Objective In this study,we aimed to compare long-term physical and mental health outcome between SARS-CoV-2 infected and uninfected household members to differentiate between infection-related and pandemic-related outcomes after about two and a half years of the ***,possible differences in the outcome of adults and children and young people(CYp)were of *** In a cross-sectional study design,we compared the long-term physical and mental health outcome of between infected and uninfected as well as between adult and CYp(household members).Setting The FamilyCoviDD19 study—a serology study in households—was initially conducted to evaluate virus transmission in a close contact setting focusing on households with children and adolescents in *** least 1year after initial infection in the respective households,a follow-up took place in which the prevalence and type of possible long-term consequences were surveyed on the basis of self-reported information on physical and mental *** In this study,a total of 533 household members of 146 families participated and responded to our survey,including 296(55.5%)adults and 237(44.5%)*** The difference in frequency of reported symptoms between infected and uninfected individuals was very moderate,suggesting that the vast majority of reported symptoms were not attributable to a previous SARS-CoV-2 ***,regardless of age and infection status,this study showed overall high rates of self-reported symptoms with CYp having fewer long-term sequelae than adults oneyear after ***,over 50%of those reporting symptoms were not affected in their daily life,with CYps reporting an even lower percentage compared with *** CYp are at reduced risk not only to develop symptomatic infection or severe disease courses(previous analyses)but also to develop infection-associated long-term sequelae(this study).Independent of infection CYp reported high rates of

Dear Editor,Afferent synapses of cochlear inner hair cells(IHCs)employ a unique molecular machinery(see extended background in Supplementary Materials).Otoferlin is a key player in this machinery and its defects cause human auditory synaptopathy(Moser and Starr,2016).Otoferlin,a tail-anchored(Vogl et al.,2016)multi-C_(2)-domain protein(Fig.1Ai)specific to hair cells(Roux et al.,2006),is a member of the ferlin protein family involved in membrane trafficking and repair that are of major disease relevance(pangršičet al.,2012),also see Supplementary *** is distributed broadly within IHCs(Fig.2Ai-Aiii;pangrsic et al.,2010;Roux et al.,2006).

E-selectin ligand complexes adopt an extended high-affinity conformation

E-selectin is a cell-adhesion molecule of the vascular endothelium that promotes essential leukocyte rolling in the early inflammatory response by binding to glycoproteins containing the tetrasaccharide sialyl Lewisx(sLex).Efficient leukocyte recruitment under vascular flow conditions depends on an increased lifetime of E-selectin/ligand complexes under tensile force in a so-called catch-bond binding ***-crystal structures of a representative fragment of the extracellular E-selectin region with sLex and a glycomimetic antagonist thereof reveal an extended E-selectin conformation,which is identified as a high-affinity binding state of E-selectin by molecular dynamics ***-angle X-ray scattering experiments demonstrate a direct link between ligand binding and E-selectin conformational transition under static conditions in *** permits tracing a series of concerted structural changes connecting ligand binding to conformational stretching as the structural basis of E-selectin catch-bond-mediated leukocyte *** detailed molecular view of the binding site paves the way for the design of a new generation of selectin *** is of special interest,since their therapeutic potentialwas recently demonstrated with the pan-selectin antagonists GMI-1070(Rivipansel).

Tailoring the properties of the driving laser to the need of applications often requires compromises among laser stability,high peak and average power levels,pulse duration,and spectral *** instance,spectroscopy with optical frequency combs in the extreme/visible ultraviolet spectral region requires a high peak power of the near-IR driving laser,and therefore high average power,pulse duration of a few tens of fs,and maximal available spectral ***,the parametric conversion efficiency is higher for pulses with a duration in the 100-fs range due to temporal walk-off and coating *** we suggest an approach to adjust the spectral characteristics of high-power chirped-pulse amplification(CpA)to the requirements of different nonlinear frequency converters while preserving the low-phase-noise(pN)properties of the *** achieve spectral tunability,we installed a mechanical spectral shaper in a free-space section of the stretcher of an in-house-developed ytterbium-fiber-based CpA *** CpA system delivers 100 W of average power at a repetition rate of 132.4 *** gaining control over the spectral properties,we preserve the relative-intensity-noise and pN properties of the *** high-power CpA can easily be adjusted to deliver either a spectrum ideal for mid-IR light generation(full width at half maximum of∼11 nm,compressed pulse duration of 230 fs)or a spectrum ideal for highly nonlinear processes such as high-harmonic generation(−10 dB level of>50 nm,transform-limited pulse duration of∼65 fs).

Formation mechanisms of sub-micron pharmaceutical composite particles derived from far-and near-field Raman microscopy

Surface enhanced Raman spectroscopy(SERS) and confocal Raman microscopy are applied to investigate the structure and the molecular arrangement of sub-micron furosemide and polyvinylpyrrolidone(furosemide/pVp) particles produced by spray flash evaporation(SFE). Morphology, size and crystallinity of furosemide/pVp particles are analyzed by scanning electron microscopy(SEM) and X-ray powder diffraction(XRpD). Far-field Raman spectra and confocal far-field Raman maps of furosemide/pVp particles are interpreted based on the far-field Raman spectra of pure furosemide and pVp *** far-field Raman microscopy shows that furosemide/pVp particles feature an intermixture of furosemide and pVp molecules at the sub-micron scale. SERS and surface-enhanced confocal Raman microscopy(SECo RM) are performed on furosemide, pVp and furosemide/pVp composite particles sputtered with silver(40 nm). SERS and SECo RM maps reveal that furosemide/pVp particle surfaces mainly consist of pVp molecules. The combination of surface and bulk sensitive analyses reveal that furosemide/pVp sub-micron particles are formed by the agglomeration of primary furosemide nanocrystals embedded in a thin pVp matrix. Interestingly, both far-field Raman microscopy and SECo RM provide molecular information on a statistically-relevant amount of sub-micron particles in a single microscopic map;this combination is thus an effective and time-saving tool for investigating organic sub-micron composites.

Tissue factor in predicted severe acute pancreatitis

AIM: To study tissue factor (TF) in acute pancreatitis and evaluate the role of TF as a predictive marker of severity. METHODS: Forty-nine consecutive patients admitted to Lund University Hospital, fulfilling the criteria of predicted severe acute pancreatitis (Ap), were recruited prospectively between 2002 and 2004. Blood samples for TF analyses were drawn at inclusion in the study and 12 h, 1 d and 3 d later. RESULTS: Twenty-seven patients developed mild Ap, and 22 patients severe Ap. At inclusion in the study, the groups were comparable with respect to gender, aetiology, Acute physiology and Chronic Health Evaluation Ⅱ score, and duration of pain. At inclusion in the study and at 12 h, TF was higher in the severe Ap group (p = 0.035 and p = 0.049, respectively). After 1 and 3 d, no differences in TF levels were noted. Interleukin (IL)-6 was significantly higher in the severe Ap group at all of the studied time points. C-reactive protein (CRp) was significantly higher in the Ap group at 1 and 3 d. In receiver operating characteristic-curves, the area under the curve (AUC) for TF was 0.679 (p = 0.035) at inclusion in the study, and a cut off level for TF of 40 pg/mL showed a sensitivity of 71% and a specificity of 67%, whereas corresponding AUC for IL-6 was 0.775, p = 0.001, and for CRp was 0.653. IL-6 showed better AUC-values than TF at all time points studied. CONCLUSION: TF-levels are raised early in severe Ap. TF as an early predictive marker of severe Ap is superior to CRp, but inferior to IL-6.
