Optimization of the Protocol for the <i>in Vitro</i>Cultivation of <i>Piper aduncum</i>L.

Piper aduncum L. (Piperaceae) has great potential for economic exploration because of the proven use of its essential oil in the agriculture and in the human health. A technique that improves its propagation, as the tissue culture, becomes necessary. Some parameters must be determined for the successful cultivation in vitro. Thus, this study aimed to determine the salts concentration of MS medium, temperature, luminosity and light quality for in vitro culture of this species. The following treatments were conducted: 1/4MS, 1/2MS, MS and 2MS;20℃, 25℃, 30℃ and 35℃;monochromatic blue, red and white lights and the combination of red and blue, using light emitting diodes (LEDs);luminosities of 17, 37, 48 and 73 μmol·m-2·s-1 obtained with cool white fluorescent lamp. After 35 days, the treatments were analyzed. To determine the salts concentration of MS, the variables were evaluated: number of shoots, leaves and roots, length and width of leaves, length and dry mass of root and shoots and percentage of death by necrosis. To the plantlets under different temperatures, length and dry mass of shoot and root, number of shoots, number, length and width of leaves and survival and rooting percentages were measured. The plantlets that were maintained under different luminosities were evaluated for length of shoot and root, dry mass of leaf and root. To evaluate the growth under the LEDs, the length and dry mass of shoot and root, number of shoots and roots, percentage of rooting and sprouting were assessed. The medium 1/4MS and the medium 1/2MS showed better responses for number and length of root, leaf width and shoot length. The temperature 25℃ provided the highest number of leaves, length of shoot and root, root dry mass and rooting percentage. The luminosity 73 μmol·m-2·s-1 provided higher values in length of root and dry mass of leaf and root. The red LED provided plantlets with greater growth. B-1 cells modulate the murine macrophage response to Leishmania major infection

AIM To investigate the modulatory effect of B-1 cells on murine peritoneal macrophages infected with Leishmania major(L. major) in *** Peritoneal macrophages obtained from BALB/c andBALB/c XID mice were infected with L. major and cultured in the presence or absence of B-1 cells obtained from wild-type BALB/c mice. Intracellular amastigotes were counted, and interleukin-10(IL-10) production was quantified in the cellular supernatants using an enzymelinked immunosorbent assay. The levels of the lipid mediator prostaglandin E2(PGE2) were determined using a PGE2 enzyme immunoassay kit(Cayman Chemical, Ann Arbor, MI), and the number of lipid bodies was quantified in the cytoplasm of infected macrophages in the presence and absence of B-1 cells. Culturing the cells with selective PGE2-neutralizing drugs inhibited PGE2 production and confirmed the role of this lipid mediator in IL-10 production. In contrast, we demonstrated that B-1 cells derived from IL-10 KO mice did not favor the intracellular growth of L. *** We report that B-1 cells promote the growth of L. major amastigotes inside peritoneal murine macrophages. We demonstrated that the modulatory effect was independent of physical contact between the cells, suggesting that soluble factor(s) were released into the cultures. We demonstrated in our co-culture system that B-1 cells trigger IL-10 production by L. major-infected macrophages. Furthermore, the increased secretion of IL-10 was attributed to the presence of the lipid mediator PGE2 in supernatants of L. major-infected macrophages. The presence of B-1 cells also favors the production of lipid bodies by infected macrophages. In contrast, we failed to obtain the same effect on parasite replication inside L. major-infected macrophages when the B-1 cells were isolated from IL-10 knockout mice. CONCLUSION Our results show that elevated levels of PGE2 and IL-10 produced by B-1 cells increase L. major growth, as indicated by the number of parasites

HPV Vaccine: Integrative Review of National and International guidelines

HPV (Human Papillomavirus) is an adenovirus of the Papillomaviridae family with approximately 200 subtypes (subdivided into low and high oncogenic risk groups), responsible for almost 99% of cervical cancers. Worldwide, there are more than 570,000 new cases and more than 311,000 women die each year from cervical cancer;for Brazil, the estimate is of more than 16,000 new cases per year. It is known that cervical cancer is one of the most easily preventable forms of cancer, as there is a highly effective vaccine against HPV. However, the morbidity and mortality rates correlated with human papillomavirus are still concerning, especially in less developed countries. In 2020, the WHO (World Health Organization) presented three goals to achieve by 2030 the worldwide extermination of cervical cancer;among these goals is vaccination. Is the world preparing for this battle? The present work is an integrative review, comparing national and international guidelines for HPV vaccination published in the last five years in the consulted databases, using the descriptors “cervical cancer”, “prevention” and “guideline” and the boolean operator “and”, from which nine articles were selected. The study made it possible to compare international and national guidelines for vaccination against HPV, showing that developed countries implemented the HPV vaccine longer ago and follow the WHO recommendations more rigorously and effectively than underdeveloped or developing countries. Adopting a single dose as a strategy, as suggested by the WHO in April 2022, could be an important step towards increasing coverage and providing protection for a greater number of girls who do not have access to HPV immunization.

Liver Histology after Chronic Use of Alcohol and Exercise Training in Rats

Objectives： To investigate the effects of physical training on the liver morphology and morphometry after chronic use of alcohol in rats. Methods： Twenty-four Wistar rats were housed in cages with controlled environment and randomly divided into four groups according to treatment received. In the initial treatment, alcohol was administered to SA （sedentary alcohol） and EA （exercise alcohol） groups. After four weeks, physical training program was held on a treadmill with EA and EC （exercise control） groups. Area, perimeter, maximum and minimum diameter and form factor of nucleus and cytoplasm of hepatocytes were analyzed. Key findings： Micro-vesicular fatty degeneration, predominantly pericentrolobular, of mild to moderate intensity, was found especially in animals treated with alcohol. EC group showed nucleus area greater than the nucleus area of EA and SA groups. The form factor was lower in the EC group than in the EA group. EA group showed maximum cytoplasm diameter is smaller than in SC （sedentary control） group. Conclusions： Physical training for two weeks was not enough to suppress histopathologic changes in the liver caused by chronic use of alcohol in rats. Chronic use of alcohol seems to have minimized the beneficial effect of physical training in the nucleus area of hepatocytes.

Allelopathy of the Leaf Extract of Eucalyptus Genetic Material on the Physiological Performance of Millet Seeds

This study aims to evaluate the possible allelopathic effects of young and mature leaf extract from of two eucalyptus genetic materials on the physiological performance of millet seeds. The experiment was carried out in a completely randomized design, in a 5 × 2 × 2 factorial arrangement [five concentrations of the extract (0, 25%, 50%, 75%, and 100%) × two genetic materials (1 - Eucalyptus grandis × Eucalyptus urophylla and 2 - Eucalyptus grandis × Eucalyptus camaldulensis) × two maturation stages of leaves (young and mature leaves)] with four replicates, each one with 25 seeds. Seeding was performed on the germitest paper substrate distributed in Petri dishes, moistened with the extract according to the treatments, being allocated in biochemical oxygen demand (B.O.D) adjusted at 25°C and 12 h photoperiod. Percentage, speed index, mean germination time, aerial part and primary root length, and dry matter weight of aerial part and root system of seedlings were evaluated. The leaf extracts of the two eucalyptus clones had all the analyzed variables changed with effect more evident on young leaf extract of the genetic material 1. It can be concluded that the tested genetic materials have a detrimental effect on the physiological performance of millet seeds.

AIM To evaluate the sex-specific effects of a hydroalcoholic extract from Eugenia punicifolia(HEEP) leaves on gastric ulcer *** In this rat study involving males, intact(cycling) females, and ovariectomized females, gastric ulcers were induced using acetic acid. A vehicle, lansoprazole, or HEEP was administered for 14 d after ulcer induction. Body weight was monitored throughout the treatment period. At the end of treatment, the rats were euthanized and the following in vivo and in vitro investigations were performed: macroscopic examination of the lesion area and organ weights, biochemical analysis, zymography, and evaluation of protein expression levels. Additionally, the concentration-dependent effect of HEEP was evaluated in terms of subacute toxicity and *** Compared to the vehicle, HEEP demonstrated a great healing capacity by substantially reducing the ulcerative lesion area in males(52.44%), intact females(85.22%), and ovariectomized females(65.47%), confirming that HEEP accelerates the healing of acetic acidinduced gastric lesions and suggesting that this effect is modulated by female sex hormones. The antiulcer effect of HEEP was mediated by prostaglandin E2 only in male rats. Overall, the beneficial effect of HEEP was the highest in intact females. Notably, HEEP promoted the expression of vascular endothelial growth factor(intact vs ovariectomized females) and decreased the expression of Caspase-8 and Bcl-2(intact female vs male or ovariectomized female). Additionally, HEEP enhanced fibroblast proliferation and migration into a wounded area in vitro, confirming its healing effect. The Cumulative Effects of Sewage Sludge Compost on Raphanus sativus L: Growth and Soil Properties

The main goal of this research study was to establish the physicochemical characteristics of sewage sludge, and the nutrients needed to enhance the properties of soil with a view to exploiting were potential for agriculture. The example used was the cultivation of radish (Raphanus sativus). The findings are intended as a contribution towards making agriculture sustainable. This study was carried out to assess the response of radish (Raphanus sativus L.) to doses of sewage sludge applied in medium texture soils, and the socioeconomic contributions that such cultivation of radish would make. Levels of 0.25 and 75 ton/ha of sewage sludge plus the equivalent dose of 25 ton/ha of sewage sludge vermin compost by California Red were used after making a randomized experimental design with three replicates. 30 days after sowing radish seeds, the following data were collected: the number of leaves per plant (NLP);plant height (PH in cm);root diameter (RD) and tubercle production (TP ton/ha). In addition, the pH, EC-electric conductivity (EC), and soil organic matter (SOM) were determined. Statistical analysis showed that 25 and 50 tons/ha gave the best results for the parameters assessed. The estimated increase in family mean income from selling radish crops fertilized with sewage sludge is around 17%.

Displacement of the non-reduction temporomandibular disk with limitation of opening. Story of a clinical case—Follow up five years

Displacement of the non-reduction disk with limitation of opening (DDwLO) can be treatment of the non-surgical form. However, it required scientific and clinic knowledge of diagnostic area of the disorders temporomandibulares. This study was performed with the patient CN, 18 years, white, female, who went to the orofacial pain ambulatory with complaints of extreme pain on the right temporomandibular joint (TMJ) and limitation of maximum opening of 29 mm with shift to right, which she has been suffering for at least three years. After a criterion anamnese the diagnosis hypothesis found was a displacement of the non-reduction disk with limitation of opening. The treatment was based on infiltration in the right TMJ with anesthesic followed by mandibular manipulation. After the therapeutic conduct the patient presented clinical signals of normality with absence of trismus and mandibular movement without shift to right. The patient was followed up during five years not presenting return of the pathology.

In Silico and in Vitro Analysis of Pyrazolone Derivatives against Zika Virus and Identification of Potential NS5 Methyltransferase Inhibitors by Molecular Docking

Zika virus (ZIKV), a mosquito-borne flavivirus, has been associated with benign infections for decades. However, it has become a public health concern due to its association with severe fetal and neurological complications. Although many efforts have been made to control ZIKV infection, approved vaccines or antiviral drugs are still lacking. Consequently, the development of new effective anti-ZIKV agents is urgently needed. In this context, we investigated the antiviral potential of pyrazolone derivatives against ZIKV replication using in silico and in vitro methods. The four pyrazolone derivatives evaluated (1a, 1b, 1c, and 1d) inhibited over 50% of ZIKV replication with low cytotoxicity. Among them, compound 1b exhibited the most potent activity (EC50 = 4.3 μM) and the highest selectivity (SI = 342). Mechanism of action studies indicated that these compounds act at early stages of virus replication, and compound 1b can also directly inactivate ZIKV particles. Molecular docking studies suggested that these compounds can bind to and block the activity of ZIKV NS5 methyltransferase. Finally, pharmacokinetic and toxicological predictions have reinforced the safety and drug-like profiles of these derivatives. In conclusion, the pyrazolone scaffold proved to be valuable for anti-ZIKV drug development, and the derivatives studied deserve further investigation.
