Assays mimicking in vitro the concentration gradients triggering biological responses like those involved in fighting infections and blood clotting are essential for biomedical research. Microfluidic assays prove especially attractive as they allow precise control of gradient shape allied to a reduction in scale. Conventional microfluidic devices are fabricated using solid plastics that prevent direct access to responding cells. Fluid-walled microfluidics allows the manufacture of circuits on standard Petri dishes in seconds, coupled to simple operating methods; cell-culture medium sitting in a standard dish is confined to circuits by fluid walls made of an immiscible fluorocarbon. We develop and experimentally validate an analytical model of diffusion between two or more aqueous streams flowing at different rates into a fluid-walled conduit with the cross-section of a circular segment. Unlike solid walls, fluid walls morph during flows as pressures fall, with wall shape changing down the conduit. The model is validated experimentally for Fourier numbers < 0.1 using fluorescein diffusing between laminar streams. It enables a priori prediction of concentration gradients throughout a conduit, so allowing rapid circuit design as well as providing bio-scientists with an accurate way of predicting local concentrations of bioactive molecules around responsive and non-responsive cells.

Source control is defined as the physical measures undertaken to eliminate the source of infection and control ongoing contamination, as well as restore anatomy and function at the site of infection. It is a key component of the management of patients with sepsis and septic shock and one of the main determinants of the outcome of infections that require source control. While not all infections may require source control, it should be considered in every patient presenting with sepsis;it is applicable and necessary in numerous infections, not only those occurring in the abdominal cavity. Although the biological rationale is clear, several aspects of source control remain under debate. The timing of source control may impact outcome;early source control is particularly relevant for patients with abdominal infections or necrotizing skin and soft tissue infections, as well as for those with more severe disease. Percutaneous procedures are increasingly used for source control;nevertheless, surgery—tailored to the patient and infection—remains a valid option for source control. For outcome optimization, adequate source control is more important than the strategy used. It should be acknowledged that source control interventions may often fail, posing a challenge in this setting. Thus, an individualized, multidisciplinary approach tailored to the infection and patient is preferable.

Obj.ctive: To systematically summarize the published literature on the genetic variants associated with nonalcoholic fatty liver disease (NAFLD). Methods: Literature from Web of Science, PubMed, and Embase between j.nuary 1980 and September 2022 was systematically searched. Meta-analyses of the genetic variants were conducted using at least five data sources. The epidemiologic credibility of the significant associations was graded using the Venice criteria. Results: Based on literature screening, 399 eligible studies were included, comprising 381 candidate gene association, 16 genome-wide association, and 2 whole-exome sequencing studies. We identified 465 genetic variants in 173 genes in candidate gene association studies, and 25 genetic variants in 17 genes were included in the meta-analysis. The meta-analysis identified 11 variants in 10 genes that were significantly associated with NAFLD, with cumulative epidemiological evidence of an association graded as strong for two variants in two genes (HFE, TNF), moderate for four variants in three genes (TM6SF2, GCKR, and ADIPOQ), and weak for five variants in five genes (MBOAT7, PEMT, PNPLA3, LEPR, and MTHFR). Conclusion: This study identified six variants in five genes that had moderate to strong evidence of an association with NAFLD, which may help understand the genetic architecture of NAFLD risk. © 2024 The Editorial Board of Biomedical and Environmental Sciences

Post-stroke epilepsy (PSE) is a severe complication that occurs after a stroke and is associated with adverse neurological outcomes and a higher risk of mortality. Achieving an accurate prediction of PSE holds significant importance for the execution of randomized controlled trials and the development of clinical diagnosis and treatment protocols. This article provides an overview of PSE, including its definition, current epidemiological status, and associated risk factors. It also offers a detailed account of the progress in PSE risk prediction models. Furthermore, it looks ahead to the construction of predictive models using a multimodal dataset combined with machine learning algorithms, with the goal of achieving precise prediction of PSE. © 2024 Chinese Medical j.urnals Publishing House ***. The effects of the gravitational redshift of gravitons upon spiral galaxy rotation energy are compared to the standard mass to light analyses in obtaining rotation curves. The derivation of the total baryonic matter compares well with the standard theory and the rotation velocity is matched to a high precision. The stellar mass distributions obtained from the fit with graviton energy loss are used to derive the surface brightness magnitudes for the galaxies, which agree well with the observed measurements. In a new field of investigation, the graviton theory is applied to the observations of gravitational lenses. The results of these applications of the theory suggest that it can augment the standard methods and may eliminate the need for dark matter.

Usually, models of globular star clusters are created by analyzing their luminosity and other observation parameters. The goal of this work is to create stable models of globular clusters based on the laws of mechanics. It is necessary to set the coordinates, velocities and masses of the stars so that as a result of their gravitational interaction the globular cluster is not destroyed. This is not an easy task, and it has been solved in this paper. Using an exact solution of the axisymmetric gravitational interaction of N-bodies, single-layer spherical structures were created. They are combined into multilayer models of globular clusters. An algorithm and a program for their creation is described. As a result of solving the problem of gravitational interaction of N bodies, evolution of 5-, 10-, and 15-layer structures was studied. During the inter-body interaction, there proceeds a transition from the initial specially organized structure to a structure with bodies, uniformly distributed in space. The number of inter-body collisions decreases, and the globular cluster model passes into the stable form of its existence. The collisions of bodies and the acquisition of rotational motion and thermal energy by them are considered. As a result of the passage to scaled dimensions, the results were recalculated to the conditions of globular star clusters. The periods of rotation and the temperatures of merged stars are calculated. Attention is paid to a decreased central-body mass in the analyzed models of globular star clusters.

Fresnel Equations Derived Using a Non-Local Hidden-Variable Particle Theory

Problem: The Fresnel equations describe the proportions of reflected and transmitted light from a surface, and are conventionally derived from wave theory continuum mechanics. Particle-based derivations of the Fresnel equations appear not to exist. Approach: The obj.ctive of this work was to derive the basic optical laws from first principles from a particle basis. The particle model used was the Cordus theory, a type of non-local hidden-variable (NLHV) theory that predicts specific substructures to the photon and other particles. Findings: The theory explains the origin of the orthogonal electrostatic and magnetic fields, and re-derives the refraction and reflection laws including Snell’s law and critical angle, and the Fresnel equations for s and p-polarisation. These formulations are identical to those produced by electromagnetic wave theory. Contribution: The work provides a comprehensive derivation and physical explanation of the basic optical laws, which appears not to have previously been shown from a particle basis. Implications: The primary implications are for suggesting routes for the theoretical advancement of fundamental physics. The Cordus NLHV particle theory explains optical phenomena, yet it also explains other physical phenomena including some otherwise only accessible through quantum mechanics (such as the electron spin g-factor) and general relativity (including the Lorentz and relativistic Doppler). It also provides solutions for phenomena of unknown causation, such as asymmetrical baryogenesis, unification of the interactions, and reasons for nuclide stability/instability. Consequently, the implication is that NLHV theories have the potential to represent a deeper physics that may underpin and unify quantum mechanics, general relativity, and wave theory.
