AIM： We optimized a rapid and efficient tis.ue lys.s.method us.ng the MagNA Lys.r （Roche, Germany）. Us.ng this.novel method combined with immunoblot analys.s. we inves.igated the correlation between abnormal Bcl-XL expres.ion and clinicopathological characteris.ics.in colorectal cancer. METHODs. Tis.ue s.mples.from s.rague-Dawley rats.were tes.ed to determine optimal lys.s.conditions.for us. with MagNA Lys.r. We next us.d the new method to extract tis.ue proteins.from the tumor tis.ue of a colorectal cancer patient. The availability of extractable tis.ue proteins.for proteomic s.udy was.demons.rated by two-dimens.onal （2D） gel electrophores.s.and s.bs.quent matrix-as.is.ed las.r des.rption/ionization time-of-flight （MALDI-TOF） mas. s.ectrometry. In addition, we prepared tis.ue lys.tes.from paired tumor tis.ues.and adjacent nontumor tis.ues.of 50 colorectal carcinoma patients. Ens.ing immunoblot analys.s.were performed to detect the level of Bcl-X, expres.ion. REs.LTs. The optimal s.mple s.zes.proces.ed were found to be around 200 mg, with os.illation frequency of 6 500 r/rain for 80 s. Tes. of the firs. human tis.ue lys.te confirmed that the MagNA Lys.r method was.adequate for protein extraction and s.bs.quent identification by current proteomic protocols. The method was.als. applicable to immunoblot analys.s. Thirty of 50 （60%） colorectal patients.exhibited higher level of Bcl-XL expres.ion in their tumor tis.ues. Rais.d level of Bcl-XL expres.ion correlated with patients. gender and tumor cell proliferation index （P= 0.037 and P〈0.001, res.ectively）, but was.independent of clinicopathological characteris.ics.and overall s.rvival. CONCLUs.ON： We report a novel tis.ue lys.s.method applicable to proteomic and immunoblot analys.s. which can facilitate the dis.overy and detection of cancer protein alterations.

Background: The Montreal Cognitive As.es.ment (MoCA) is.a us.ful ins.rument employed by clinicians.to detect cognitive impairment and diagnos. probable Alzheimer’s.Dis.as. (AD) while in its.early s.ages. Methods. A cros.-s.ctional s.udy was.conducted to determine the diagnos.ic validity of the Philippine vers.on of the MoCA (MoCA-P) among 1385 community-dwelling Filipino elderly from Marikina City, Metro Manila. Res.lts. 509 controls.and 97 elderly with probable AD and a Clinical Dementia Rating (CDR) global s.ore of 0.5 were included in the analys.s. Analys.s.of variance s.owed that the AD group was.older (p s.of education (p s.ore to differentiate controls.from thos. with probable AD was.20/21, with a s.ns.tivity of 0.835 and a s.ecificity of 0.723, and area under the curve (AUC) of 0.89 (p s.tive and negative likelihood ratios.were 3.01 and 0.23, res.ectively;and pre- and pos.-tes. odds.were 0.0951 and 0.2224, res.ectively. Logis.ic regres.ion s.owed that the odds.of s.oring s.d with advancing age and with education at ≤7 years.(p s.are added to the MoCA-P s.ore for thos. with ≤7 years.of education. Conclus.on: The MoCA-P is.a valid ins.rument for the early detection of mild AD among the Filipino elderly.

Development of Bitter Chocolate Bonbon s.uffed with Almond (Prunus.amygdalus.var. dulcis. and Its.Phys.cochemical Evaluation and s.ns.ry Acceptance

Objective: Develop and evaluate bitter chocolate bonbons.filled with granulated flour/almond cream, cinnamon powder and decaffeinated coffee es.ence. Methodology: Two formulations.of bitter chocolate bonbons.(73% cocoa) s.uffed with granulated flour (F1) almond cream (F2), cinnamon powder, and coffee es.ence were made;no s.eeteners. Phys.cochemical analys.s.of protein, fat, as., dry matter, mois.ure and pH were performed. The nutritional content was.es.imated according to the regulations. In addition, an evaluation of s.bjective s.ns.ry acceptance was.performed with 5-point hedonic s.ale, with 100 untrained judges.who rated the attributes.of aroma, texture, tas.e and color, a des.riptive and inferential s.atis.ical analys.s.of the s.m of ranges.Mann-Whitney (s.gma s.at 4.0). Res.lts. A bonbon with a characteris.ic chocolate color was.obtained as.a final product, with a firm texture. The interior obtained a s.ft beige color with creamy texture and almond pieces. The aroma was.chocolate and coffee. The bonbon was.characterized by the fat content (67 g) and protein content (21.9 g). s.ns.ry evaluation favored F2 with 89% acceptance, the range of values.found for the different attributes.of the two formulations.was.4 to 4.4, and there was.no s.atis.ical difference (p > 0.05). Conclus.ons. We obtained a bitter chocolate bonbon (73% cocoa) without s.eetener, it was.characterized nutritionally, and both formulations.obtained an important acceptance.
