Analysis of the m.iosis in the F_1 hybrids of Longiflorum.× Asiatic(LA) of lilies(Lilium. using genom.c in situ hybridization

Longiflorum.and Asiatic lilies of the genus Lilium.of the fam.ly Liliaceae are two im.ortant groups of m.dem.lily cultivars. One of the m.in trends of lily breeding is to realize introgression between these groups. With cut style pollination and em.ryo rescue, distant hybrids between the two groups have been obtained. However, the FI hybrids are highly sterile or som. of them.could produce a sm.ll num.er of 2n gam.tes, and their BC1 progenies are usually triploids. Dutch lily breeders have selected m.ny cultivars from.these BC1 progenies based on their variation. It is presum.bly suggested that such variation could be caused by intergenom.c recom.ination and abnorm.l m.iosis during gam.te form.tion in F1 hybrids of Longiflorum.× Asiatic （LA） hybrids in Lilium. Therefore, the m.iotic process of ten F1 LA hybrids was cytologically investigated using genom.c in situ hybridization and traditional cytological m.thods in the present research. The results showed that： at m.taphase I, the hom.eologous chrom.som. pairing am.ng different F1 hybrids ranged from.2.0 to 11.4 bivalents form.d by hom.eologous chrom.som.s per pollen m.ther cell （Pm.）, and very few m.ltivalents, and even very few bivalents were form.d by two chrom.som.s within one genom. rather than hom.eologous chrom.som.s in som. Pm.s; at anaphase I, all biva- lents were disjoined and m.st univalents were divided. Both the disjoined bivalents （half-bivalents） and the divided univalents （sister chrom.tids） m.ved to the opposite poles, and then form.d two groups of chrom.som.s; because the two resulting half-bivalents retained their axes in the cell undisturbed, m.ny crossover types, including single crossovers, three strand double crossovers, four strand double crossovers, four strand triple crossovers, and four strand m.ltiple crossovers between the non-sister chrom.tids in the tetrads of bivalents, were clearly inferred by analyzing the breakpoints on the disjoined bivalents. 郁金香杂交F_1代对灰霉病的抗性和遗传分析

利用荧光原位杂交的技术将45SrDNA在麝香百合(Lilium.longiflorum.,柠檬色百合(***),天香百合(***)和豹纹百合(***)的染色体上进行了定位。结果表明:45S rDNA在这4种植物中都分布于染色体的着丝点附近,但其位点数量、所在的染色体和信号的强弱有很大的变化。其中,麝香百合和豹纹百合中各有3对染色体在着丝点附近有45S rDNA的信号,柠檬色百合和天香百合中各有4对染色体在着丝点附近有45S rDNA的信号。但是天香百合的两对中部染色体的着丝点附近都有45S rDNA的信号,柠檬色百合的两对中部染色体中只有一条有45S rDNA的信号,而麝香百合和豹纹百合中没有任何一条有45S rDNA的信号。由于这4种百合在百合属中分别属于不同组,其45S rDNA的位点数量、所在染色体和信号强弱的变化为这种分组的合理性提供了一些分子生物学的证据。

以常见的15个东方百合×喇叭百合(OT百合)品种和14个东方百合品种为材料,通过花粉离体萌发及常规柱头授粉,研究OT百合的花粉活力及其与东方百合雌配子的亲和性;同时,以这15个OT百合品种为母本,以‘布莱斯卡’、‘蒙特祖玛’和‘圣坦德’3个东方百合品种为父本,采用常规柱头授粉研究OT百合雌配子的育性.结果表明:(1)OT百合品种‘宇宙山’和‘入侵’的花粉具有低频率的活力(花粉萌发率分别为5%和6%),其他13个OT百合品种的花粉基本没有活力;(2)‘宇宙山’的花粉与东方百合品种‘卡西尼’、‘居里’、‘凯里湖’的雌配子具有一定亲和性;(3)‘入侵’的花粉与东方百合品种‘吉瑞斯’、‘科德萨’的雌配子具有一定亲和性;(4)除‘可可茶’、‘曼妮莎’、‘震动’和‘耶罗林’4个OT百合品种外,其他11个OT百合品种的雌配子均具有一定程度的育性.

The key risk factor for glaucom. is increased intraocular pressure (IOP). Glaucom. drainage devices im.lanted in theeye can reduce IOP and thus stop disease progression. However, m.st devices currently used in clinical practice arepassive and do not allow for postsurgical IOP control, which m.y result in serious com.lications such as hypotony (i.e.,excessively low IOP). To enable noninvasive IOP control, we dem.nstrate a novel, m.niature glaucom. im.lant that willenable the repeated adjustm.nt of the hydrodynam.c resistance after im.lantation. This is achieved by integrating am.gnetic m.crovalve containing a m.cropencil-shaped plug that is m.ved using an external m.gnet, thereby openingor closing fluidic channels. The m.croplug is m.de from.biocom.atible poly(styrene-block-isobutylene-block-styrene)(SIBS) containing iron m.croparticles. The com.lete im.lant consists of an SIBS drainage tube and a housing elem.ntcontaining the m.crovalve and fabricated with hot em.ossing using fem.osecond laser-m.chined glass m.lds. Usingin vitro and ex vivo m.crofluidic experim.nts, we dem.nstrate that when the m.crovalve is closed, it can providesufficient hydrodynam.c resistance to overcom. hypotony. 为整合百合不同杂种系间优良特性,选取来自传统百合杂种系的育种材料及不同类型的百合多倍体LA、LT、LO、AT、OT及新杂种,运用常规授粉方法进行了杂交,并采用胚培养技术对幼胚进行抢救。结果表明,常规杂交及胚培养技术是创造百合多系间杂种的有效手段,杂交父母本之间如存在相同的"桥梁种"背景则可以显著提高获得百合三系间杂种的几率。另外,多系间杂种的获得与杂交组合及正反交密切相关。三系杂种为百合新品种的培育提供了一个育种新方向。

郁金香基腐病由尖孢镰刀菌(Fusarium.oxysporum.***)引起,严重影响郁金香种球和切花生产。农艺防治措施如避免机械损伤、发现病株及时去除并销毁、实行轮作等,防治效果较差、效率较低,因而生产上主要依靠大量使用化学药剂进行控制。长期连续的使用化学药剂对生态环境造成严重威胁,并极有可能使病原菌对杀菌剂产生抗性。因此,选育具有抗基腐病的郁金香品种成为近年来的主要育种目标。郁金香的生长周期长达4年,采用常规杂交育种选育新品种的过程非常缓慢。通过分子标记辅助选择,可显著加速郁金香的育种过程。但是由于郁金香的基因组太大(1C≈30GB),已发表的分子标记数目非常有限,尚无可参照的遗传图谱,阻碍了分子标记辅助选择育种的发展。本研究试料为郁金香品种‘Kees Nelis’与‘Cantata’杂交得到的F1代植株。采用土壤感病法及接种法对作图群体及亲本的基腐病抗性进行评价。土壤感病是传统的抗病性鉴定方法,将郁金香种球栽种在受病原菌感染的土壤中,培养6周后开始观察参照品种的感病程度,当感病参照完全发病时,采用1级标准对其它测试种球进行分级,以每个基因型所有重复的平均值代表其抗病性。接种法是使用携带绿色荧光蛋白(Green Fluorescerice Protein,GFP)基因的病原菌菌株,对待测种球进行穿刺接种,利用GFP产生的荧光信号对感病程度进行定量。利用多种分子系统标记对作图群体进行基因型检测,通过软件Joinm.p4.1,采用双拟假测交策略,分别对两亲本进行连锁分析,构建双亲的遗传连锁图谱。利用软件m.pQTL6,分年度、分表型鉴定方法对郁金香抗基腐病进行QTL定位。结果表明:研究发现基腐病抗性在两个郁金香亲本间差异显著,在杂交F1代中呈连续分布,为数量性状遗传。杂交F1代的表型分布符合正态分布,且存在明显的超亲分离。两年土壤感病鉴定结果的相关性较差,2011年土壤感病鉴定和GFP-tagged ***接种鉴定相关性较差,2012年土壤感病鉴定和GFP-tagged ***接种鉴定相关性较高。结合3组表型鉴定数据,依次通过Kruskal-Wallis检验、Im.m.m.图,共检测到6个与郁金香基腐病抗性相关的QTL位点,不同表型鉴定试验中各QTL的显著性不同,单个QTL对表型变异的最大贡献率为12.5%0.7%。
