Impact of Fusobacterium nucleatum in the gastrointestinal tract on natural killer cells

BAcKGROUND Gut microbial dysbiosis contributes to the development and progression of colorectal cancer(cRc).Natural killer(NK)cells are involved in early defense mechanisms to kill infective pathogens and tumor cells by releasing chemokines and *** better understand the relationship between the gut microbiome and cRc,it was hypothesized here that a high abundance of Fusobacterium nucleatum(***)in the gastrointestinal tract could cause reduced NK cell *** To identify associations between gastrointestinal tract *** levels and NK cell *** In vitro experiments were performed on NK cells treated with ***,Peptostreptococcus anaerobius,and Parvimonas micra to identify the effects of gut microbiome species on NK *** 24 and 48 h of treatment,NK cell counts were *** parallel studies,c57BL/6 mice were given broadspectrum antibiotics in their drinking water to reduce resident gut *** 3 wk,the mice received the various bacterial species or phosphate-buffered saline(PBs)via oral gavage every 2 d for 6 *** the study end,blood samples were acquired to perform NK cell activity assessment and cytokine *** tissues were collected and analyzed via immunohistochemistry(IHc).REsULTs The data show that after 3 wk of broad-spectrum antibiotic treatment,levels of total bacteria and *** were markedly decreased in *** of *** significantly decreased NK cell activity relative to the activities of cells from mice treated with antibiotics only and *** administration of *** decreased the proportion of NK46+cells based on IHc staining and increased the production of interleukin-1βand tumor necrosis factor-α.cONcLUsION High levels of *** in the gastrointestinal tract reduced NK cell activity in mice,and the decrease in NK cell activity might be affected by increased proinflammatory cytokines after *** treatment.

Tai chi for Management of Type 2 Diabetes Mellitus:A systematic Review

Objective： Tai chi has been recommended for treating type 2 diabetes mellitus. The purpose of this systematic review was to evaluate evidence from controlled clinical trials testing the effectiveness of tai chi in treating type 2 diabetes mellitus. Methods： systematic searches were conducted on 14 electronic databases without restrictions on either population characteristics or language of publication. The outcome measures considered for inclusion were changes in fasting blood glucose （FBG）, glycosylated haemoglobin A1c （HbA1c） and quality of life （QOL）. Results： Eight randomised clinical trials （RcTs） and two controlled clinical trials （ccTs） met all inclusion criteria. Three RcTs from 1 trial compared the effects of tai chi with sham exercise and failed to show effectiveness of tai chi on FBG, HbA1c, or QOL. The other 3 RcTs tested the effects of tai chi compared with other types of exercise on FBG. The meta-analysis failed to show an FBG-lowering effect of tai chi [n=118, weighted mean difference （WMD）： -0.14 mmol/L, 95% cI：-0.86 to 0.58,P=0.70]. Four studies （2 RcTs and 2 ccT） compared tai chi with no treatment or self-management programme and failed to report significant differences between the experimental and control groups except for QOL from 1 RcT and 1 ccT. conclusion： The existing evidence does not suggest that tai chi is an effective therapy for type 2 diabetes. currently, there are few high-quality trials on which to make definitive judgements.

Diagnosis,treatment and complications of radial head and neck fractures in the pediatric patient

Radial head and neck fractures represent up to 14%of all pediatric elbow fractures and can be a difficult challenge in the pediatric *** up to 39%of proximal radius fractures,there is a concomitant fracture,which can easily be overlooked on the initial standard *** treatment options for proximal radius fractures in children range from non-surgical treatment,such as immobilization alone and closed reduction followed by immobilization,to more invasive options,including closed reduction with percutaneous pinning and open reduction with internal *** choice of treatment depends on the degree of angulation and displacement of the fracture and the age of the patient;an angulation of less than 30 degrees and translation of less than 50%is generally accepted,whereas a higher degree of displacement is considered an indication for surgical *** with limited displacement and non-surgical treatment generally result in superior outcomes in terms of patient-reported outcome measures,range of motion and complications compared to severely displaced fractures requiring surgical *** proper management,good to excellent results are achieved in most cases,and long-term sequelae are ***,severe complications do occur,including radio-ulnar synostosis,osteonecrosis,rotational impairment,and premature physeal closure with a malformation of the radial head as a result,especially after more invasive *** follow-up is therefore warranted.

Treatment satisfaction after I year high-power potassiumtitanyl-phosphate photoselective vaporization of the prostate

To investigate the factors that influence treatment satisfaction after high-power potassium-titanyl-phosphate （KTP） laser vaporization of the prostate, we compared the characteristics between patients who were satisfied and those who were not satisfied. In all, 97 patients aged between 53-82 years （median age 67 years） underwent high-power KTP laser vaporization of the prostate for lower urinary tract symptoms due to benign prostatic hyperplasia. At 12 months postoperatively, 60 patients were satisfied with the treatment, whereas 37 were dissatisfied. Although there were no differences in International Prostate symptom score （IPss） values at baseline, the satisfied group scored better in total IPss at 1, 3, 6, and 12 months postoperatively （P 〈 0.05）. At baseline, the maximum flow rate （Qmax） was lower in the dissatisfied group and remained low throughout the follow-up period, with the exception of 1 month postoperatively （P 〈 0.05）, compared with the satisfied group. There were no differences in other objective data between the two groups, including post-void residual and the number of voids based on the frequency-volume charts. In a multivariate model, a higher bladder contractility index was associated with a greater likelihood of treatment satisfaction 12 months after high-power KTP laser vaporization （odds ratio 1.024, 95% confidence interval 1.001-1.048, P 〈 0.05）. Patients who were not satisfied following the surgery had a smaller improvement in subjective symptoms and Qmax. In addition, our findings suggest that the relative risk of treatment dissatisfaction following high-power KTP laser vaporization was increased in patients with weak detrusor contractility.

Aim： To investigate whether the biological process of superparamagnetic iron oxide （sPIO）-labeled human mesenchymal stem cells （hMscs） may be monitored non-invasively by using in vivo magnetic resonance （MR） imaging with conventional 1.5-T system examinations in corpus cavernosa of rats and rabbits. Methods： The labeling efficiency and viability of sP10-labeled hMscs were examined with Prussian blue and Tripan blue, respectively. After sPIO-labeled hMscs were transplanted to the corpus cavernosa of rats and rabbits, serial T2-weighted MR images were taken and histological examinations were carried out over a 4-week period. Results： hMscs loaded with sPIO compared to unlabeled cells had a similar viability. For sPIO-labeled hMscs more than lx 105 concentration in vitro, MR images showed a decrease in signal intensity. MR signal intensity at the areas of sPIO-labeled hMscs in the rat and rabbit corpus cavernosa decreased and was confined locally. After injection of sPIO-labeled hMscs into the corpus cavernosum, MR imaging demonstrated that hMscs could be seen for at least 12 weeks after injection. The presence of iron was confirmed with Prussian blue staining in histological sections. conclusion： sP10-labeled hMscs in corpus cavernosa of rats and rabbits can be evaluated non-invasively by molecular MR imaging. Our findings suggest that MR imaging has the ability to test the long-term therapeutic potential of hMscs in animals in the setting of erectile dysfunction.

Presence of ccK-A, B receptors and effect of gastrin and cholecystokinin on growth of pancreatobiliary cancer cell lines

AIM: To investigate the effects of gastrin and cholecystokinin (ccK) and their specific antagonists on the growth of pancreatic and biliary tract cancer cell lines. METHODs: Five pancreatic and 6 biliary cancer cell lines with 2 conrtol cells were used in this study. cell proliferation study was done using 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyl tetrazolium bromide (MTT) test and direct cell count method. Reverse transcription-polymerase chain reaction (RT-PcR) and slot blot hybridization were performed to examine and quantify the expression of hormonal receptors in these cell lines. REsULTs: sNU-308 showed a growth stimulating effect by gastrin-17, as did sNU-478 by both gastrin-17 and ccK-8. The trophic effect of these two hormones was completely blocked by specific antagonists (L-365, 260 for gastrin and L-364, 718 for ccK). Other cell lines did not respond to gastrin or ccK. In RT-PcR, the presence of ccK-A receptor and ccK-B/gastrin receptor mRNA was detected in all biliary and pancreatic cancer cell lines. In slot blot hybridization, compared to the cell lines which did not respond to hormones, those that responded to hormones showed high expression of receptor mRNA. cONcLUsION: Gastrin and ccK exert a trophic action on some of the biliary tract cancers.

Bamboo salt suppresses skin inflammation in mice with 2, 4-dinitrofluorobenzene-induced atopic dermatitis

Bamboo salt(Bs) is a traditional Korean food, and has been reported to have anti-cancer, anti-inflammatory, and anti-metastatic effects. However, the anti-atopic dermatitis(AD) activity of Bs has not been described yet. In the present study, we examined the preventive effect of Bs on AD. The effect of oral administration of Bs was tested in a 2, 4-dinitrofluorobenzene(DNFB)-induced AD animal model, by histological analysis, enzyme-linked immunosorbent assay, reverse transcription-polymerase chain reaction, caspase-1 assay, and Western blotting analysis. Bs administration reduced the total clinical severity and scratching frequencies, compared with the AD group. In the serum of DNFB-induced AD mice, the levels of IgE, histamine, thymic stromal lymphopoietin(TsLP), interleukin(IL)-5, and IL-13 were significantly reduced by Bs treatment. Bs significantly reduced the protein and mRNA expression of TsLP, IL-6, and tumor necrosis factor-α in the AD skin lesions. Bs markedly reduced the infiltration of inflammatory cells. Furthermore, the activation of caspase-1 was reduced by Bs in the AD skin lesions. Our results suggested that Bs should be considered as a candidate treatment for allergic inflammatory diseases including AD.

Promoter hypomethylation and reactivation of MAGE-A1 and MAGE-A3 genes in colorectal cancer cell lines and cancer tissues

AIM： To verify the expression and methylation status of the MAGE-A1 and MAGE-A3 genes in colorectal cancer tissues and cancer cell lines. METHODs： We evaluated promoter demethylation status of the MAGE-A1 and MAGE-A3 genes by RT-PcR analysis and methylation-specific PcR （Ms-PcR）, as well as sequencing analysis, after sodium bisulfite modification in 32 colorectal cancer cell lines and 87 cancer tissues. REsULTs： Of the 32 cell lines, MAGE-A1 and MAGE-A3 expressions were observed in 59% and 66%, respectively. subsequent to sodium bisulfite modification and MsPcR analysis, the promoter hypomethylation of MAGE-A1 and MAGE-A3 was confirmed in both at 81% each. Promoter hypomethylation of MAGE-A1 and MAGE-A3 in colorectal cancer tissues was observed in 43% and 77%, respectively. Hypomethylation of MAGE-A1 and MAGE-A3 genes in corresponding normal tissues were observed in 2% and 6%, respectively. cONcLUsION： The promoter hypomethylation of MAGE genes up-regulates its expression in colorectal carcinomas as well as in gastric cancers and might play a significant role in the development and progression of human colorectal carcinomas.
