Mesenchymal stem cells(MsCs)play diverse roles ranging from regeneration and wound healing to immune *** investigations have indicated the crucial role of these multipotent stem cells in regulating various aspects of the immune *** express unique signaling molecules and secrete various soluble factors that play critical roles in modulating and shaping immune responses,and in some other cases,MSCs can also exert direct antimicrobial effects,thereby helping with the eradication of invading ***,it has been demonstrated that MSCs are recruited at the periphery of the granuloma containing Mycobacterium tuberculosis and exert"Janus"-like functions by harboring pathogens and mediating host protective immune *** leads to the establishment of a dynamic balance between the host and the *** function through various immunomodulatory factors such as nitric oxide(NO),IDO,and immunosuppressive ***,our group has shown that *** uses MSCs as a niche to evade host protective immune surveillance mechanisms and establish *** also express a large number of ABC efflux pumps;therefore,dormant *** residing in MSCs are exposed to a suboptimal dose of ***,it is highly likely that drug resistance is coupled with dormancy and originates within *** this review,we discussed various immunomodulatory properties of MsCs,their interactions with important immune cells,and soluble *** also discussed the possible roles of MsCs in the outcome of multiple infections and in shaping the immune system,which may provide insight into therapeutic approaches using these cells in different infection models.

Condensed tannins(CTs)are one of the promising compounds due to their potentially health-promoting *** this study,CTs were extracted from a Leucaena leucocephala hybrid-Rendang and subjected to various biological studies including antioxidant(using Ferric reducing antioxidant power(FRAP),DPPH and ABTS radical scavenging assay),anti-microbial(against different pathogens)and cytotoxic activities(toward human breast adenocarcinoma(MCF-7),human colon carcinoma(HT29),human cervical carcinoma(HeLa)and human liver carcinoma(HepG2)cell lines)in cancer cells through in vitro *** structural characteristics and purity of CTs extract were determined using 13C *** results showed that CTs exhibited higher in vitro antioxidant activities(2257.12±80.55 mg TEAC/g extract,605.3±1.82 mg TEAC/g extract and 1014.03±1.20 mg TEAC/g extract in FRAP,ABTS and DPPH assay,respectively)and demonstrated anti-microbial activities toward selected Gram’s positive and Gram’s negative bacteria tested with MIC and MBC value at 6.25–50 mg/***,among other selected cancer cells,CTs also demonstrated cytotoxic activity toward human breast cancer cells(MCF-7)(IC50=38.33±2.08mg/mL).Characteristic of apoptosis such as cell shrinkage,nuclear condensation and apoptotic bodies were shown in *** preliminary investigations have provided scientific rationale to use CTs as an alternative therapy for various oxidative and inflammatory associated diseases.©2016 Beijing Academy of Food *** and hosting by Elsevier *** rights reserved.

A new series of N-arylquinoline derivatives 5a-x bearing 2-morpholinoquinoline moiety has been synthesized by one pot base catalyzed cyclocondensation reaction of 2-morpholinoquinoline-3-carbaldehydes 2a-c,malononitrile 3 andβ-enaminones 4a-h. All the synthesized compounds were screened for their in vitro antimicrobial activity against six bacterial pathogens,namely Streptococcus pneumoniae,Clostridium tetani,Bacillus subtilis,Salmonella typhi,Vibrio cholerae,Escherichia coli and against two fungal pathogens,Aspergillus fumigatus and Candida albicans using broth microdilution MIC *** the compounds studied,majority of the compounds were found to active against ***,*** and *** as compared to first-line standard drugs.

Determination of asenapine in presence of its inactive metabolites in human plasma by LC-MS/MS

A highly selective and sensitive liquid chromatography-tandem mass spectrometry （LC-MS/MS） assay has been described for the determination of asenapine （ASE） in presence of its inactive metabolites N-desmethyl asenapine （DMA） and asenapine-N-glucuronide （ASG）. ASE, and ASE 13C-d3, used as internal standard （IS）, were extracted from 300 μL human plasma by a simple and precise liquid-liquid extraction procedure using methyl tert-butyl ether. Baseline separation of ASE from its inactive metabolites was achieved on Chromolith Performance RPse （100 mm ×4.6 mm） column using acetonitrile- 5.0 mM ammonium acetate-10% formic acid （90：10：0.1, v/v/v） within 4.5 min. Quantitation of ASE was done on a triple quadrupole mass spectrometer equipped with electrospray ionization in the positive mode. The protonated precursor to product ion transitions monitored for ASE and ASE 13C-d3 were m/z 286.1 → 166.0 and m/z 290.0 → 166.1, respectively. The limit of detection （LOD） and limit of quantitation （LOQ） of the method were 0.0025 ng/mL and 0.050 ng/mL respectively in a linear concentration range of 0.050-20.0 ng/mL for ASE. The intra-batch and inter-batch precision （% CV） and mean relative recovery across quality control levels were 〈 5.8% and 87.3%, respectively. Matrix effect, evaluated as IS-normalized matrix factor, ranged from 1.03 to 1.05. The stability of ASE under different storage conditions was ascertained in presence of the metabolites. The developed method is much simpler, matrix free, rapid and economical compared to the existing methods. The method was suc- cessfully used for a bioequivalence study of asenapine in healthy Indian subjects for the first time.
