Diagnosing, planning and evaluating osteochondral ankle defects with imaging modalities

This current concepts review outlines the role of diffe-rent imaging modalities in the diagnosis, preoperative planning, and follow-up of osteochondral ankle defects. An osteochondral ankle defect involves the articular cartilage and subchondral bone(usually of the talus) and is mostly caused by an ankle supination trauma. Conventional radiographs are useful as an initial imaging tool in the diagnostic process, but have only moderate sensitivity for the detection of osteochondral defects. Computed tomography(CT) and magnetic resonance imaging(mRI) are more accurate imaging modalities. Recently, ultrasonography and single photon emission CT have been described for the evaluation of osteochondral talar defects. CT is the most valuable modality for assessing the exact location and size of bony lesions. Cartilage and subchondral bone damage can be visualized using mRI, but the defect size tends to be overestimated due to bone edema. CT with the ankle in full plantar flexion has been shown a reliable tool for preoperative planning of the surgical approach. Postoperative imaging is useful for objective assessment of repair tissue or degenerative changes of the ankle joint. Plain radiography, CT and mRI have been used in outcome studies, and different scoring systems are available.

In plants, resistance to necrotrophic pathogens depends on the interplay between different hormone systems, such as those regulated by salicylic acid （SA）, jasmonic acid （ja）, ethylene, and abscisic acid. Repression of auxin signaling by the SA pathway was recently shown to contribute to antibacterial resistance. Here, we demonstrate that Arabidopsis auxin signaling mutants axrl, axr2, and axr6 that have defects in the auxin-stimulated SCF （Skpl-Cullin- F-box） ubiquitination pathway exhibit increased susceptibility to the necrotrophic fungi Plectosphaerella cucumerina and Botrytis cinerea. Also, stabilization of the auxin transcriptional repressor AXR3 that is normally targeted for removal by the SCF-ubiquitin/proteasome machinery occurs upon P. cucumerina infection. Pharmacological inhibition of auxin transport or proteasome function each compromise necrotroph resistance of wild-type plants to a similar extent as in non-treated auxin response mutants. These results suggest that auxin signaling is important for resistance to the necrotro- phic fungi P. cucumerina and B. cinerea. SGTlb （one of two Arabidopsis SGT1 genes encoding HSP90/HSC70 co-chaperones） promotes the functions of SCF E3-ubiquitin ligase complexes in auxin and ja responses and resistance conditioned by certain Resistance （R） genes to biotrophic pathogens. We find that sgtlb mutants are as resistant to P. cucumerina as wild-type plants. Conversely, auxin/SCF signaling mutants are uncompromised in RPP4-triggered resistance to the obligate biotrophic oomycete, Hyaloperonospora parasitica. Thus, the predominant action of SGTlb in R gene-conditioned resis- tance to oomycetes appears to be at a site other than assisting SCF E3-ubiquitin ligases. However, genetic additivity of sgtlb axrl double mutants in susceptibility to H. parasitica suggests that SCF-mediated ubiquitination contributes to lim- iting biotrophic pathogen colonization once plant-pathogen compatibility is established.

Transcription factors(TFs)regulate gene expression by binding to cis-regulatory sequences in the promoters of target *** research is helping to decipher in part the cis-regulatory code in eukaryotes,including plants,but it is not yet fully understood how paralogous TFs select their *** we addressed this question by studying several proteins of the basic helix-loop-helix(bHLH)family of plant TFs,all of which recognize the same DNA *** focused on the mYC-related group of bHLHs,that redundantly regulate the jasmonate(ja)signaling pathway,and we observed a high correspondence between DNA-binding profiles in vitro and mYC function in *** demonstrated that A/T-rich modules flanking the mYC-binding motif,conserved from bryophytes to higher plants,are essential for TF recogni*** observed particular DNA-shape features associated with A/T modules,indicating that the DNA shape may contribute to mYC DNA *** extended this analysis to 20 additional bHLHs and observed correspondence between in vitro binding and protein function,but it could not be attributed to A/T modules as in *** conclude that different bHLHs may have their own codes for DNA binding and specific selection of targets that,at least in the case of mYCs,depend on the TF-DNA interplay.

In this study,a new algorithm of fractional beta chaotic maps is proposed to generate chaotic sequences for image *** proposed technique generates multi random sequences by shuffling the image pixel *** technique is used to blur the pixels connecting the input and encrypted images and to increase the attack *** proposed algorithm makes the encryption process sophisticated by using fractional chaotic maps,which hold the properties of *** fractional beta sequences are utilized to alter the image pixels to decryption *** experimental results proved that the proposed image encryption algorithm successfully encrypted and decrypted the images with the same *** output findings indicate that our proposed algorithm has good entropy and low correlation *** translates to enhanced security against different attacks.A mATLAB programming tool was used to implement and assess the image quality measures.A comparison with other image encryption techniques regarding the visual inspection and signal-to-noise ratio is provided.

Activation of Defense Response Pathways by OGs and Fig22 Elicitors in Arabidopsis Seedlings

We carried out transcriptional profiling analysis in 10-d-old Arabidopsis thaliana seedlings treated with oligogalacturonides （OGs）, oligosaccharides derived from the plant cell wall, or the bacterial flagellin peptide Fig22, general elicitors of the basal defense response in plants. Although detected by different receptors, both OGs and Fig22 trigger a fast and transient response that is both similar and comprehensive, and characterized by activation of early stages of multiple defense signaling pathways, particularly ja-associated processes. However, the response to Fig22 is stronger in both the number of genes differentially expressed and the amplitude of change. The magnitude of induction of individual genes is in both cases dose-dependent, but, even at very high concentrations, OGs do not induce a response that is as comprehensive as that seen with Fig22. While high doses of either microbe-associated molecular pattern （mAmP） elicit a late response that includes activation of senescence processes, SA-dependent secretory pathway genes and PR1 expres- sion are substantially induced only by Fig22. These results suggest a lower threshold for activation of early responses than for sustained or SA-mediated late defenses. Expression patterns of amino-cyclopropane-carboxylate synthase genes also implicate ethylene biosynthesis in regulation of the late innate immune response.
