveillance data was ***’s correlation test was used to examine trends and an exploratory spatial data analysis was conducted to assess the global and local patterns of spatial autocorrelation in TB rates(Moran’s I and local indicator of spatial association[LIsA])at the prefecture/province *** including living in a prefecture versus living in a province,annual rainfall,annual mean temperature,population density,and AIDs incidence were *** ordinary least squares regression was thus performed and both spatial dependence and heteroscedasticity were ***:A decrease in TB incidence rate was seen between 1995 and 2014(Kendall’s tau b=−0.72;Pst years)was considered,TB rate remained stable and as high as 84 per 100000 population per year(95%CI:83.7-84.3).The highest incidence rates were seen in Tanger-Assilah,Fez,Tetouen-M’diq Fnidaq,Inezgane Ait Melleoul,and *** 2005 to 2014,while TB incidence rate was stable in Fez(P=0.500),Tetouen-M’diq Fnidaq(P=0.300),Casablanca(P=0.500),Mohammadia(P=0.146),Al Hoceima(P=0.364),and Guelmim(P=0.242),an increase in TB incidence rate was seen in Tanger-Assilah(Kendall’s tau=0.49;P=0.023)and a decrease in salé(Kendall’s tau b=−0,54;P=0.014)and Inezgane-Ait Melloul(Kendall’s tau b=−0,67;P=0.0023).TB is strongly clustered in space(P-values of Moran’s Istinct spatial regimes that affect TB spatial clustering were identified(east and west).In the east,both annual rainfall(P=0.003)and AIDs(P=0.0002)exert a statistically significant effect on TB *** the west,only the living area(prefecture versus province)was associated with TB rate(P=0.048).Conclusions:New information on TB incidence and TB-related predictors

Harmonic standing-Wave Excitations of simply-supported Thick-Walled Hollow Elastic Circular Cylinders:Exact 3D Linear Elastodynamic Response

The forced-vibration response of a simply-supported isotropic thick-walled hollow elastic circular cylinder subjected to two-dimensional harmonic standing-wave excitations on its curved surfaces is studied within the framework of linear *** semi-analytical solutions for the steady-state displacement field of the cylinder are constructed using recently-published parametric solutions to the Navier-Lam´e *** application of the standing-wave boundary conditions generates three parameter-dependent 66 linear systems,each of which can be numerically solved in order to determine the parametric response of the cylinder’s displacement field under various *** method of solution is direct and demonstrates a general approach that can be applied to solve many other elastodynamic forcedresponse problems involving isotropic elastic *** an application,and considering several examples,the obtained solution is used to compute the steady-state frequency response in a few specific low-order excitation *** each case,the solution generates a series of resonances that are in exact correspondence with a unique subset of the natural frequencies of the simply-supported *** considered problem is of general theoretical interest in structural mechanics and acoustics and more practically serves as a benchmark forced-vibration problem involving a thickwalled hollow elastic cylinder.

On a Novel Extended Lomax Distribution with Asymmetric Properties and Its statistical Applications

In this article,we highlight a new three-parameter heavy-tailed lifetime distribution that aims to extend the modeling possibilities of the Lomax *** is called the extended Lomax *** considered distribution naturally appears as the distribution of a transformation of a random variable following the logweighted power distribution recently introduced for percentage or proportion data analysis *** a result,its cumulative distribution has the same functional basis as that of the Lomax distribution,but with a novel special logarithmic term depending on several *** modulation of this logarithmic term reveals new types of asymetrical shapes,implying a modeling horizon beyond that of the Lomax *** the first part,we examine several of its mathematical properties,such as the shapes of the related probability and hazard rate functions;stochastic comparisons;manageable expansions for various moments;and quantile *** particular,based on the quantile functions,various actuarial measures are *** the second part,the distribution’s applicability is investigated with the use of themaximumlikelihood *** behavior of the obtained parameter estimates is validated by a simulation *** claim data are *** show that the proposed distribution outperforms eight well-known distributions,including the Lomax distribution and several extended Lomax *** addition,we demonstrate that it gives preferable inferences from these competitor distributions in terms of risk measures.

Warped product manifolds are known to have applications in physics. For instance, they provide an excellent setting to model space-time near a black hole or a massive star （cf. [9]）. The studies on warped product manifolds with extrinsic geometric point of view were intensified after the B.Y. Chen＇s work on CR-warped product submanifolds of Kaehler manifolds （cf. [6], [7]）. Later on, similar studies were carried out in the setting of 1.c.K. manifolds and nearly Kaehler manifolds （el. [3], [11]）. In the present article, we investigate a larger class of warped product submanifolds of 1.c.K. manifolds, ensure their existence by constructing an example of such manifolds and obtain some important properties of these submanifolds. With regard to the CR-warped product submanifold, a special case of generic warped product submanifolds, we obtain a characterization under which a CR-submanifold is reducesd to a CR-warped product submanifold.
