In Brassica napus L.(rapeseed),complete genic male sterility(GMS)plays an important role in the utilization of *** microRNAs(miRNAs)play essential regulatory roles during bud development,knowledge of how GMS is regulated by miRNAs in rapeseed is rather *** this study,we obtained a novel recessive GMS system,*** sterile line CN12A has defects in tapetal diferentiation and *** sequencing was employed to examine the expression of miRNAs in the buds of CN12A and the fertile line *** identifed 85 known miRNAs and 120 novel miRNAs that were expressed during rapeseed anther *** comparing the expression levels of miRNAs between CN12A and CN12B,19 and 18 known miRNAs were found to be difrentially expressed in 0.5--1.0 mm buds and in 2.5--3.0 mm buds,*** these,the expression levels of 14 miRNAs were higher and the levels of 23 miRNAs were lower in CN12A compared with *** predicted target genes of these iferentially expressed miRNAS encode protein kinases,F-box domain-containing proteins,MADS-box family proteins,SBP-box gene family members,HD ZIP proteins,floral homeotic protein APETAL A 2(AP2)。and nuclear factor Y,subunit *** targets have previously been reported to be involved in pollen development and male sterility,suggesting that miRNAs might act as regulators of GMS in rapeseed ***,RT-qPCR data suggest that one of the dffrentially expressed miRNAs,bna-miR159,plays a role in tapetal differentiation by regulating the expression of transcription factor BnMYB101 and participates in tapetal degradation and infuences callose degradation by manipulating the expression of *** findings contibute to our understanding of the roles of miRNAs during anther development and the occurrence of GMS in rapeseed.

AIM:To evaluate the proinflammatory effects and molecular mechanisms of interleukin(IL)-17 in intestinal epithelial cell line ***:HT-29 cells were cultured with IL-17,tumor necrosis factor(TNF)-α,or the combination of both IL-17 and TNF-α.Real-time PCR and Western blot were used to measure the gene expression levels of neutrophil chemokines CXCL1,CXCL2,CXCL5,CXCL6,IL-8and TH-17 cell chemokine CCL20,the phosphorylation levels of p38 and TNF-α,and the expression level of IL-8,after using the p38 inhibitor in HT-29 *** stable Act1 knockdown HT-29 cell line was established to further test the phosphorylation changes of p38,after using IL-17 and TNF-α.RESULTS:After HT-29 cells were cultured with IL-17and TNF-α,the expression levels of neutrophil chemokines(CXCL1,CXCL2,CXCL5,CXCL6,IL-8)and Th17chemokine(CCL20)significantly improved(24.96±2.53,28.47±2.87,38.08±2.72,33.47±2.41,31.7±2.38,44.37±2.73,respectively),and the differences were all statistically significant(P<0.01).Western blot results showed that IL-17 obviously enhanced the phosphorylation level of p38,which was induced by TNF-α.Compared with the control group,the expression level of IL-8 significantly declined(9.47±1.36 vs 3.06±0.67,P<0.01)when TH-29 cells were cultured with IL-17and TNF-α.p38 inhibition assay showed that the p38pathway played an essential role in the inflammatory response induced by IL-17.p38 phosphorylation levels could not be changed after using IL-17 and TNF-αin the stable Act1 knockdown HT-29 cell ***:IL-17 significantly promoted the gene expression levels of TNF-α-induced neutrophil chemokines and Th17 cell *** is obvious that IL-17and TNF-αhave synergistic effects on p38.

An association between assisted reproductive technology （ART） and neurobehavioral imprinting disorders has been reported in many studies, and it seems that ART may interfere with imprint reprogramming. However, it has never been explored whether epigenetic erros or imprinting disease susceptibility induced by ART can be inherited transgenerationally. Hence, the aim of this study was to determine the effect of in vitro fertilization and embryo transfer （IVF-ET） on transgenerational inheritance in am inbred mouse model. Mice derived from IVF-ET were outcrossed to wild-type C57BL/6J to obtain their female and male line F2 and F3 generations. Their behavior, morphology, histology, and DNA methylation status at several important differentially methylated regions （DMRs） were analyzed by Morris water maze, hematoxylin and eosin （H＆E） staining, and bisulfite genomic sequencing. No significant differences in spatial learning or phenotypic abnormality were found in adults derived from IVF （F1） and female and male line F2 and F3 generations. A borderline trend of hypomethylation was found in H19 DMR CpG island 3 in the female line-derived F3 generation （0.40±0.118, P=0.086）. Methylation status in H19/Igf2 DMR island 1, Igf2 DMR, KvDMR, and Snrpn DMR displayed normal patterns. Methylation percentage did not differ significantly from that of adults conceived naturally, and the expression of the genes they regulated was not disturbed. Transgenerational integrity, such as behavior, morphology, histology, and DNA methylation status, was maintained in these generations, which indicates that exposure of female germ cells to hormonel stimulation and gamete manipulation might not affect the individuals and their descendents.

Objective： To analyze the effect of Chinese medicine （CM） on mortality and quality of life （QOL） of acquired immunodeficiency syndrome （AIDS） patients treated with combined antiretroviral therapy （cART）. Methods： A random sample of AIDS patients enrolled in the National Chinese Medicine Treatment Trial Program （NCMTP） that met the inclusion criteria was included in this study. NCMTP patients were included as the CM＋cART group, and those not in the NCMTP were included as the cART group. Survival from September 2004 to September 2012 was analyzed by retrospective cohort study. QOL was analyzed by cross-sectional study. Results： The retrospective cohort study included 528 AIDS patients, 322 in the CM＋cART group and 206 in the cART group. After 8 years, the mortality in the CM＋cART group was 3.3/100 person-years, which was lower than the cART group of 5.3/100 person-years （P〈0.05）. The hazard ratio （HR） for mortality in the cART group was 1.6 times that of the CM＋cART group by Cox proportional hazard model analysis. After controlling for gender, age, marital status, education, and CD4＋ T-cell count, the HR was 1.9 times higher in the cART group compared with the CM＋cART group （P〈0.05）. The cross-sectional study investigated 275 AIDS patients. The mean scores of all QOL domains except spirituality/personal beliefs were higher in the CM＋cART group than in the cART group （P〈0.05）. Conclusions： For AIDS patients, CM could help to prolong life, decrease mortality, and improve QOL. However, there were limitations in the study, so prospective studies should be carded out to confirm our primary results.

BACKGROUND: The study aims to investigate the performance of a metagenomic next-generationsequencing (NGS)-based diagnostic technique for the identifi cation of potential bacterial and viral infectionsand eff ects of concomitant viral infection on the survival rate of intensive care unit (ICU) sepsis ***: A total of 74 ICU patients with sepsis who were admitted to our institution from February1, 2018 to June 30, 2019 were enrolled. Separate blood samples were collected from patients for bloodcultures and metagenomic NGS when the patients’ body temperature was higher than 38 °C. Patients’demographic data, including gender, age, ICU duration, ICU scores, and laboratory results, were *** correlations between pathogen types and sepsis severity and survival rate were ***: NGS produced higher positive results (105 of 118;88.98%) than blood cultures(18 of 118;15.25%) over the whole study period. Concomitant viral infection correlated closelywith sepsis severity and had the negative effect on the survival of patients with sepsis. However,correlation analysis indicated that the bacterial variety did not correlate with the severity of ***: Concurrent viral load correlates closely with the severity of sepsis and thesurvival rate of the ICU sepsis patients. This suggests that prophylactic administration of antiviraldrugs combined with antibiotics may be benefi cial to ICU sepsis patients.
