The precise regulation of microRNA(miRNA)biogenesis is crucial for plant development,which requires core microprocessors and many fine tuners to coordinate their miRNA processing activity/specificity in fluctuating cellular *** de-etiolation,light triggers a dramatic accumulation of core microprocessors and primary miRNAs(pri-miRNAs)but decreases pri-miRNA processing activity,resulting in relatively constant miRNA *** mechanisms underlying these seemingly contradictory regulatory changes remain *** this study,we identified fork.ead-associated domain 2(FHA2)as a light-stabilized suppressor of miRNA *** found that FHA2 deficiency increased the level of mature miRNAs,accompanied by a reduction in pri-miRNAs and target *** assays showed that FHA2 associates with the core microprocessors DCL1,HYL1,and SE,forming a complex to suppress their pri-miRNA processing *** analyses revealed that FHA2 promotes HYL1 binding but inhibits the binding of DCL1-PAZ-RNase-RNA-binding domains(DCL1-PRR)to miRNAs,whereas FHA2 does not directly bind to these ***,we found that FHA2 protein is unstable in the dark.but stabilized by light during ***,disruption of FH A led to defects in light-triggered changes in miRNA expression and reduced the survival rate of deetiolated seedlings after prolonged light ***,these data suggest that FHA2 is a novel light-stabilized suppressor of miRNA biogenesis and plays a role in fine-tuning miRNA processing during de-etiolation.

An open-label study of alefacept plus ultravioletB light as combination therapy for chronic plaque psoriasisOrtonne j.P./k.emis A./k.o j./choi j. [***-onne, l’ Archet II, 15, R. de S. Antoine de Ginestiere, Nice, Cé dex 03, France]Back.round: Alefacept, a fully human LFA-3/IgG1 fusion protein, is a selective biological agent approved in theUnited States for the treatment of chronic plaque psoriasis. In phase 3 trials, clinical improvement and prolonged off-treatment remission of psoriasis correlated with reductions in circulating memory T cells. Reductions in pathogenic epidermal T cells in psoriatic lesions also have been noted following phototherapy with ultraviolet B (UVB) light. Because alefacept and UVB target T cells in different ways, combination therapy with these two agents may lead to greater efficacy. Obj.ctives: To determine the safety, tolerability, and efficacy trends of combination therapy with alefacept plus UVB light in patients with chronic plaque psoriasis. Methods: In an open-label, parallel-group study conducted at two sites, one in France and one in the United States, patients with chronic plaque psoriasis who were candidates for phototherapy received 12-week.y intramuscular inj.ctions of alefacept, 15 mg. In addition, patients were randomized to one of three treatment arms: no UVB treatment, 6-week.UVB treatment, and 12-week.UVB treatment. UVB treatment consisted of narrowband (NB) UVB at the site in France and broadband (BB) UVB at the site in the United States. The 12-week.treatment period was followed by a 12-week.follow-up period. Clinic visits occurred week.y during treatment and every 2-4 week. during follow-up. Results: A total of 60 patients (n =30/site)were enrolled in the study. Alefacept was well tolerated when administered in combination with UVB treatment and as monotherapy. There was no evidence of increased phototoxicity or photosensitivity with the combination. Diamond-lik. carbon（DLC） films were prepared on silicon substrates by the radio frequency（RF） plasma enhanced chemical vapor deposition（PECVD） method using methane（CH4） and hydrogen（H2） *** examined the effects of the post annealing temperature on the tribological properties of the DLC films using friction force microscopy（FFM）.The films were annealed at various temperatures ranging from 300 to 900 oC in steps of 200 oC using rapid thermal annealing（RTA） equipment in nitrogen *** variation of structure according to the annealing treatment was examined using X-ray photoelectron spectroscopy（XPS）.The wettability of the films was examined using contact angle *** friction coefficients of annealed DLC films were investigated by atomic force microscopy（AFM） in FFM mode.

BACk.ROUND Intraductal papillary mucinous neoplasms(IPMNs)are non-invasive pancreatic precursor lesions that can potentially develop into invasive pancreatic ductal ***,the International Consensus Guidelines(ICG)for IPMNs provides the basis for evaluating suspected IPMNs on computed tomography(CT)*** using the ICG,it remains challenging to accurately predict whether IPMNs harbor high grade or invasive disease which would warrant surgical resection.A supplementary quantitative radiological tool,radiomics,may improve diagnostic accuracy of radiological evaluation of *** hypothesized that using CT whole lesion radiomics features in conj.nction with the ICG could improve the diagnostic accuracy of predicting IPMN *** To evaluate whole lesion CT radiomic analysis of IPMNs for predicting malignant histology compared to International Consensus *** Fifty-one subj.cts who had pancreatic surgical resection at our institution with histology demonstrating IPMN and available preoperative CT imaging were included in this retrospective *** lesion semi-automated segmentation was performed on each preoperative CT using Healthmyne software(Healthmyne,Madison,WI).Thirty-nine relevant radiomic features were extracted from each lesion on each available contrast *** analysis of the 39 radiomics features was performed for each contrast phase and values were compared between malignant and benign IPMN groups using logistic *** quantitative and qualitative CT measurements were also compared between groups,viaχ2(categorical)and Mann Whitney U(continuous)*** Twenty-nine subj.cts(15 males,age 71±9 years)with high grade or invasive tumor histology comprised the"malignant"cohort,while 22 subj.cts(11 males,age 70±7 years)with low grade tumor histology were included in the"benign"*** analysis showed 18/39 precontrast,19/39 arterial phase,and 21/39 venous phase features

AIM： To compare ecabet sodium and cimetidine in relieving symptoms of functional dyspepsia. METHODS： We performed a multi-center, prospective, randomized, double-blinded controlled trial to compare the clinical efficacy of ecabet sodium and cimetidine in patients with functional dyspepsia. Two-hundred and seventy-two patients with dyspeptic symptoms fulfilling the Rome-II criteria were enrolled from 7 centers. In the study group （115 patients）, 1.5 g ecabet sodium was given twice a day. In the control group （121 patients）, 400 mg cimetidine was given twice a day. Symptoms and parameters of quality of life were analyzed at baseline, 3, 14, and 28 d after initiating the treatment. RESULTS： Two-hundred and thirty-six patients completed the clinical trial. After 4 wk.of treatment, the rates of improvement in patients with dyspeptic symptoms were not different between two groups （77.4% in the ecabet group and 79.3% in the cimetidine group, respectively, P 〉 0.05）. Lik.wise, the rates of symptomatic improvement were not different at 3 d and 14 d. The parameters of quality of life did not change significantly during the study period in both groups. There was no clinically significant adverse event in both groups.

OBj.CTIVE:To investigate the optimal timing and underlying mechanism of electroacupuncture(EA)at Baihui(GV 20)and Dazhui(GV 14)for improved long-term functional recovery after focal cerebral ischemia in a photothrombotic strok. mouse ***:Totally 50 adult male C57 BL/6 j.mice were assigned into 5 groups:(a)the control group,sham-operated mice(n=10);(b)the vehicle group,focal cerebral ischemia induction without EA(n=10);(c)the acute EA group,mice received EA immediately post-ischemia,followed by once-daily treatments for 7 consecutive days(n=10);(d)the subacute EA group,mice received EA 4 days post-ischemia,followed by once-daily treatments for 7 consecutive days(n=10);(e)the delayed EA *** stimulation(2 Hz,2 V for 20 min)was applied to acupuncture points(acupoints),Baihui(GV 20)and Dazhui(GV 14),once a day for 7 consecutive days beginning immediately(acute treatment),4 d(subacute treatment)and 10 d(delayed treatment)after focal cerebral ischemia in C57 BL/6 j.*** assessments were conducted 21 and 28 d post-ischemia and histopathological analyses were performed 28 days ***:The subacute EA treatment at Baihui(GV20)and Dazhui(GV 14)significantly improved functional recovery compared to the vehicle group 28 d after ischemic brain inj.ry,although brain atrophy was not *** number of Neu N+and Neu N+/Brd U+cells as well as GFAP intensity in the ipsilateral cortex were significantly increased in the subacute group compared to the vehicle group 28 d *** concluded that EA stimulation 4 d post-ischemia(subacute treatment)enhanced neurogenesis and astrogliosis,lik.ly contributing to long-term functional recovery following focal cerebral ***:Our findings suggest that the timing of the EA therapy at Baihui(GV 20)and Dazhui(GV 14)determines the therapeutic effects in mice with focal cerebral ischemia induced by photothrombotic occlusion.
