The epigenetic regulation of gene expression is critical for ensuring the proper deployment and stability of defined genome transcription programs at specific developmental stages. The cellular memory of stable gene expression states during animal and plant development is mediated by the opposing activities of Polycomb group （PcG） factors and trithorax group （trxG） factors. Yet, despite their importance, only a few trxG factors have been characterized in plants and their rotes in regulating plant development are poorly defined. In this work, we report that the closely related Arabidopsis trxG genes ULTRAPETALA1 （ULT1） and ULT2 have overlapping functions in regulating shoot and floral stem cell accumula- tion, with ULT1 playing a major role but ULT2 also making a minor contribution. The two genes also have a novel, redun- dant activity in establishing the apical-basal polarity axis of the gynoecium, indicating that they function in differentiating tissues. Like ULT1 proteins, ULT2 proteins have a dual nuclear and cytoplasmic localization, and the two proteins physically associate in planta. Finally, we demonstrate that ULT1 and ULT2 have very similar overexpression phenotypes and regulate a common set of key development target genes, including floral MADS-box genes and class I KNOX genes. Our results reveal that chromatin remodeling mediated by the ULT1 and ULT2 proteins is necessary to control the development of mer- istems and reproductive organs. They also suggest that, like their animal counterparts, plant trxG proteins may function in multi-protein complexes to up-regulate the expression of key stage- and tissue-specific developmental regulatory genes.

Repeated video head impulse testing in patients is a stable measure of the passive vestibulo-ocular reflex

Objectives The video head impulse test(vHIT)is used as a measure of compensation yet it’s stability in patients with vestibular pathology is *** 144 patients(n=72 female,mean 54.46±15.8 years)were grouped into one of three primary diagnoses(Peripheral,central,or Mixed).Subjects were further categorized based on sex(male versus female),ear(left versus right;ipsilesional versus contralesional),age(six groups ranging from 19 to 84 years),and duration between visits(five groups,mean 191.46±SE 29.42 days,median 55.5 days).The gain of the VOR during passive head rotation was measured for each semicircular canal(horizontal,anterior,posterior).Results There was no difference in the VOR gain within any semicircular canal between the two visits(horizontal:p=0.179;anterior:p=0.628;posterior:p=0.613).However,the VOR gain from the horizontal canals was higher than the vertical canals for each visit(pcantly lower(p≤0.001)horizontal semicircular canal gains at each *** was no difference in VOR gain between sex(p=0.215)or age groupings(p=0.331).Test-retest reliability of vHIT in patient subjects is good(Icc=0.801)and the VOR gain values across two separate visits were significant and positively correlated(r=0.67)regardless of sex,ear,age,or duration between *** The vHIT is a stable measure of VOR gain over two different times across a variety of vestibular patients with no influence of age or sex.

Epigenetic alterations are widespread in cancer and can complement genetic alterations to influence cancer progression and treatment *** determine the potential contribution of DNA methylation alterations to tumor phenotype in non-small cell lung cancer(NScLc)in both smoker and never-smoker patients,we performed genome-wide profiling of DNA methylation in 17 primary NScLc tumors and 10 matched normal lung samples using the complementary assays,methylated DNA immunoprecipitation sequencing(MeDIP-seq)and methylation sensitive restric-tion enzyme sequencing(MRE-seq).We reported recurrent methylation changes in the promoters of several genes,many previously implicated in cancer,including FAM83A and SEPT9(hy-pomethylation),as well as PcDH7,NKX2-1,and SOX17(hypermethylation).Although many methylation changes between tumors and their paired normal samples were shared across patients,several were specific to a particular smoking *** example,never-smokers displayed a greater proportion of hypomethylated differentially methylated regions(hypoDMRs)and a greater number of recurrently hypomethylated promoters,including those of ASPScRI,TOP2A,DPP9,and USP39,all previously linked to *** outside of promoters were also widespread and often recurrent,particularly methylation loss over repetitive elements,highly enriched for ERV1 *** hypoDMRs were enriched for several transcription factor binding motifs,often for genes involved in signaling and cell *** example,71%of recurrent promoter hypoDMRs contained a motif for ***,the majority of DMRs were located within an active chromatin state in tissues profiled using the Roadmap Epigenomics data,suggesting that methylation changes may contribute to altered regulatory programs through the adaptation of cell type-specific expression programs.

Visual rating and the use of image analysis for assessing canopy density in a pecan provenance collection during leaf fall

The roles of inter- and intra-sexual selection in behavioral isolation between native and invasive pupfishes

Male-male competition and female mate choice may both play important roles in driving and maintaining reproductive isolation between species. When previously allopatric species come into secondary contact with each other due to introductions, they provide an opportunity to evaluate the identity and strength of reproductive isolating mechanisms. If reproductive isolation is not maintained, hybridization may occur. We examined how reproductive isolating mechanisms medi- ate hybridization between endemic populations of the Red River pupfish cyprinodon rubrofluviati- lis and the recently introduced sheepshead minnow c. variegatus. In lab-based dominance trials, males of both species won the same number of competitions. However, male c. rubrofluviatilis that won competitions were more aggressive than c, variegatus winners, and more aggression was needed to win against competitor c. variagatus than allopatric c. rubrofluviatilis. Duration of fights also differed based on the relatedness of the competitor. In dichotomous mate choice trials, there were no conspecific or heterospecific preferences expressed by females of either species. Our findings that male-male aggression differs between closely and distantly related groups, but female choice does not suggest that male-male competition may be the more likely mechanism to impede gene flow in this system.

Background： Hamburger is the most consumed beef product in North America, but lacks in nutritional appeal due to its high fat content and high proportion of saturated fatty acids （SFA）. Objectives of the present study were to improve the FA profiles of hamburgers made with perirenal fat （PRF） and subcutaneous fat （ScF） when feeding steers different diets along with examining differences in sensory attributes and oxidative stability. Diets included a control diet containing 70：30 red clover silage： barley based concentrate, a diet containing sunflower-seed （SS） substituted for barley, and diets containing SS with 15% wheat dried distillers＇ grain with solubles （DDGS-15） or 30% DDGS （DDGS-30）. Hamburgers were made from triceps brachii and either PRF or ScF （80：20 w/w）. Results： Perirenal fat versus ScF hamburgers FA had 14.3% more （P 〈0.05） 18：0, 11.8% less cis （c）9-18：1 （P 〈0.05）, and 1.82% more total trdns （t）-18：1 mainly in the form of tl 1-18：1. During sensory evaluation, PRF versus ScF hamburgers had greater （P 〈0.05） mouth coating, but the difference was less than one panel unit. Examining effects of steer diet within PRF hamburgers, feeding the SS compared to the control diet increased （P 〈0.05） t-18：1 by 2.89% mainly in the form of tl 1-18：1, feeding DGGS-15 diet led to no further changes （P 〉0.05）, but feeding DDGS-30 diet reduced the proportions of （P 〈0.05） of t-18：1 chiefly tl 1-18：1. Feeding SS and DDGS diets had small but significant （P 〈0.05） effects on hamburger sensory attributes and oxidative stability. conclusions： Feeding high-forage diets including SS and 15% DDGS, and taking advantage of the FA heterogeneity between fat depots offers an opportunity to differentially enhance beef hamburgers with 18：2n-6 biohydrogenation products （Le., t11-18：1） with potential human health benefits without compromising their sensory attributes and oxidative stability during retail display.

Effect of opioid dependence or abuse on opioid utilization after shoulder arthroplasty

AIM To examine whether opioid dependence or abuse has an effect on opioid utilization after anatomic or reverse total shoulder arthroplasty(TSA).METHODS All anatomic TSA(IcD-9 81.80) and reverse shoulder arthroplasty(RSA)(IcD-9 81.88) procedures from 2007 to 2015 were queried from within the Humana claims database utilizing the Pearl Diver supercomputer(colorado Springs, cO). Study groups were formed based on the presence or absence of a previous history of opioid dependence(IcD-9 304.00 and 304.03) or abuse(IcD-9 305.50 and 305.53). Opioid utilization among the groups was tracked monthly up to 1 year post-operatively utilizing National Drug codes. A secondary analysis was performed to determine risk factors for pre-operative opioid dependence or *** Two percent of TSA(157 out of 7838) and 3% of RSA(206 out of 6920) patients had a history of opioid dependence or abuse. For both TSA and RSA, opioid utilization was significantly higher in opioid dependent patients at all post-operative intervals(P cidence of opioid use among groups was similar within the first post-operative month. After TSA, opioid dependent patients were over twice as likely to fill opioid prescriptions during the post-operative months 1-12. Following RSA, opioid dependent patients were over 3 times as likely to utilize opioids from months 3-12. Neuronal injury in the motor cortex after chronic stroke and lower limb motor impairment: a voxelbased lesion symptom mapping study

Many studies have examined motor impairments using voxel-based lesion symptom mapping, but few are reported regarding the corresponding relationship between cerebral cortex injury and lower limb motor impairment analyzed using this technique. This study correlated neuro- nal injury in the cerebral cortex of 16 patients with chronic stroke based on a voxel-based lesion symptom mapping analysis. Neuronal injury in the corona radiata, caudate nucleus and putamen of patients with chronic stroke could predict walking speed. The behavioral measure scores were consistent with motor deficits expected after damage to the cortical motor system due to stroke. These findings suggest that voxel-based lesion symptom mapping may provide a more accurate prognosis of motor recovery from chronic stroke according to neuronal injury in cerebral motor cortex.
