＜正＞ CKD-501 is a novel peroxisome proliferator-activated receptor α/γ dual agonist for the treatment of diabetes mellitus. This study aimed to investigate safety, tolerability, and pharmacokinetics of CKD-501 in healthy volunteers. A randomized, placebo-controlled, double-blind, par.llel-group, dose-rising study was performed. Thirty-six healthy male subjects received single oral doses ranging from 0.5-8 mg CKD- 501 or placebo. In the multipledose study, 24 subjects received 1-4 mg once daily for 7 days. Serial blood and urine samples were collected. No serious adverse events （AEs） were observed and AEs reported were all of mild severity. In the single-dose study, mean Cmax and AUCinf increased linearly up to the 8 mg dose *** and t1/2 ranged from 0.5-4 h and 7.8-9.8 h. respectively. Less than 1 % CKD-501 was excreted in urine. After multiple dosing, accumulation index was around 1.2. Mean appar.nt clearance, Tmax and t1/2 in steady state were independent of dose and time. Single oral doses up to 8 mg CKD- 501 and multiple doses up to 4 mg were safe and well tolerated. Mean Cmax and AUCinf were dose proportional and there was no remarkable accumulation after multiple dosing.

Atypical onset of bicalutamide-induced liver injury

Anti-androgen therapy is the leading treatment for advanced prostate cancer and is commonly used for neoadjuvant or adjuvant treatment. Bicalutamide is a non-steroidal anti-androgen, used during the initiation of androgen deprivation therapy along with a luteinizing hormone-releasing hormone agonist to reduce the symptoms of tumor-related flares in patients with advanced prostate cancer. As side effects, bicalutamide can cause fatigue, gynecomastia, and decreased libido through competitive androgen receptor blockade. Additionally, although not as common, drug-induced liver injury has also been reported. Herein, we report a case of hepatotoxicity secondary to bicalutamide use. Typically, bicalutamideinduced hepatotoxicity develops after a few days; however, in this case, hepatic injury occurred 5 mo after treatment initiation. Based on this rare case of delayed liver injury, we recommend careful monitoring of liver function throughout bicalutamide treatment for prostate cancer.

Interstitial lung disease （ILD） is a comprehensive term referring to a group of lung diseases affecting the interstitium of the lung. Idiopathic pulmonary fibrosis （IPF） is the most common idiopathic ILD, and nonspecific interstitial pneumonia （NSIP） is the second most common. As the name suggests, NSIP is diagnosed atter many other diseases are excluded. The main pathological finding in NSIP is homogeneous interstitial inflammation with or without fibrosis. NSIP can be categorized by cellular type or fibrotic type, according to the grade of inflammation and fibrosis. The cellular type has mostly inflammatory lesions with good responses to steroid, but the fibrotic type has a large proportion of fibrosis mixed with inflammatory lesions and a relatively poor response to steroid treatment So far, the exact mechanism underlying idiopathic lED has not been clarified. Determining key regulators of these ILDs will be helpful in the diagnosis and development of novel drugs for ILD.

背景与目的利妥昔单抗是一种靶向CD20的单克隆抗体,在治疗边缘区淋巴瘤(marginal zone lymphoma,MZL)中可以单药或联合化疗使用,可提高缓解率和生存率,但治疗后仍可能会复发。因此,仍需探索如何延长MZL患者的缓解期的治疗方案。这项多中心、单臂、开放标签的Ⅱ期研究评估了对一线R-CVP(rituximab,cyclophosphamide,vincristine,and prednisolone;利妥昔单抗、环磷酰胺、长春新碱和泼尼松龙)治疗有反应的Ⅲ–Ⅳ期CD20阳性MZL患者进行利妥昔单抗维持治疗2年的生存率。本研究旨在明确R-CVP后利妥昔单抗维持治疗的策略是否有进一步研究的价值。方法在利妥昔单抗维持治疗之前,Ⅲ–Ⅳ期MZL患者应接受6–8周期的R-CVP一线治疗。每个周期为3周,第1 d静脉输注利妥昔单抗(375 mg/m^2)、环磷酰胺(750 mg/m^2)和长春新碱(1.4 mg/m^2;最大2 mg),第1–5 d口服泼尼松龙(100 mg)。对R-CVP治疗达到完全缓解(complete response,CR)、部分缓解(par. response,PR)或疾病稳定(stable response,SD)的患者进行利妥昔单抗维持治疗,每8周静脉注射利妥昔单抗375mg/m^2,直到12个周期。主要终点为无进展生存期(progression-free surviva,PFS),次要终点为总生存期(overall survival,OS)和治疗安全性。结果共纳入47例患者,其中45(96%)例接受了利妥昔单抗维持治疗。15(33%)例患者有淋巴结MZL。在进行R-CVP一线治疗后,分别有20(44%)例、22(49%)例和3(7%)例的患者达到了CR、PR和SD。中位随访38.2个月后,患者的3年PFS率为81%。在利妥昔单抗维持治疗期间,R-CVP治疗后有6例PR和1例SD患者获得CR。LDH升高和B症状是PFS的重要预后因素(P=0.003),3年OS率为90%。利妥昔单抗维持治疗的耐受性良好,常见的治疗性不良事件为感觉性神经病(18%)、肌痛(13%)、疲劳(9%)和中性粒细胞减少(9%)。结论一线R-CVP治疗后利妥昔单抗维持治疗在Ⅲ–Ⅳ期MZL患者中不仅表现出较好的PFS,而且药物毒性反应评价良好。

Melatonin modulates adiponectin expression on murine colitis with sleep deprivation

AIM To determine adiponectin expression in colonic tissue of murine colitis and systemic cytokine expression after melatonin treatments and sleep *** The following five groups of C57BL/6 mice were used in this study:(1) group Ⅰ,control;(2) group Ⅱ,2% DSS induced colitis for 7 d;(3) group Ⅲ,2% DSS induced colitis and melatonin treatment;(4) group Ⅳ,2% DSS induced colitis with sleep deprivation(SD) using specially designed and modified multiple platform water baths; and(5) group V,2% DSS induced colitis with SD and melatonin treatment. Melatonin(10 mg/kg) or saline was intraperitoneally injected daily to mice for 4 d. The body weight was monitored daily. The degree of colitis was evaluated histologically after sacrificing the mice. Immunohistochemical staining and Western blot analysis was performed using anti-adiponectin antibody. After sampling by intracardiac punctures,levels of serum cytokines were measured by ELISA. RESULTS Sleep deprivation in water bath exacerbated DSS induced colitis and worsened weight loss. Melatonin injection not only alleviated the severity of mucosal injury,but also helped survival during stressful condition. The expression level of adiponectin in mucosa was decreased in colitis,with the lowest level observed in colitis combined with sleep deprivation. Melatonin injection significantly(P < 0.05) recovered the expression of adiponectin. The expression levels of IL-6 and IL-17 were increased in the serum of mice with DSS colitis but decreased after melatonin injection. CONCLUSION This study suggested that melatonin modulated adiponectin expression in colonic tissue and melatonin and adiponectin synergistically potentiated antiinflammatory effects on colitis with sleep deprivation.

Early Ordovician (Late Tremadoc) graptolites are procured from the nine sections of the Yeongwol area, Korea: the Dojang-gol, Ugga-gol, Karae-jae, Ppelchigol, Dae-gol, Myungjeon, Namaeri, Dumok, Namkyo Sections, which contain the boundary between the Mungok and Yeonghung Formations. The boundary between the Mungok and Yeongheung Formations reveals the ribbon rock, marlstone to shale, and grainstone to packstone facies of the Mungok Formation, and the thin laminated lime mudstone, bioturbated limestone, and wackestone to grainstone facies of the Yeongheung Formation.

薄膜晶体管液晶电视(TFT-LCDTV)因为具有薄、轻、紧凑和可随意放置的特点,已经占据了大部分电视机市场。除了这些物理特性以外,最重要的特性是已具有了良好像质的对比度。为了将对比度提高到1:600以上,对偏振片膜、背光源板、滤色片树脂、电极锥角和摩擦条件等都进行了研究。优化的背光板组合,光滑的电极锥角和摩擦方法的控制是提高对比度的主要控制因素。应用新开发的滤色片树脂,对获得高对比度最为有效。

将普遍应用于工业上的几种辅料,通过不同比例的混配并进行纳米级粉碎,加工成具有杀菌活性的纳米粉末,形状似黏土,它对革兰氏阴性菌、革兰氏阳性菌和真菌均有高效杀菌活性,这种纳米黏土既能应用到生物农药领域也有益于环境保护。实验室条件下测定了2个纳米黏土样品的不同质量浓度稀释液对烟草根黑腐菌、革兰氏阳性菌和革兰氏阴性菌的抑菌作用,对其抑菌效果进行了初步研究,以期为该纳米粘土作为生物农药的应用奠定理论基础。本试验选取了真菌——烟草根黑腐菌(Thielaviopsis basicola)、革兰氏阳性菌(Bacillusthuringiensis,Bt)和革兰氏阴性菌(*** TOP10)作为试验所需的病菌。将上述病菌经平板活化培养后,进行平板活菌计数。根据活菌计数结果,真菌取活菌数在100个和1 000个孢子/ml数量级的菌液备用,两株细菌均取活菌数在×10个/ml数量级的菌液备用。准确称取纳米黏土样品6mg和20mg,分别加入1 ml上述菌液,配成6mg/ml和20mg/ml的溶液。于30℃200rpm摇培作用3h后,取各处理200μl混合液与15ml PDA/LB培养基混匀倒平板,各设3个重复,30℃培养48h后检查菌落数。设3个菌的菌液200μl与PDA/LB培养基混合为空白对照。抑菌率%=(对照活菌数一样品处理活菌数)×100%/对照活菌数。试验结果表明:纳米黏土样品1和纳米黏土样品2对革兰氏阳性菌(Bacillus thuringiensis,Bt)和革兰氏阴性菌(*** TOP10)的抑菌试验中,样品1的抑菌效果很好,3h的抑菌率均达到99%以上,抑菌率随生物农药质量浓度的增加而增强;纳米黏土样品1和纳米黏土样品2对烟草根黑腐菌(Thielaviopsis basicola)的抑菌试验中,样品2的抑菌试验效果比较好,3h的抑菌率达到了82.90%,抑菌率随生物农药质量浓度的增加而增强。综上所述,在室内条件下,样品1的抑菌谱比较广,对革兰氏阴性菌和革兰氏阳性菌的抑菌效果都很好,样品2对烟草根黑腐菌的抑菌效果比较好。通过这2种纳米黏土的混合使用,可以有效地抑制大田中的各种病原菌,既可以杀灭各种细菌,对真菌也具有比较好的抑菌效果。

AIM:To determine whether magnified observation of short-segment Barrett’s esophagus(BE)is useful for the detection of specialized intestinal metaplasia(SIM).METHODS:Thirty patients with suspected short-segment BE underwent magnifying endoscopy up to×*** magnified images were analyzed with respect to their pit-patterns,which were simultaneously classified into five epithelial types[Ⅰ(small round),Ⅱ(straight),Ⅲ(long oval),Ⅳ(tubular),Ⅴ(villous)]by Endo’s ***,a 0.5%solution of methylene blue(MB)was sprayed over columnar *** patterns of the magnified image and MB staining were *** were obtained from the regions previously observed by magnifying endoscopy and MB ***:Three of five patients with a typeⅤ(villous)epithelial pattern had SIM,whereas 21 patients with a non-typeⅤepithelial patterns did not have *** sensitivity,specificity,accuracy,positive predictive value,and negative predictive value of pit-patterns in detecting SIM were 100%,91.3%,92.3%,60%and100%,respectively(P=0.004).Three of the 12 patients with positive MB staining had SIM,whereas 14patients with negative MB staining did not have *** sensitivity,specificity,accuracy,positive predictive value,and negative predictive value of MB staining in detecting SIM were 100%,60.9%,65.4%,25%and100%,respectively(P=0.085).The specificity and accuracy of pit-pattern evaluation were significantly superior compar.d with MB staining for detecting SIM by compar.son with the exact McNemar’s test(P=0.0391).CONCLUSION:The magnified observation of a shortsegment BE according to the mucosal pattern and its classification can be predictive of SIM.
