There has been a growing interest over the past few years in the impact of male nutrition on fertility. Infertility has been linked to male overweight or obesity, and conventional semen parameter values seem to be altered in case of high body mass index （BMI）. A few studies assessing the impact of BMI on sperm DNA integrity have been published, but they did not lead to a strong consensus. Our objective was to explore further the relationship between sperm DNA integrity and BMI, through a 3-year multicentre study. Three hundred and thirty male partners in subfertile couples were included. Using the terminal uridine nick-end labelling （TUNEL） assay, we observed an increased rate of sDerm DNA damage in obese men （odds ratio （95% confidence interval）： 2.5 （1.2-5.1））.

Tumor necrosis factor α （TNF-α） has been recognized as an activator of nuclear factor kB （NF-kB）, a factor implicated in the protection of many cell types from apoptosis. We and others have presented evidence to suggest that Fas-induced apoptosis may be an important aspect of the resolution of inflammation, and that delayed resolution of inflammation may be directly associated with NF-kB-dependent resistance to Fas. Because TNF-α activates NF-kB in many cell types including inflammatory cells such as eosinophils, we examined effects of TNF-α signaling on the Fas-mediated killing of an eosinophilic cell line AML14. While agonist anti-Fas （CHII） treatment induced apoptosis in AML14 cells, no significant cell death occurred in response to TNF-α alone. Electrophoretic mobility shift assay （EMSA） revealed that TNF-α induced NF-kB transactivation in AMLI4 cells in a time- and dose-dependent fashion, and subsequent supershift assays indicated that the translocated NF-kB was the heterodimer p65 （RelA）/p50. Pre-treatment of cells with TNF-α dramatically decreased the CHll-induced cell death in a transient fashion, accompanied by suppression of activation of caspase-8 and caspase-3 activation. Inhibition of NF-kB transactivation by inhibitors, BAY 11-7085 and parthenolide, reversed the suppression of Fas-mediated apoptosis by TNF-α. Furthermore, TNF-α up-regulated X-linked inhibitor of apoptosis protein （XIAP） transiently and XIAP levels were correlated with the temporal pattern of TNF-α protection against Fas-mediated apoptosis. This finding suggested that TNF-α may contribute to the prolonged survival of inflammatory cells by suppression of Fas-mediated apoptosis, the process involved with NF-kB transactivation, anti-apoptotic XIAP up-regulation and caspase suppression. Petrography of alluvial sands as a past and present environmental indicator:Case of the Loire River(France)

Four sample sets of the Upper and Middle Loire river sands were analyzed in order to study the impact of natural and anthropogenic factors on their petrographic composition in space(on an 800 km stretch) and *** was determined by modal analysis of three sand-size fractions using a polarizing optical microscope and calculated for each sample("standard sand" = Sst).The watershed is composed mainly of endogenic(Massif Central) and sedimentary(southern Parisian Basin) rocks.B-set sands collected in channels for different water flows in 1996 show that Sst compositions vary by only 5%.Present-day sands in the Upper Loire and Middle Loire have very high petrographic immaturity comparing to others worldwide fluvial sands,although bio-climatic conditions favor sand maturation by source-rock weathering in the *** shows the strong impact of the Massif Central on sediment yield due to relief rejuvenation as a consequence of the formation of the Alps during the *** sands stored during the Weichselian and the Holocene in the Middle Loire floodplain,although partly weathered since their deposition,show higher inputs from the endogenic rocks of the Massif Central than present-day *** can be explained by Weichselian periglacial conditions and the development of crop farming since the Neolithic,which favored mechanical erosion,particularly in the Massif Central which is characterized by a cold,humid climate and steep *** upstream-downstream change in the composition of presently deposited sand is low in the diked *** shows however that basalt and some heavy mineral grains are vulnerable to abrasion during transport and indicates a marked sediment yield from ancient sediment stored in the *** is in line with the high incision of the river bed over the last 150 years partly due to dam construction and aggregate mining.

Actigraphy: A new diagnostic tool for hepatic encephaiopathy

AIM： To assess the actigraphy, an ambulatory and continuous monitoring of wrist motor activity fitted to study sleep/wake patterns in hepatic encephalopathy （HE）. METHODS： Twenty-five cirrhotic patients （17 M, 8 F, mean age 56± 11 years, 24/25 alcoholic, Child-Pugh A, B, C： 2, 6, 17） were included. The patients were classified into 3 groups： stage 0 group （n = 12）, stage 1-2 group （n = 6）, and stage 3-4 group （n = 7） of encephalopathy. Over three consecutive days, patients had clinical evaluation 3 times a day with psychometric test, venous ammoniemia, flash visually evoked potentials （VEP）, electroencephalogram and continuous actigraphic monitoring for 3 d, providing 5 parameters： mesor, amplitude, acrophase, mean duration of activity （MDAI） and inactivity （MDII） intervals. RESULTS： Serum ammonia and VEP did not differ among the 3 groups. Electroencephalography mean dominant frequency （MDF） correlated significantly with clinical stages of HE （r=0.65, P=0.003）. The best correlation with HE stage was provided by actigraphy especially with MDAI （r= 0.7, P〈10^-4） and mesor （r= 0.65, P〈 10^-4）. MDAI correlated significantly with MDF （r= 0.62, 0.004） and was significantly shorter in case of HE compared to patients without HE （stage 0： 5.33± 1.6 min; stage 1-2：3.28±1.4 min; stage 3-4：2.52±1.1 min; P〈0.05）. Using a threshold of MDAI of less than 4.9 min, sensitivity, specificity, positive predictive value, negative predictive value for HE diagnosis were 85%, 67%, 73% and 80%, respectively. CONCLUSION： Actigraphy may be an objective method to identify HE, especially for early HE detection. Motor activity at the wrist correlates well with clinical stages of HE. MDAI and mesor are the most relevant parameters.

Laboratory pull-out tests were conducted on the following rock bolts and cable bolts:steel rebars,smooth steel bars,fiberglass reinforced polymer threaded bolts,flexible cable bolts,IR5/IN special cable bolts and Mini-cage cable *** diameter of the tested bolts was between 16 mm and 26 *** bolts were grouted in a sandstone sample using resin or cement *** tests were conducted under either constant radial stiffness or constant confining pressure boundary conditions applied on the outer surface of the rock *** most tests,the rate of displacement was about 0.02 mm/*** tests were performed using a pull-out bench that allows testing a wide range of *** paper provides an extensive database of laboratory pull-out test results and confirms the influence of the confining pressure and the embedment length on the pull-out response(rock bolts and cable bolts).It also highlights the sensitivity of the results to the operating conditions and to the behavior of the sample as a whole,which cannot be neglected when the test results are used to assess the bolt-grout or the grouterock interface.

Hepato-biliary profile of potential candidate liver progenitor cells from healthy rat liver

AIM： To evaluate the presence of progenitor cells in healthy adult rat liver displaying the equivalent ad- vanced hepatogenic profile as that obtained in humans. METHODS： Rat fibroblastic-like liver derived cells （rFLDC） were obtained from collagenase-isolated liver cell suspensions and characterized and their phenotype profile determined using flow cytometry, immunocyto- chemistry, reverse transcription polymerase chain reac- tion and functional assays. RESULTS： rFLDC exhibit fibroblastoid morphology, ex- press mesenchymal （CD73, CD90, vimentin, m-smooth muscle actin）, hepatocyte （UGTIA1, CK8） and biliary （CK19） markers. Moreover, these cells are able to store glycogen, and have glucose 6 phosphatase activity, but not UGTIA1 activity. Under the hepatogenic differentia- tion protocol, rFLDC display an up-regulation of hepatocyte markers expression （albumin, tryptophan 2,3-di- oxygenase, G6Pase） correlated to a down-regulation of the expression of the biliary marker CK19. CONCLUSION： Advanced hepatic features observed in human liver progenitor cells could not be demonstrated in rFLDC. However, we demonstrated the presence of an original rodent hepato-biliary cell type.

BACKGROUND Gastrointestinal tumors are among the most common cancer types,and early detection is paramount to improve their ***-free DNA(cfDNA)liquid biopsy raises significant hopes for non-invasive early *** To describe current applications of this technology for gastrointestinal cancer detection and *** A systematic review of the literature was performed across the PubMed *** reporting the use of cfDNA liquid biopsy in the screening or diagnosis of gastrointestinal cancers were included in the *** A total of 263 articles were screened for eligibility,of which 13 articles were *** investigated colorectal cancer(5 studies),pancreatic cancer(2 studies),hepatocellular carcinoma(3 studies),and multi-cancer detection(3 studies),including gastric,oesophageal,or bile duct cancer,representing a total of 4824 *** sensitivities ranged from 71% to 100%,and specificities ranged from 67.4% to 100%.Pre-cancerous lesions detection was less performant with a sensitivity of 16.9% and a 100% specificity in one *** study using a large biobank demonstrated a 94.9% sensitivity in detecting cancer up to 4 years before clinical symptoms,with a 61% accuracy in tissue-of-origin *** cfDNA liquid biopsy seems capable of detecting gastrointestinal cancers at an early stage of development in a non-invasive and repeatable manner and screening simultaneously for multiple cancer types in a single blood *** trials in clinically relevant settings are required to determine the exact place of this technology in gastrointestinal cancer screening and diagnosis strategies.

Mycotoxins are fungal secondary metabolites detected in many agricultural commodities, especially cereals. Due to their high consumption of cereals, pigs are exposed to these toxins. In the European Union, regulations and/or recommendations exist in pig feed for aflatoxins, ochratoxin A, fumonisins,zearalenone, and trichothecenes, deoxynivalenol and T-2 toxin. These mycotoxins have different toxic effects, but they all target the immune system. They have immunostimulatory or immunosuppressive effects depending on the toxin, the concentration and the parameter investigated. The immune system is primarily responsible for defense against invading organisms. The consequences of the ingestion of mycotoxin-contaminated feed are an increased susceptibility to infectious diseases, a reactivation of chronic infection and a decreased vaccine efficacy. In this review we summarized the data available on the effect of mycotoxins on the immune system and the consequences for pig health.
