The pathological implication of amyloid precursor protein(APP)in Alzheimer’s disease has been widely documented due to its involvement in the generation of amyloid-β ***,the physiological functions of APP are still poorly *** is considered a multimodal protein due to its role in a wide variety of processes,both in the embryo and in the adult ***,APP seems to play a key role in the proliferation,differentiation and maturation of neural stem *** addition,APP can be processed through two canonical processing pathways,generating different functionally active fragments:soluble APP-α,soluble APP-β,amyloid-β peptide and the APP intracellular c-terminal *** fragments also appear to modulate various functions in neural stem cells,including the processes of proliferation,neurogenesis,gliogenesis or cell ***,the molecular mechanisms involved in these effects are still *** this review,we summarize the physiological functions of APP and its main proteolytic derivatives in neural stem cells,as well as the possible signaling pathways that could be implicated in these *** knowledge of these functions and signaling pathways involved in the onset or during the development of Alzheimer’s disease is essential to advance the understanding of the pathogenesis of Alzheimer’s disease,and in the search for potential therapeutic targets.

The Arabidopsis XBAT35 is one of five structurally related ankyrin repeat-containing really Interesting New Gene （rING） E3 ligases involved in ubiquitin-mediated protein degradation, which plays key roles in a wide range of cel- lular processes. Here, we show that the XBAT35 gene undergoes alternative splicing, generating two transcripts that are constitutively expressed in all plant tissues. The two splice variants derive from an exon skipping event that excludes an in-frame segment from the XBAT35 precursor mrNA, giving rise to two protein isoforms that differ solely in the presence of a nuclear localization signal （NLS）. Transient expression assays indicate that the isoform lacking the NLS localizes in the cytoplasm of plant cells, whereas the other is targeted to the nucleus, accumulating in nuclear speckles. Both isoforms are functional E3 ligases, as assessed by in vitro ubiquitination assays. Two insertion mutant alleles and rNA-interference （rNAi） silencing lines for XBAT35 display no evident phenotypes under normal growth conditions, but exhibit hyper- sensitivity to the ethylene precursor 1-aminocyclopropane-l-carboxylate （Acc） during apical hook exaggeration in the dark, which is rescued by an inhibitor of ethylene perception. Independent expression of each XBAT35 splice variant in the mutant background indicates that the two isoforms may differentially contribute to apical hook formation but are both functional in this ethylene-mediated response. Thus, XBAT35 defines a novel player in ethylene signaling involved in negatively regulating apical hook curvature, with alternative splicing controlling dual targeting of this E3 ubiquitin ligase to the nuclear and cytoplasmic compartments.

Goat whey is an industry-discarded dairy by-product that has interesting nutritional value and a nutrient composition with important functional potential,where proteins have a high biological value and play an important role in the organism’s ***,the aim of this study was to identify,quantify,concentrate and investigate the capacity of goat whey proteins to perform different biological activities and to evaluate their acute *** proteins were quantified and identified by Bradford,Kjehdal and SDS-PAGE *** activity was performed against Listeria monocytogenes,Salmonella spp.,Staphylococcus aureus,Pseudomonas aeruginosa and Escherichia coli microorganisms and showed MIc ranging from 15 to 120μg mL-1,with the highest inhibition for *** *** activity was performed against human malignant melanoma cells and their proliferation inhibition was up to 66.8%within 72 h of *** highest antioxidant capacity was obtained at concentration 2 mg that showed 29.69%of inhibition of DPPH radical *** acute toxicity test was performed using Artemia franciscana larvae in which whey proteins were able to preserve larval survival throughout ***,we can conclude that goat’s whey has proteins that can play a positive effect on health,with antimicrobial activity and without *** present study highlights the great potential of goat’s whey to be developed as a safe biopreservative or as a functional ingredient in the food industry.

AIM To identify common copy number alterations on gastric cancer cell *** four gastric cancer cell lines(AcP02, AcP03, AGP01 and PG100) underwent chromosomal comparative genome hybridization and array comparative genome hybridization. We also confirmed the results by fluorescence in situ hybridization analysis using the bacterial artificial chromosome clone and quantitative real time Pcr *** The amplification of 9p13.3 was detected in all cell lines by both methodologies. An increase in the copy number of 9p13.3 was also confirmed by fluorescence in situ hybridization analysis. Moreover, the interleukin 11 receptor alpha(IL11rA) and maternal embryonic leucine zipper kinase(MELK) genes, which are present in the 9p13.3 amplicon, revealed gains of the MELK gene in all the cell lines studied. Additionally, a gain in the copy number of IL11 rA and MELK was observed in 19.1%(13/68) and 55.9%(38/68) of primary gastric adenocarcinoma samples, respectively. cONcLUSION The characterization of a small gain region at 9p13.3 in gastric cancer cell lines and primary gastric adenocarcinoma samples has revealed MELK as a candidate target gene that is possibly related to the development of gastric cancer.

Background:Identification of relevant in vivo biomarkers for fertility remains a challenge for the livestock *** of the small peptide hormone oxytocin(OXT),involved in male reproductive function and present in the seminal plasma(SP)of several species could be a robust *** study characterized concentrations of SPOXT in ejaculates from boars used in artificial insemination(AI)programs aiming to evaluate its relationship with sperm quality variables and in vivo fertility of their liquid-stored *** OXT concentrations(ng/mL)were measured in 169 ejaculates from 61 boars of the Duroc,Pietrain,Landrace and Large White breeds using a direct competitive immunoassay test based on AlphaLISA®***(ejaculate volume,sperm concentration,total sperm count)and sperm parameters(motility,viability,intracellular generation of reactive oxygen species,plasma membrane fluidity)were assessed at 0 h and 72 h in AI-semen samples stored at 17℃.In vivo fertility included only 18 Large White and Landrace boars whose AI-semen was used to inseminated>100 sows and evaluated both farrowing rate and litter size of 3,167 ***:The results showed that SP-OXT differed between boars and between ejaculates within boar(Preeds(Duroc,Pietrain,Landrace and Large White).Ejaculates with higher SP-OXT concentration/mL(hierarchically grouped;Prger volume and came from younger boars(Pculates of boars showing positive farrowing rate deviation exhibited higher(Pconcentration/mL than those with negative farrowing rate ***:The SP concentrations of OXT are boar,ejaculate and age dependent,and positively related with ejaculate volume and farrowing rates of liquid-stored semen AI-doses.

Microbial electrosynthesis(MES) can potentially provide a mean for storing renewable energy surpluses as chemical energy. However, the fluctuating nature of these energy sources may represent a threat to MES, as the microbial communities that develop on the biocathode rely on the continuous existence of a polarized electrode. This work assesses how MES performance, product generation and microbial community evolution are affected by a long-period(6 weeks) power off(open circuit). Acetogenic and H2-producing bacteria activity recovered after reconnection. However, few days later syntrophic acetate oxidation bacteria and H2-consuming methanogens became dominant, producing cH4 as the main product, via electromethanogenesis and the syntrophic interaction between eubacterial and archaeal communities which consume both the acetic acid and the hydrogen present in the cathode environment. Thus,the system proved to be resilient to a long-term power interruption in terms of electroactivity. At the same time, these results demonstrated that the system could be extensively affected in both end product generation and microbial communities.

Spacesuits and spacecraft must endure high velocity impacts from micrometeoroids. This work considers the impact of 100 μm diameter projectiles into composite targets at velocities from 0.5 km/s to 2 km/*** work begins by presenting an energy-based theoretical model relating depth of penetration(Do P)and impact force to impact velocity, characteristic time, and threshold velocity and force. Next, this work compares numerical simulations of normal impact on composites to the theoretical model. Numerical simulations are conducted with LS-DYNA and the well-known composite model, MAT-162. The numerical models consider unidirectional S2-glass fiber reinforced Sc-15 epoxy composite laminates. The numerical model shows good correlation with the theoretical model. The numerical model also investigates lateral impact, parallel to the fiber direction, and oblique impact at angles from 30°to 82.5°.This work decomposes oblique impact into normal and lateral components, and compares them with normal and lateral impact results. The results show good correlation of the normal component of oblique results with the theoretical model. This numerical and theoretical study focuses on Do P, velocity, and penetration resistance force as functions of time. The theoretical model and numerical simulations are used to determine new Do P parameters: characteristic time of depth of penetration and threshold impact velocity. These models are a first step in developing the capability to predict Do P for oblique,microscale, high-speed impact on composite materials.

Background： Sperm DNA fragmentation（sDf） has been proved to be an important parameter in order to predict in vitro the potential fertility of a semen sample. colloid centrifugation could be a suitable technique to select those donkey sperm more resistant to DNA fragmentation after thawing. Previous studies have shown that to elucidate the latent damage of the DNA molecule, sDf should be assessed dynamically, where the rate of fragmentation between treatments indicates how resistant the DNA is to iatrogenic damage. The rate of fragmentation is calculated using the slope of a linear regression equation. However, it has not been studied if s Df dynamics fit this model. The objectives of this study were to evaluate the effect of different after-thawing centrifugation protocols on sperm DNA fragmentation and elucidate the most accurate mathematical model（linear regression, exponential or polynomial） for DNA fragmentation over time in frozen-thawed donkey ***： After submitting post-thaw semen samples to no centrifugation（UDc）, sperm washing（SW） or single layer centrifugation（SLc） protocols, sD f values after 6 h of incubation were significantly lower in SLc samples than in SW or *** of determination（r-2） values were significantly higher for a second order polynomial model than for linear or exponential. The highest values for acceleration of fragmentation（aSDf） were obtained for SW, fol owed by SLc and ***： SLc after thawing seems to preserve longer DNA longevity in comparison to UDc and SW. Moreover,the fine-tuning of models has shown that sDf dynamics in frozen-thawed donkey semen fit a second order polynomial model, which implies that fragmentation rate is not constant and fragmentation acceleration must be taken into account to elucidate hidden damage in the DNA molecule.

Leaf wetness duration(LWD)is a critical parameter used to predict plant disease,but its determination under actual field conditions is a major *** this study,a method for determining LWD using thermal infrared imaging was developed and applied to cucumber plants grown in a solar *** images of the plant leaves were captured using an infrared scanning camera,and a leaf wetness area segmentation method consisting of two procedures was ***,a color space conversion was performed automatically by an image-processing ***,the K-means clustering algorithm was applied to enable the segmentation of the wetness area on the thermal ***,to enable overall thermal image analysis,an initial leaf wetness threshold(LWT)of 5%was defined(where wetness values higher than 5%indicated that the leaf was in a wet state).The results of comparative experiments conducted using thermal images of plant leaves captured using an infrared scanning camera and human visual observation indicated that the estimated LWD values were generally higher than the observed LWD values,because slight leaf wetness condensations were overlooked by the human eye but detected by the infrared scanning *** these differences were not found to be statistically significant in this study,the proposed method for determining LWD using thermal infrared imaging may provide a new LWD detection method for cucumber and other plants grown in solar greenhouses.

Effect of age on visual and refractive results after LASIK: mechanical microkeratome versus femtosecond laser

AIM: To evaluate the effect of age on visual and refractive results after laser in situ keratomileusis(LASIK) obtained with a mechanical microkeratome or a femtosecond ***: retrospective, nonrandomized, cohort study. A total of 3826 eyes were included in the study(1725 eyes treated with mechanical LASIK and 2101 eyes treated with femtosecond LASIK). The relationship between patient age and the 3-month postoperative visual and refractive results of both procedures were analyzed by linear regression ***: Three months postoperatively, we found a significant correlation between age and the postoperative spherical equivalent(SE; r2=0.004, P=0.006), efficacy(r2=0.006, P=0.001), and safety indexes(r2=0.05, P=0.0001) in the mechanical LASIK group. On the other hand, we found a significant correlation between age and the postoperative SE(r2=0.02, P=0.0001) and the efficacy index(r2=0.01, P=0.0001) but not the safety index in the femtosecond laser group. Mechanical LASIK provided slightly but significantly better efficacy and predictability in patients 18 to 40 years of age and femtosecond LASIK did so in patients older than 40 years of age. The femtosecond laser provided better safety results than the mechanical microkeratome in both age groups. cONcLUSION: A tendency toward undercorrection and less predictability is found with aging after myopic LASIK regardless of whether the flap was created with a mechanical microkeratome or a femtosecond laser. However, femtosecond laser provides significantly betteroutcomes in terms of efficacy, safety and predictability compared to mechanical microkeratome for the correction of myopia in patients over 40 y.
