Bone marrow-derived mesenchymal stem cells versus adipose-derived mesenchymal stem cells for peripheral nerve regeneration

Studies have confirmed that bone marrow-derived mesenchymal stem cells （MSCs） can be used for treatment of several nervous system diseases. However, isolation of bone marrow-derived MSCs （BMSCs） is an invasive and painful process and the yield is very low. Therefore, there is a need to search for other alterative stem cell sources. Adipose-derived MSCs （ADSCs） have phenotypic and gene expression profiles similar to those of BMSCs. The production of ADSCs is greater than that of BMSCs, and ADSCs proliferate faster than BMSCs. To compare the effects of venous grafts containing BMSCs or ADSCs on sciatic nerve injury, in this study, rats were randomly divided into four groups： sham （only sciatic nerve exposed）, Matrigel （MG; sciatic nerve injury ＋ intravenous transplantation of MG vehicle）, ADSCs （sciatic nerve injury ＋ intravenous MG containing ADSCs）, and BMSCs （sciatic nerve injury ＋ intravenous MG containing BMSCs） groups. Sciatic functional index was calculated to evaluate the function of injured sciatic nerve. Morphologic characteristics of nerves distal to the lesion were observed by toluidine blue staining. Spinal motor neurons labeled with Fluoro-Gold were quantitatively assessed. Compared with sham-operated rats, sciatic functional index was lower, the density of small-diameter fibers was significantly increased, and the number of motor neurons significantly decreased in rats with sciatic nerve injury. Neither ADSCs nor BMSCs significantly improved the sciatic nerve function of rats with sciatic nerve injury,increased fiber density, fiber diameters, axonal diameters, myelin sheath thickness, and G ratios （axonal diameter/fiber diameter ratios） in the sciatic nerve distal to the lesion site. There was no significant difference in the number of spinal motor neurons among ADSCs, BMSCs and MG groups. These results suggest that neither BMSCs nor ADSCs provide satisfactory results for peripheral nerve repair when using MG as the conductor for engraftment.

Huge Recalcitrant Warts Treated with Pulse Dye Laser after Conventional Therapies and Others Destructive Physical Modalities Failed

Cutaneous infections with the human papilloma virus, despite being almost always asymptomatic, can induce slow-growing lesions knowed as warts that can remain subclinical for long periods of time. There are several therapeutical options namely keratolytics, retinoids, imiquimod, cryotherapy, electrosurgery, lasers and classic surgery, but none has antiviral effects. We describe the case of a patient with large warts located on the dorsal surface of the right hand, treated with cryotherapy, CO2 laser and topical imiquimod having either no clinical response or quick relapse. Treatment with PDL was started and after 13 sessions, the patient was without lesions, with a good cosmetic result and so far without relapse. This case report is relevant to us and deserves to be published because of the size of the lesions and the great response to PDL despite all the failures with the other treatments.

Avalonia,get bent!–Paleomagnetism from SW Iberia confirms the Greater Cantabrian Orocline

The amalgamation of Pangea formed the contorted Variscan-Alleghanian orogen,suturing Gondwana and Laurussia during the Carboniferous.From all swirls of this orogen,a double curve in Iberia stands out,the coupled Cantabrian Orocline and Central Iberian curve.The Cantabrian Orocline formed at ca.315–290 Ma subsequent to the Variscan orogeny.The formation mechanism of the Cantabrian Orocline is disputed,the most commonly proposed mechanisms include either(1)that south-westernmost Iberia would be an Avalonian(Laurussian)indenter or(2)that the stress field changed,buckling the orogen.In contrast,the geometry and kinematics of the Central Iberian curve are largely unknown.Whereas some authors defend both curvatures are genetically linked,others support they are distinct and formed at different times.Such uncertainty adds an extra layer of complexity to our understanding of the final stages of Pangea’s amalgamation.To solve these issues,we study the late Carboniferous–early Permian vertical-axis rotations of SW Iberia with paleomagnetism.Our results show up to 70counterclockwise vertical-axis rotations during late Carboniferous times,concurring with the anticipated kinematics if SW Iberia was part of the southern limb of the Cantabrian Orocline.Our results do not allow the necessary penecontemporaneous clockwise rotations in Central Iberia to support a concomitant formation of both Cantabrian and Central Iberian curvature.The coherent rotation of both Gondwanan and Avalonian pieces of SW Iberia discards the Laurussian indenter hypothesis as a formation mechanism of the Cantabrian Orocline and confirms the Greater Cantabrian Orocline hypothesis.The Greater Cantabrian Orocline likely formed as a consequence of a change in the stress field during the late Carboniferous and extended beyond the Rheic Ocean suture affecting the margins of both Laurussia and Gondwana.

Background:Topical photodynamic therapy(PDT)with 5-aminolevulinic acid(ALA)was first described by Kennedy et al.in 1990;currently,accumulated evidence shows that topical PDT is effective in the treatment of actinic keratoses,Bowen’s disease and superficial basal cell carcinoma,being potentially advantageous where size,anatomical location or number of lesions limit the efficacy and/or the acceptability of conventional therapies.It involves the use of photosensitizing drugs and the administration of an appropriate light source to activate the sensitizing agent,which leads to the production of activated oxygen species for tissue destruction.The topically administered prodrug 5-ALA is converted within cells into the active photosensitizer protoporphyrin IX,via haem cycle,with preferential accumulation in tumour cells.Methods:The authors describe their experience with topical PDT,using 20%5-ALA and two different incoherent light sources,’Waldman 1200 L’and ’VersaLightTM’,in the treatment of non-melanoma skin cancer and precursor lesions.More than 100 patients were treated since 1997,with up to a 7-year follow-up period.Results:Cases are illustrated and results are presented and discussed in the light of the relevant Literature.Conclusion:The study suggests that 5-ALA PDT is effective in the treatment of superficial skin cancer and premalignant lesions.

Analysis of a Plasmonic Based Optical Fiber Optrode With Phase Interrogation

Optical fiber optrodes are attractive sensing devices due to their ability to perform point measurement in remote locations. Mostly, they are oriented to biochemical sensing, quite often supported by fluorescent and spectroscopic techniques, but with the refractometric approach considered as well when the objective is of high measurement performance, particularly when the focus is on enhancing the measurand resolution. In this work, we address this subject, proposing and analyzing the characteristics of a fiber optic optrode relying on plasmonic interaction. A linearly tapered optical fiber tip is covered by a double overlay： the inner one - a silver thin film and over it - a dielectric layer, with this combination allowing to achieve, at a specific wavelength range, surface plasmonic resonance （SPR） interaction sensitive Typically, the interrogation of the SPR sensing to the refractive index of the surrounding medium. structures is performed, considering spectroscopic techniques, but in principle, a far better performance can be obtained, considering the reading of the phase of the light at a specific wavelength located within the spectral plasmonic resonance. This is the approach which is studied here in the context of the proposed optical fiber optrode configuration. The analysis performed shows the combination of a silver inner layer with a dielectric titanium oxide layer with tuned thicknesses enables sensitive phase reading and allows the operation of the fiber optic optrode sensor in the third telecommunication wavelength window.

A voltammetric sensor based on the electropolymerization of cobalt-poly(methionine)(Co-poly(Met)) on a glassy carbon electrode (GCE) was developed and applied for the determination of estriol by differential pulse voltammetry (DPV) for the first time. The electrochemical properties of the Co-poly(Met)/GCE were analysed by cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS). Scanning electron microscopy (SEM) and energy dispersive spectroscopy (EDS) were used to characterize the polymers on the GCE surface. The deposition of the Co-poly(Met) film on the GCE surface enhanced the sensor electronic transfer. CV studies revealed that estriol exhibits an irreversible oxidation peak at t0.58 V for the Co-poly(Met)/GCE (vs. Ag/AgCl reference electrode) in 0.10 mol/L Britton-Robinson buffer solution (pH=7.0). Different voltammetric scan rates (10-200 mV/s) suggested that the estriol oxidation on the Co-poly(Met)/GCE surface is controlled by adsorption and diffusion processes. Based on the optimized DPV conditions, the linear responses for estriol quantification were from 0.596 μmol/L to 4.76 μmol/L (R2 =0.996) and from 5.66 μmol/L to 9.90 μmol/L (R2 =0.994) with a limit of detection (LOD) of 0.0340 μmol/L and a limit of quantification (LOQ) of 0.113 μmol/L. The DPV-Co-poly(Met)/GCE method provided good intra-day and inter-day repeatability with RSD values lower than 5%. Also, no interference of real sample matrices was observed on the estriol voltammetric response, making the DPV-Copoly( Met)/GCE highly selective for estriol. The accuracy test showed that the estriol recovery was in the ranges 96.7%-103% and 98.7%-102% for pharmaceutical tablets and human urine, respectively. The estriol quantification in pharmaceutical tablets performed by the Co-poly(Met)/GCE-assisted DPV method was comparable to the official analytical protocols.

Experimental Study on Split-Ring Resonators with Different Slit Widths

Metamaterial one-dimensional periodic structures are composed of split-ring resonators, which can display electric permittivity and magnetic permeability simultaneously negative, are studied experimentally. In the present study, each resonator is made up of two concentric circular copper rings patterned on a substrate of kapton, with slits diametrically opposite each other and with the line of the splits along the longitudinal direction of the periodic array containing seven split rings evenly spaced. The experiments consist in inserting the metamaterial slab into a square waveguide of side length 6 mm, corresponding to a cutoff frequency of 25 GHz. Transmission bands due to magnetic and electrical responses are identified for slits with aperture widths of 1 mm and 2 mm, centered at 5.67 and 6.12 GHz frequencies, respectively, values well below the 25 GHz frequency cutoff, so characterizing a medium with negative permeability and permittivity.

An Experimental Study of Self-Compacting Concrete Made with Filler from Construction and Demolition Waste

This study evaluated the influence of the Portland cement replacement by 0, 5%, 10%, 15% and 20% of Construction and Demolition Waste (CDW) filler contents in the production of self-compacting concrete (SCC). The SCC mixtures were evaluated on fresh state by slump flow, J-ring, resistance of segregation, specific gravity, and on hardened state by compressive and splitting tensile strength, specific gravity, air voids and absorption rate. The results indicated that all SCC produced with CDW filler met the limits established at any level of substitution without changes of the w/c ratio or superplasticizer content. It was possible to verify that the presence of CDW filler, in substitution of cement, by volume, improves the resistance to segregation and up to 5% of CDW filler decreases the loss of fluidity with time as compared to reference. It was found that all SCC mixtures, at 28 days, had the average compressive strength above 50 MPa, without showing significant loss with up to 20% of CDW filler. Cryptococcal Meningitis in Patient with Chronic Myeloid Leukemia

Objective: This study aimed to report the case of a female patient with chronic myeloid leukemia affected by cryptococcal meningitis. Case report: ML, white, 48 years old, female sex, previously diagnosed with chronic myeloid leukemia that has been refractive to the use of imatinib and who has recently begun using nilotinib, was admitted complaining of sudden and disabling migraine in the last 1 month associated with asthenia, adinamia, anorexia, disinterest for daily activities, dizziness, nausea, and vomiting. She evolved with ataxia, and started to stroll with help and showed decrease of muscular strength in her upper limbs. She also presented episodes of decrease of consciousness, with look fixation, no respond to sound stimulation, and short-term hearing loss. The cerebrospinal fluid showed presence of Cryptococcus sp. and, therefore, we began treatment with intravenous liposomal amphotericin B in the dose of 3 mg/kg/day, for 6 weeks. A new cerebrospinal fluid analysis, at the end of treatment, also showed rare structures that are compatible with Cryptococcus sp. As sequelae, she continued with hearing loss in her right ear and enhancement in her right auditory canal, seen in the magnetic resonance imaging. After stabilization and clinical improvement, she was discharged. After 3 weeks, she was hospitalized again with degeneration of the condition, and died due to intracranial hypertension secondary to cryptococcal infection. Final Considerations: This report reinforces the need of reflecting on fungi pathologies, especially in immunosuppressant patients, as well as the importance of early diagnosing and making a fast intervention, with the aims of providing quality of life and comfort to the patient and of minimizing neurological sequelae to the patient.

Objective: To evaluate the synergic effects of a novel oral supplement formulation, containing prebiotics,yeast b-glucans, minerals and silymarin(Silybum marianum), on lipid and glycidic metabolism, inflammatory and mitochondrial proteins of the liver, in control and high-fat diet-induced obese mice.Methods: After an acclimation period, 32 male C57 BL/6 mice were divided into the following groups:nonfat diet(NFD) vehicle, NFD supplemented, high-fat diet(HFD) vehicle and HFD supplemented. The vehicle and experimental formulation were administered orally by gavage once a day during the last four weeks of the diet(28 consecutive days). We then evaluated energy homeostasis, inflammation, and mitochondrial protein expression in these groups of mice.Results: After four weeks of supplementation, study groups experienced reduced glycemia, dyslipidemia,fat, and hepatic fibrosis levels. Additionally, proliferator-activated receptor-α, AMP-activated protein kinase-1α, peroxisome proliferator-activated receptor γ co-activator-1α, and mitochondrial transcription factor A expression levels were augmented;however, levels of inhibitor of nuclear factor-κB kinase subunit a and p65 nuclear factor-κB expression, and oxidative markers were reduced. Notably, the cortisol/C-reactive protein ratio, a well-characterized marker of the hypothalamic–pituitary–adrenal axis immune interface status, was found to be modulated by the supplement.Conclusion: We discovered that the novel supplement was able to modify different antioxidant, metabolic and inflammatory pathways, improving the energy homeostasis and inflammatory status, and consequently alleviated hepatic steatosis.
