A multi-shot target assembly and automatic alignment procedure for laser–plasma proton acceleration at high repetition rate are *** assembly is based on a multi-target rotating wheel capable of hosting more than 5000 targets,mounted on a 3D motorized stage to allow rapid replenishment and alignment of the target material between laser *** automatic alignment procedure consists of a detailed mapping of the impact positions at the target surface prior to the irradiation that ensures stable operation of the target,which alongside the purpose-built design of the target wheel,enables operation at rates up to 10 *** and continuous laser-driven proton acceleration at 10 Hz is demonstrated,with observed cut-off energy stability about 15%.

目的探讨山楂叶总黄酮对血管紧张素Ⅱ(AngⅡ)诱导的心肌肥大的影响,及其可能作用机制。方法将H9c2细胞分为对照组(正常培养),模型组(100 nmol·L^(-1)AngⅡ处理24 h),低、中、高剂量实验组(先给予100 nmol·L^(-1)AngⅡ处理24 h后,再分别给予25、50、100 mg·L^(-1)的山楂叶总黄酮处理24 h),anti-miR-NC、anti-miR-21a-5p组(分别转染anti-miR-NC、anti-miR-21a-5p后,给予100 nmol·L^(-1)AngⅡ处理24 h),miR-NC+高剂量组和miR-21a-5p+高剂量组(分别转染miR-NC、miR-21a-5p mimics,再给予100 nmol·L^(-1)AngⅡ+100 mg·L^(-1)山楂叶总黄酮各处理24 h)。用CCK-8法和流式细胞术检测细胞活力和凋亡情况,用实时荧光定量聚合酶链反应法检测miR-21a-5p的表达水平,用蛋白质印迹法检测环氧化酶-2(COX2)/前列腺素E2(PGE2)的表达水平。结果对照组、模型组和高剂量实验组的细胞活力分别为1.03±0.09、0.51±0.05和0.93±0.08,凋亡率分别为(7.69±0.61)%、(23.04±1.82)%和(9.43±0.71)%,miR-21a-5p表达水平分别为1.00±0.09、2.43±0.18和1.09±0.08,COX2蛋白相对表达水平分别为0.42±0.03、0.85±0.08和0.40±0.04,PGE2蛋白相对表达水平分别为0.34±0.03、0.75±0.07和0.35±0.03,模型组的上述指标与高剂量实验组和对照组相比,在统计学上差异均有统计学意义(均PmiR-NC组、anti-miR-21a-5p组、miR-NC+高剂量组和miR-21a-5p+高剂量组的细胞活力分别为0.52±0.04、1.12±0.08、0.94±0.09和0.57±0.04,凋亡率分别为(23.04±1.82)%、(9.86±0.73)%、(9.47±0.64)%和(24.96±1.94)%,miR-21a-5p表达水平分别为1.00±0.10、0.43±0.04、1.00±0.09和2.12±0.18,COX2蛋白相对表达水平分别为0.86±0.05、0.39±0.04、0.41±0.03、0.78±0.07,PGE2蛋白相对表达水平分别为0.74±0.06、0.38±0.07、0.36±0.02、0.71±0.05,anti-miR-21a-5p组的上述指标与anti-miR-NC组比较,miR-21a-5p+高剂量组的上述指标与miR-NC+高剂量组比较,在统计学上差异均有统计学意义(均PmiR-21a-5p和COX2/PGE2通路的表达,从而抑制AngⅡ诱导的心肌肥大。

The influence of ultrasonic loading on reverse current-voltage characteristics of mo/n-n＋-Si structures has been investigated. The research of leakage current variation has been carried out for various ultrasonic wave frequencies （4.1 and 8.4 mHz）, intensities （up to 0.8 W/cm2） and loading temperatures （130-330 K）. The observed reversible acoustically induced increase in reverse currents was as large as 60%. It has been found that dominant charge transfer mechanisms are the thermionic emission （at high temperature） and the phonon-assisted tunneling （at low temperature）. The ultrasound loading affects both processes due to the decrease of Schottky barrier height and binding energy of the electron on the trap.

The CELLULOSE SYNTHASE-LIKE C （CSLC） family is an ancient lineage within the CELLULOSE SYNTHASE/CELLULOSE SYNTHASE-LIKE （CESA/CSL） polysaccharide synthase superfamily that is thought to have arisen before the divergence of mosses and vascular plants. As studies in the flowering plant Arabidopsis have suggested synthesis of the （1,4）-β-glucan backbone of xyloglucan （XyG）, a wall polysaccharide that tethers adjacent cellulose microfibrils to each other, as a probable function for the CSLCs, CSLC function was investigated in barley （Hordeum vulgate L.）, a species with low amounts of XyG in its walls. Four barley CSLCgenes were identified （designated HvCSLC1-4）. Phylogenetic analysis reveals three well supported clades of CSLCs in flowering plants, with barley having representatives in two of these clades. The four barley CSLCs were expressed in various tissues, with in situ PCR detecting transcripts in all cell types of the coleoptile and root, including cells with primary and secondary cell walls. Co-expression analysis showed that HvCSLC3 was coordinately expressed with putative XyG xylosyltransferase genes. Both immuno-Em and membrane fractionation showed that HvCSLC2 was located in the plasma membrane of barley suspension-cultured cells and was not in internal membranes such as endoplasmic reticulum or Golgi apparatus. Based on our current knowledge of the sub-cellular locations of polysaccharide synthesis, we conclude that the CSLC family probably contains more than one type of polysaccharide synthase.

metformin(met)is a drug developed for the treatment of patients with typeⅡ ***,met estimation in pharmaceutical formulations and human fluids has gained a growing *** extend requisite data that can be used to assessment of met quantitatively based on charge-transfer(CT)complexation,the present study describes the synthesis and characterization of CT complexes that formed between drug met and the organicπ-acceptors picric acid(PA),chloranilic acid(CLA),chloranil(CHL),7,7',8,8'-tetracyanoquinodimethane(TCNQ),and dichlorodicyanobenzoquinone(DDQ).The properties of the formed CT complexes were investigated by elemental,spectral(UV-visible,IR,and Raman spectroscopies),thermal(TG)and morphological(SEm)*** results indicated that the complexation of met with either PA or CLA acceptors occurs through proton transfer interaction,whereas its complexation with CHL,TCNQ,or DDQ acceptors occurs through n→π*interaction.

Cadmium(Cd)induces several effects in different tissues,but our knowledge of the toxic effects on organelles is insuffi *** observe the progression of Cd effects on organelle structure and function,HuH-7 cells(human hepatic carcinoma cell line)were exposed to CdCl2 in increasing concentrations(1μm–20μm)and exposure times(2 h–24 h).During Cd treatment,the cells exhibited a progressive decrease in viability that was both time-and *** treated cells displayed progressive morphological changes that included cytoplasm retraction and nuclear condensation preceding a total loss of cell *** with 10μm for 12 h led to irreversible *** these drastic and irreparable damages,treated cells(5μm for 12 h)presented a progressive loss of mitochondrial function and cytoplasm acidifi cation as well as dysfunction and disorganization of microfi laments and endoplasmic *** damages led to the induction of apoptotic events and an increase in autophagic bodies in the *** results revealed that Cd affects multiple intra-cellular targets that induce alterations in the mitochondria,cytoskeleton,endoplasmic reticulum and acidic compartments,ultimately culminating in cell death via apoptotic and autophagic pathways.
