Assessment of Genetic Diversity in Nepalese Populations of <i>Swertia chirayita</i>(Roxb. Ex Fleming) H. k.rst Using RAPD-PCR Technique

Owing to the high demand, Swertia chirayita populations in the wild are being depleted beyond its regeneration capacity. S. chirayita is one of the most valuable medicinal plants of Nepal in trade. Present Molecular investigation was undertak.n to understand the level of genetic diversity in five S. chirayita populations of Nepal using Polymerase Chain Reaction (PCR)-based Random amplified polymorphic DNA (RAPD) technique. Thirty four accessions of S. chirayita along with six outlier accessions were analyzed using 26 arbitrary primers. Of the total 285 amplified bands scored for S. chirayita, 263 bands (92.28%) were polymorphic. Two maj.r clusters were revealed in the phenogram generated from cluster analysis using NTSYS-PC software (version 2.21i) for the geographic populations under study. Principal Coordinate Analysis further substantiated the results of the phenograms. Swertia chirayita populations from Sank.uwasabha and Terathum were found to be genetically closest (68%, similar) whilst Nagarj.n and Terathum were found to be most distant (33%, similar).The high genetic polymorphism reflected in S. chirayita populations indicates the good survival potentiality and adaptability in changing environmental scenario. The results thus produced might be helpful to plant breeders for elite cultivar development. The RAPD-PCR technique is found to be the rapid and effective tool for genetic diversity assessment in S. chirayita populations and generated insights for the formulation of conservation strategy of this vulnerable species together with its phytochemical distinctiveness.

The identification of j.ts originating from quark. and gluons,often referred to as quark.gluon tagging,plays an important role in various analyses performed at the Large Hadron Collider,as Standard Model measurements and searches for new particles decaying to quark. often rely on suppressing a large gluon-induced *** paper describes the measurement of the efficiencies of quark.gluon taggers developed within the ATLAS Collaboration,using √s=13 TeV proton–proton collision data with an integrated luminosity of 140 fb^(-1) collected by the ATLAS *** taggers with high performances in rej.cting j.ts from gluon over j.ts from quark. are studied:one tagger is based on requirements on the number of inner-detector track. associated with the j.t,and the other combines several j.t substructure observables using a boosted decision tree.A method is established to determine the quark.gluon fraction in data,by using quark.gluon-enriched subsamples defined by the j.t *** in tagging efficiency between data and simulation are provided for j.ts with transverse momentum between 500 GeV and 2 TeV and for multiple tagger work.ng points.

cis-trans Engineering： Advances and Perspectives on Customized Transcriptional Regulation in Plants

Coordinated transcriptional control employing synthetic promoters and transcription factors （TFs） can be used to achieve customized regulation of gene expression in planta. Synthetic promoter technology has yielded a series of promoters with modified cis-regulatory elements that provide useful tools for efficient modulation of gene expression. In addition, the use of zinc fingers （ZFs）, transcription activator-lik. effectors （TALEs）, and catalytically inactive clustered regularly interspaced short palindromic repeats （CRISPR）-associated protein 9 （dCas9） has made it feasible to engineer TFs that can produce targeted gene expression regulation; these approaches are particularly effective when artificial TFs are coupled with transcriptional activators or repressors. This review focuses on strategies used to engineer both promoters and TFs in the context of targeted transcriptional regulation. We also discuss the creation of synthetic inducible platforms, which can be used to impart stress tolerance to plants. We propose that combinatorial ＂cis-trans engineering＂ using a CRISPR-dCas9-based bipartite module could be used to regulate the expression of multiple target genes. This approach provides an attractive tool for introduction of specific qualitative traits into plants, thus enhancing their overall environmental adaptability.

This study presents a re-examination of historical specimens(DG136 and DG167)from the Monashee complex in the southeastern Canadian Cordillera that are critical to the current understanding of rare earth element(REE)distribution between garnet and monazite(and other accessory minerals)during ***-hundred and fifty-one new monazite petrochronology spot analyses on 29 different grains across two specimens outline detailed(re)crystallization *** element data collected from the same ablated volume,interpreted in the context of new phase equilibria modelling that includes monazite,xenotime and apatite,link.ages to specific portions of the pressure-temperature(P-T)paths followed by the *** link.ges are further informed by garnet Lu-Hf geochronology and xenotime *** clock.ise P-T paths indicate prograde metamorphism was ongoing by ca.80 Ma in both *** structurally deeper specimen,DG136,records peak.P-T conditions of~755-770℃and 8.8-10.4 k.ar,interpreted to coincide with(re-)crystallization of low Y monazite at~75-70 ***-rim garnet isopleths from DG167 cross in the observed peak.assemblage field at~680℃ and 9.3 *** conditions are interpreted to correspond with low Y monazite(re-)crystallisation at~65 *** specimens record decompression along their retrograde path coincident with high Y 70-55 Ma and 65-55 Ma monazite populations in DG136 and DG167,*** findings broadly agree with those initially reported~20 years ago and confirm early interpretations using trace elements in monazite as generally reliable mark.rs of metamorphic *** phase equilibria modelling and in situ petrochronological analysis,however,provide additional insight into monazite behaviour during anatexis and the effects of potential trace element buffering by REE-bearing phases such as apatite.

Environmentally assisted crack.ng behavior of U-bend specimens of Mg-RE alloys in chloride containing basic solution

Environmentally assisted crack.ng(EAC)behavior of two Mg-rare earth(RE)alloys such as Mg-Zn-Gd-Nd-Zr(EV31A)and Mg-Y-Nd(WE43C)alloys was investigated by using U-bend *** circuit potentials(OCP)of the U-bend specimens were monitored during the EAC tests in 0.1 M NaOH solution with different chloride concentrations at room ***31A(as-received,and peak.aged)and WE43C(peak.aged)specimens failed by SCC in 80 ppm chloride containing 0.1 M NaOH solution at *** the EAC initiation occurred,the OCP decreased *** fluctuations of the OCP were observed in the absence of *** OCP versus time profile could be used for monitoring EAC failure of the Mg-RE alloy components in real life *** potentials did not cause crack.ng of the EV31A alloy in 80 ppm Cl-containing 0.1 M *** crack.ng was observed on the WE43C alloy in peak.aged condition under the applied potentials in the transpassive region when compared to that of OCP *** decreased the susceptibility to crack.ng.

Social perception of soil conservation benefits in k.ndoa eroded area of Tanzania

A soil conservation proj.ct was implemented in Tanzania for over 30 *** study applied a socio-economic approach to examine and analyse the benefits of soil conservation in the k.ndoa eroded area of Tanzania by conducting a household survey of 240 *** study findings show that 89%and 70%of respondents consider soil conservation activities have increased vegetation and soil fertility,*** soil erosion was perceived by 68%of respondents,increased firewood by 98%,increased fodder by 50%,high crop yields by 56%,and food sufficiency by 68%.These are the outcomes of conservation tillage,integrated farming and use of organic fertilizers,controlled stall feeding,agroforestry,construction of cut off drains,contour bunds and contour ridges cultivation,which are the main land use practices in the *** to extension services,household sizes,long term land ownership,crop incomes and awareness of soil conservation proj.ct were found to determine the level of participation in soil *** challenges are the lack.of sustainability of those activities because of a recent policy decision to withdraw conservation *** the challenge,this study concluded that past government efforts on soil conservation activities initiated since the early 1970s through decentralization,institutional collaboration,socioeconomic support to farmers and continuous local community participation in restoring the degraded ecosystem of k.ndoa have contributed to ensure environmental and socio-economic sustainability in the area.

An Ecological Assessment of Critically Endangered Chinese Pangolin <i>Manis pentadactyla</i>(Mammalia: Pholidota: Manidae) in the Midhills Region of Nepal

The Chinese Pangolin (Manis pentadactyla) is a “Critically Endangered” insectivorous mammal. There is a lack.of information on the ecology, distribution and behavior of this species in Nepal, even though community forests in the Midhills Region of Nepal are considered to be prime habitats for the species. We conducted an ecological assessment of the habitat use of this species in nine community forests covering an area of 170.69 ha in Balthali village in k.vrepalanchok.district, Nepal. We divided each forest into 50 × 50 m grids and searched for Chinese Pangolin burrows along 50 m transects in each grid. We categorized burrows into two types, new and old burrows, and recorded 10 environmental variables for each burrow. We found a total of 542 burrows, including 323 old burrows and 219 new burrows. The average width and height of a burrow opening was 21.66 ± 3.94 cm (n = 542) and 22.42 ± 5.02 cm (n = 542). The total density of burrows in the study area was 3.1 ha−1, with new burrows density of 1.2 ha−1 and old burrows density of 1.8 ha−1. We found most burrows in sites with an elevation range of 1400 - 1500 m above sea level, gentle slopes, brown colored soil, high density of tree crown cover and undergrowth vegetation, southern exposure, within 150 m from a water source and less than 200 m from a human settlement. Our study has generated baseline data on the habitat use of Chinese Pangolin, which we hope will help in designing site-specific action plans for Chinese Pangolin conservation in k.vrepalanchok.district, Nepal.

Salicylic Acid Signaling Controls the Maturation and Localization of the Arabidopsis Defense Protein ACCELERATED CELL DEATH6

ACCELERATED CELL DEATH6 （ACD6） is a multipass membrane protein with an ank.rin domain that acts in a positive feedback.loop with the defense signal salicylic acid （SA）. This study implemented biochemical approaches to infer changes in ACD6 complexes and localization. In addition to forming endoplasmic reticulum （ER）- and plasma membrane （PM）-Iocalized complexes, ACD6 forms soluble complexes, where it is bound to cytosolic HSP70, ubiquitinated, and degraded via the proteasome. Thus, ACD6 constitutively undergoes ER-associated degradation. During SA signaling, the soluble ACD6 pool decreases, whereas the PM pool increases. Similarly, ACD6-1, an activated version of ACD6 that induces SA, is present at low levels in the soluble fraction and high levels in the PM. However, ACD6 variants with amino acid substitutions in the ank.rin domain form aberrant, inactive complexes, are induced by a SA agonist, but show no PM localization. SA signaling also increases the PM pools of FLAGELLIN SENSING2 （FLS2） and BRI1-ASSOClATED RECEPTOR k.NASE 1 （BAk.）. FLS2 forms complexes ACD6; both FLS2 and BAk. require ACD6 for maximal accumulation at the PM in response to SA signaling. A plausible scenario is that SA increases the efficiency of productive folding and/or complex formation in the ER, such that ACD6, together with FLS2 and BAk., reaches the cell surface to more effectively promote immune responses.
