Measuring NO Production by Plant Tissues and Suspension Cultured Cells

We describe an inexpensive and reliable detector for measuring NO emitted in the gas phase from plants. The method relies on the use of a strong oxidizer to convert NO to NO2 and subsequent capture of NO2 by a Griess reagent trap. The set-up approaches the sensitivity for NO comparable to that of instruments based on chemiluminescence and photoacoustic detectors. We demonstrate the utility of our set-up by measuring NO produced by a variety of well established plant sources. NO produced by nitrate reductase （NR） in tobacco leaves and barley aleurone was readily detected, as was the production of NO from nitrite by the incubation medium of barley aleurone. Arabidopsis mutants that overproduce NO or lack NO-synthase （AtNOS1） also displayed the expected NO synthesis phenotype when assayed by our set-up. We could also measure NO production from elicitor-treated suspension cultured cells using this set-up. Further, we have focused on the detection of NO by a widely used fluorescent probe 4-amino-5-methylamino-2＇,7＇-difluorofluorescein （DAF-FM）. Our work points to the pitfalls that must be avoided when using DAF-FM to detect the production of NO by plant tissues. In addition to the dramatic effects that pH can have on fluorescence from DAF-FM, the widely used NO scavengers 2-phenyl- 4,4,5, 5-tetramethylimidazoline- 1-oxyl-3-oxide （PTIO） oxide （cPTIO） can produce anomalous and unexpected to bind to dead cells and remain NO-sensitive. and 2-（4-carboxyphenyl）-4,4,5,5-tetramethylimidazoline- 1-oxyl-3- results. Perhaps the most serious drawback of DAF-FM is its ability to bind to dead cells and remain NO-sensitive.

Oxidative damages to DNA by indoor PM10s: Their relationships with trace element compositions

Plasmid DNA assay and ICP-MS analysis were conducted in order to investigate the bioreactivity of inhalable particles （PM_ 10） and the relationship between bioreactivity and trace element compositions of PM_ 10 in Beijing air. A total of four PM_ 10 samples were carefully selected to represent the indoor and corresponding outdoor environments: one from urban smoker's home, two from non-smoker's homes, and the other from the outdoor. In general, the oxidative damage by indoor PM_ 10 was slightly higher than that of outdoor. Among the four sets of samples, the PM_ 10 from the smoker's home had a lowest TD50 （toxic dose of PM_ 10 causing 50% DNA to be damaged）, being 100 μg·ml -1, suggesting the highest bioreactivity. The heavy metals are believed to be the main reason for oxidative damage to plasmid DNA. The ICP-MS analysis combined with the DNA assay showed that the water-soluble zinc levels had better relationship with TD50 values than other elements, implying that water-soluble zinc might play an important role in the damage of DNA. It is concluded that the PM_ 10 in smoker's home had the highest level of water-soluble zinc as well as the lowest TD50 （highest bioreactivity）. Iron is considered to be one of the most bioreactive elements, but it will cause little damage to plasmid DNA, probably because iron is mainly in water-insoluble state in Beijing PM_ 10.

Effect of Composts,lime and Diammonium Phosphate on the Phytoavailability of Heavy Metals in a Copper Mine Tailing Soil

Soil samples from a historic copper mine tailing site at the Parys Mountain,North Wales(UK) were amended with green waste compost(GC),GC+30% sewage sludge(GCS),lime and diammonium phosphate(DAP),to determine the effect of amendments on DTPA-and Ca(NO3)2-extractable metals in the mine tailing and on the phytoavailability of heavy metals by a lettuce(lactuca sativa l.).Both compost were added at the rate of 10% by weight,lime was added as calcium carbonate equivalent(pH = 7) and DAP at a 2300 mg kg-1 soil *** experiment was arranged in randomised complete design with three replicates in pots under control *** of lime resulted in the largest reduction in metal extractability with DTPA and Ca(NO3)2 and phytoavailability of Cu,Fe and Zn while DAP was effective in lowering Pb extractability and *** exception of Zn,all other metals extracted decreased with time after amendment *** distribution of heavy metals between and within the four procedures of potentially bioavailable sequential extraction(PBASE) varied significantly(P lationships were noted between the metals extracted with PBASE SE1 and Cu,Pb(P lts indicate that addition of lime is sufficient to restore the vegetative cover to a high metal mine waste while DAP is good for stabilizing Pb,but its detrimental role on plant growth and the risk associated with presence of N in DAP(through N leaching) may restrict its chances for remediation of contaminated sites.

Sensitivity analysis of key input parameters in conditional cell transmission model for oversaturated arterials

A novel conditional cell transmission model （CCTM） is a potential simulation tool because it accommodates all traffic conditions from light condition to oversaturated condition. To test the performance of the CCTM, a series of experiments for sensitivity analysis were designed and performed for a multilane, two-way, three-signal sample network. Experiment 1 shows that the model is performed in a logical and expected manner with variations in traffic demand with time and direction. Experiment 2 shows when the possibility of the occurrence of a useful gap increases to 60% and 100%, the delays in left rams decrease by 5% and 15%, respectively. In Experiment 3, comparing the possibility of a conditional cell of 0 with 100%, delay of left turn and delay of the entire network were underestimated by 58% and 11%, respectively. Hence, sensitivity analysis demonstrates that by reflecting local drivers＇ behaviors properly, the CCTM provides an accurate representation of traffic flow in simulating oversaturated traffic conditions.

Weight regain after intragastric balloon for pre-surgical weight loss

Pazopanib is the first and only tyrosine kinase inhibitor currently approved for the treatment of multiple histological subtypes of soft tissue sarcoma(STS).Initially developed as a small molecule inhibitor of vascular endothelial growth factor receptors,preclinical work indicates that pazopanib exerts an anticancer effect through the inhibition of both angiogenic and oncogenic signaling *** the establishment of optimal dosing and safety profiles in early phase studies and approval for the treatment of advanced renal cell carcinoma,pazopanib was investigated in STS.A landmark phase III randomized study demonstrated improved progression-free survival with pazopanib compared to that with placebo in pretreated patients with STS of various *** efficacy of pazopanib in specific STS subtypes has been further described in real-world-based case series in both mixed and subtype-specific STS *** present,there are no clinically validated predictive biomarkers for use in selecting patients with advanced STS for pazopanib therapy,limiting the clinical effectiveness and cost-effectiveness of the *** this review,we summarize the preclinical and clinical data for pazopanib,outline the evidence base for its effect in STS and explore reported studies that have investigated putative biomarkers.

DETERMINE ON INITIAl DIMENSIONAl STABIlITY OF CHEMICAl MODIFIED WOOD

Strain development along radial direction was continuously determined by D-200 linear Variable Differential Transformers (l.V.D.T). When a wood specimen was put into water and begin to swell from absolute dry moisture content to the maximum. Experiment results show that the differences of swelling with soaking time between untreated and treated wood specimens due to uptaking water. It would also give an indication to the effectiveness of various chemicals on the dimensional stability of modified wood. ASE of modified wood samples with SA-EP, EP and SA were found to be better than MA-AGE. SA-EP had got the best effetivencss on dimensional stability in reducing the swelling.

Response of nitrogen fractions in the rhizosphere and bulk soil to organic mulching in an urban forest plantation

Nitrogen is an essential component in forest ecosystem nutrient *** fractions,such as dissolved nitrogen,ammonium,nitrate,and microbial biomass nitrogen,are sensitive indicators of soil nitrogen pools which affect soil fertility and nutrient ***,the responses of nitrogen fractions in forest soils to organic mulching are less well *** rhizosphere is an important micro-region that must be considered to better understand element cycling between plants and the soil.A field investigation was carried out on the effect of mulching soil in a 15-year-old ligustrum lucidum urban *** in total nitrogen and nitrogen fractions in rhizosphere and bulk soil in the topsoil(upper 20 cm)and in the subsoil(20-40 cm)were evaluated following different levels of mulching,in addition to nitrogen contents in fine roots,leaves,and organic *** relationships between nitrogen fractions and other measured variables were *** mulching had no significant effect on most nitrogen fractions except for the rhizosphere microbial biomass nitrogen(MBN),and the thinnest(5 cm)mulching layer showed greater effects than other *** MBN was more sensitive to mulching compared to bulk soil,and was more affected by soil environmental *** and soil depth had more pronounced effects on nitrogen fractions than *** nitrogen and dissolved nitrogen were correlated to soil phosphorus,whereas other nitrogen fractions were strongly affected by soil physical properties(temperature,water content,bulk density).Mulching also decreased leaf nitrogen content,which was more related to soil nitrogen fractions(except for MBN)than nitrogen contents in either fine roots or organic *** applications of small quantities of organic mulch contribute to nitrogen transformation and utilization in urban forests.
