ETM study of electroporation influence on cell morphology in human malignant melanoma and human primary gingival fibroblast cells

Objective:To estimate electroporation(EP) influence on malignant and normal ***: Two cell lines including human malignant melanoma(Me-43) and normal human gingival fibroblast(HCFs) were *** parameters were the following:230,1000,1 730,2 300 V/cm;30 μ s by 3 impulses for every *** viability of cells after EP was estimated by MTT assay. The ullrastructural analysis was observed by transmission electron microscope(Zeiss EM 900). Results:In the current study we observed the intracellular effect following EP on Me-43 and HGF *** the conditions applied,we did not observe any significant damage of mitochondrial activity in both cell lines treated by ***,we showed that EP in some conditions can stimulate cells to *** changes induced by EP were only visible in electron *** fibroblast cells we observed significant changes in lower parameters of EP(230 and 1 000 V/cm).After applying higher electric field intensities(2 300 V/cm) we detected many vacuoles,myelin-like bodies and swallowed endoplasmic *** melanoma cells such strong pathological modifications after EP were not observed,in comparison with control cells. The ultrastructure of both treated cell lines was changed according to the applied parameters of ***:We can claim that EP conditions are cell line *** terms of the intracellular morphology,human fibroblasts are more sensitive to electric field as compared with melanoma *** conditions should be determined for each cell *** our study,we can conclude that EP is not an invasive method for human normal and malignant cells. This technique can be safely applied in chemotherapy for delivering drugs into tumor cells.

There is a wide diversity of plants and seasonal crops in Peru, due to the presence of many climatic zones. Numerous plants are used to cure or prevent diseases. These plants are promising candidates for functional foods products. The most frequent form in which they are used is an aqueous infusion or decoction. In this study, we compared the antioxidant properties of ten Peruvian plants infusions and investigated their relation to the phenolic content. The studied plants were: Uncaria tomentosa (cat’s claw), Lepidium meyenii (maca), Berberis vulgaris L. (barberry, agracejo), Phyllantus niruri (chanca piedra), Annona muricata L. (graviola, soursop), Gentianella alborosea (hercampure), Geranium dielsianum (pasuchaca), Tabebuia ochracea (tahuari), Notholaena nivea (“cuti cuti”) and Tiquilia paronychioides (“flor de arena”). Infusions of all studied plants have shown antioxidant activity, though there was a large diversity between the results. The antioxidant properties, determined with DPPH and ABTS scavenging assays as well as FRAP test, were strongly correlated with total phenolic content, while there was no correlation with the carotenoid content.

The most common clinical manifestation of gram negative Salmonella typhimurium infection is gastroenteritis. In Poland extraintestinal infections occur in about 0.4% of cases of reported salmonellosis. Poor personal hygiene practices of parents are the most common cause of Salmonella spp. infections in the neonatal period. The neonatal salmonellosis is significantly more frequently reported in endemic areas such as Central Africa or India, than in Poland. The authors present a case of a newborn with Salmonella typhimurium sepsis, who was treated in the Department of Intensive Care and Congenital Malformations of the Neonates and Infants, Polish Mother’s Memorial Research Institute.

AIM: Transforming growth factor (TGF)-β1, metalloproteinase (MMP)-1 and its tissue inhibitor (TIMP)-I are considered predictive biomarkers of chronic hepatitis activity and *** aim of this study was to evaluate the effect of lamivudine treatment on the plasma levels of these peptides in patients with chronic hepatitis ***: TGF-β1, MMP-1 and TIMP-1 plasma concentrations were measured with an enzyme immunoassay in 40 patients treated with lamivudine for 48 wk. Elimination of HBV-DNA and HBV antigens was evaluated 24 wk after treatment ***: Baseline TGF-β1(29.6±2.2 ng/mL) and TIMP-1(1 578±93 ng/mL) significantly exceeded normal values(18.3±1.6 ng/mL and 1 102±67 ng/mL respectively). Lamivudine treatment resulted in a significant decrease of TGF-β1 and TIMP-1 during treatment with an increase after 24 wk of treatment. Pretreatment MMP-1 levels (6.7±0.7 ng/mL) were significantly lower than normal values (11.9±0.9 ng/mL) and increased during treatment and follow-up. A significant correlation was noted between TGF-β1 or TIMP-1 and aminotransferases as well as fibrosis scored in liver biopsy specimens. There were no statistically significant differences of TGF-β1, TIMP-1 and MMP-1 between four groups at baseline, 24 and 48 wk of treatment. TGF-β1 and TIMP-1 levels increased significantly in non-responders and normalized in responders at wk 72. MMP-1 also normalized in responders and decreased to values significantly lower than normal in ***: These findings support the role of TGF-β1,TIMP-1 and MMP-1 in the pathogenesis of chronic hepatitis *** of their association with hepatic injury and antiviral treatment efficacy, determination of these peptides may be useful in disease management.

Flood frequency analysis (FFA) concentrates on peak flows of flood hydrographs. However, floods that last years devastated large parts of Poland lead us to revision of the views on the assessment of flood risk in Poland. It turned out that it is the prolonged exposure to high water on levees that causes floods, not only the water overflowing the levee crest. This is because, the levees are weakened by water and their disruption occurs when it seems that the danger is over, i.e. after passing culmination. Two main causes of inundation of embanked rivers, namely over-crest flow and wash out of the levees, are combined to assess the total risk of inundation. Therefore the risk of inundation is the total of risk of exceeding embankment crest by flood peak and risk of washout of levees. Hence, while modeling the flood events in addition to the maximum flow one should consider also the duration of high water in a river channel, Analysis of the frequency of annual peak flows based on annual maxima and peaks over threshold is the subject of countless publications. Therefore we will here mainly modeling the duration of high water levels. In the paper the two-component model of flood hydrograph shape i.e. “duration of flooding-discharge- probability of nonexceedance” (DqF), with the methodology of its parameters estimation for stationary case was developed as a completion to the classical FFA with possible extension to non stationary flood regime. The model combined with the technical evaluation of probability of levees breach due to the d-days duration of flow above alarm stage gives the annual probability of inundation caused by the embankment breaking. The results of theoretical research were supplemented by a practical example of the model application to the series for daily flow in the Vistula River in Szczucin.
