Identification of optimal contemporary antiemetic prophylaxis for doxorubicin-cyclophosphamide chemotherapy in Chinese cancer patients: post-hoc analysis of 3 prospective studies

Objective:Chemotherapy-induced nausea and vomiting(CINV)are common with doxorubicin-cyclophosphamide(AC)*** antiemetic regimens incorporate neurokinin-1 receptor antagonist(NK1 RA),5-hydroxytryptamine type-3 receptor antagonist(5 hT3 RA),corticosteroid,and dopamine *** post-hoc analysis compared results of 3 prospective antiemetic studies conducted among Chinese breast cancer patients who received(neo)adjuvant AC,in order to identify optimal antiemetic ***:A total of 304 patients were included:Group 1,ondansetron/dexamethasone(D1);Group 2,aprepitant/ondansetron/dexamethasone(D1);Group 3,aprepitant/ondansetron/dexamethasone(D1–3);Group 4,aprepitant/ondansetron/dexamethasone(D1–3)/olanzapine;and Group 5,netupitant/palonosetron/dexamethasone(D1–3).Antiemetic efficacies of Groups 3,4,and 5 during cycle 1 of AC were individually compared with Group *** addition,emesis outcomes of patients in Groups 3 and 5,and those of Groups 2 and 3,were ***:When comparing efficacies of a historical doublet(5 hT3 RA/dexamethasone)with triplet antiemetic regimens(NK1 RA/5 hT3 RA/dexamethasone)with/without olanzapine,complete response(CR)percentages and quality of life(QOL)in overall phase of cycle 1 AC were compared between Group 1 and the other groups:Group 1 vs.3,41.9%vs.38.3%(P=0.6849);Group 1 vs.4,41.9%vs.65.0%(P=0.0107);and Group 1 vs.5,41.9%vs.60.0%(P=0.0460).Groups 4 and 5 achieved a better *** comparing netupitant-based(Group 3)with aprepitant-based(Group 5)triplet antiemetics,CR percentages were 38.3%vs.60.0%,respectively(P=0.0176);Group 5 achieved a better *** comparing 1 day(Group 2)vs.3 day(Group 3)dexamethasone,CR percentages were 46.8%and 38.3%,respectively(P=0.3459);Group 3 had a worse ***:Aprepitant-containing triplets were non-superior to doublet ***-containing triplets and adding olanzapine to aprepitant-containing triplets were superior to ***/palono

Proteomics and Metabolomics of Arabidopsis Responses to Perturbation of Glucosinolate Biosynthesis

To understand plant molecular networks of glucosinolate metabolism, perturbation of aliphatic glucosinolate biosynthesis was established using inducible RNA interference （RNAi） in Arabidopsis. Two RNAi lines were chosen for examining global protein and metabolite changes using complementary proteomics and metabolomics approaches. Pro- teins involved in metabolism including photosynthesis and hormone metabolism, protein binding, energy, stress, and defense showed marked responses to glucosinolate perturbation. In parallel, metabolomics revealed major changes in the levels of amino acids, carbohydrates, peptides, and hormones. The metabolomics data were correlated with the pro- teomics results and revealed intimate molecular connections between cellular pathways/processes and glucosinolate me- tabolism. This study has provided an unprecedented view of the molecular networks of glucosinolate metabolism and laid a foundation towards rationale glucosinolate engineering for enhanced defense and quality.

Ehlers-Danlos syndrome:prevalence and outcomes in gender affirming surgery-a single institution experience

Aim:Patients with Ehlers-Danlos Syndrome(EDS)are considered to have an increased risk for wound healing *** may therefore be hesitant to offer elective surgeries,including gender-affirming surgeries(GAS),to EDS *** our center,we frequently encountered patients presenting for GAS evaluation with the co-existing diagnosis of *** study aims to establish the prevalence of EDS diagnosis in our GAS patients and compare their post-operative complications to patients without EDS ***:This is a single-institution retrospective case-control study on all patients who underwent GAS from *** include EDS diagnosis,demographics,operation,and complications(including minor wound healing issues).Results:Of 1363 patients presenting for GAS,36(2.6%)had EDS diagnoses and were matched with 108 control *** complications requiring surgical intervention in the OR occurred in 6 patients(4.2%),(2.8% EDS vs.5.4%controls;P=0.63),while 8.3% of EDS and 14% of controls required minor interventions(P=0.38).The rate of wound healing issues of any severity was 28% in EDS vs.47% in control groups(P=0.04).Conclusion:The prevalence of EDS diagnosis in our patient population is 132 times the highest reported prevalence in the general *** healing issues and the need for additional post-operative interventions in the group with EDS diagnosis were not significantly different from the control *** findings suggest that patients with a diagnosis of EDS undergoing GAS have comparable outcomes to patients without *** for post-operative complications should not be a barrier to offering GAS to patients presenting with an EDS diagnosis.

Comparative transcriptomics reveals hidden issues in the plant response to arthropod herbivores

Plants experience different abiotic/biotic stresses,which trigger their molecular machinery to cope with them. Besides general mechanisms prompted by many stresses, specific mechanisms have been introduced to optimize the response to individual threats. however, these key mechanisms are difficult to identify. here, we introduce an in-depth species-specific transcriptomic analysis and conduct an extensive meta-analysis of the responses to related species to gain more knowledge about plant responses. The spider mite Tetranychus urticae was used as the individual species, several arthropod herbivores as the related species for meta-analysis, and Arabidopsis thaliana plants as the common host. The analysis of the transcriptomic data showed typical common responses to herbivory, such as jasmonate signaling or glucosinolate biosynthesis. Also, a specific set of genes likely involved in the particularities of the Arabidopsis-spider mite interaction was discovered. The new findings have determined a prominent role in this interaction of the jasmonateinduced pathways leading to the biosynthesis of anthocyanins and tocopherols. Therefore, tandem individual/general transcriptomic profiling has been revealed as an effective method to identify novel relevant processes and specificities in the plant response to environmental stresses.

Background:Extensive loss of donor neural stem cell(NSCs)due to ischemic stress and low rate of differentiation at the site of cell graft are two of the major issues that hamper optimal outcome in NSCs transplantation *** that histone deacetylases(hDACs)modulate various cellular processes by deacetylating histones and non-histone proteins,we hypothesized that combined treatment with small molecules,sodium butyrate(NaB;a known hDAC inhibitor)and nicorandil,will enhance the rate neuronal differentiation of NSCs besides their preconditioning to resist oxidative ***:NSCs derived from 14-day old Sprague Dawley rat ganglion eminence were characterized for tri-lineage *** with 1 mM NaB significantly changed their culture characteristics while continuous treatment for 10 days enhanced their neural *** treatment also preconditioned the cells for their resistance to oxidative ***:The highest rate of neural differentiation and preconditioning effect was achieved when the NSCs were treated concomitantly with NaB and *** proliferation assay showed that concomitant treatment with NaB and nicorandil retarded their rate of ***:These data conclude that preconditioning of NSCs with NaB and nicorandil effectively enhances their differentiation capacity besides preconditioning the cells to support their survival under ischemic conditions.

Wereport on the colloidal and rheological characterization of suspensions of commercial single-walled carbon nanotubes(SWCNT)and oxidized single walled carbon nanotubes(SWCNT-oxi24h),in water and fetal bovine serum(FBS).The effects of two surfactants,dodecyl-benzene sodium sulfonate(SBDS)and sodium dodecyl sulfate(SDS)at different concentrations(0.5-2.0%)on the zeta potential and the rheology of the suspensions is also *** suspensions were characterized by Transmission Electron Microscopy(TEM)and Atomic Force Microscopy(AFM).The colloidal stability was determined by measurements of zeta potential in the physiological ph range 6.0hhe results obtained show that SWCNT suspensions in FBS,with and without addition of surfactants,depict a high tendency to aggregate with the bovine serum albumin(BSA),a processwe could further characterize using Molecular Dynamics(MD)*** suspensions in aqueous media show a negative zeta potential between−60 and−80 mV,whereas in FBS the stability is lower,with values of-20 *** observed no significant changes in the zeta potential with the different *** viscosity values obtained are smaller in water than in FBS,demonstrating the higher stability in water.

GSTM3, but not IZUMO1, is a cryotolerance marker of boar sperm

Background: Cryopreservation is currently the most efficient method for long-term preservation of mammalian gametes and is extensively used in swine artificial insemination(AI) centres. however, it is well-known that cryopreservation procedures induce changes in the water phase in both intra and extracellular compartments,which alter the content and localisation of several proteins and ends up curtailing the structural integrity of functional sperm(i.e., cryoinjuries). Alterations and deficiencies of sperm-oocyte binding proteins during gamete recognition are one of the causes of reproductive failure both in vitro and in vivo. In this sense, characterisation of cryopreservation effects upon oocyte-binding proteins of sperm, such as IZUMO1 and GSTM3, is essential when assessing the impact of this technique in swine ***: Cryopreservation was found to induce changes in the localisation of IZUMO1 and GSTM3 in boar ***, the relative content of both proteins was not altered after thawing. Furthermore, whereas IZUMO1 content was found not to be related to the cryotolerance of boar sperm, GSTM3 content was observed to be higher in poor(PFE) than in good(GFE) freezability ejaculates in both pre-frozen(1.00 INT·mm^2± 0.14 INT·mm^2 vs.0.72 INT·mm^2± 0.15 INT·mm^2;P hawed(0.96 INT·mm^2± 0.20 INT·mm^2 vs. 70 INT·mm^2± 0.19 INT·mm^2;P higher in those spermatozoa that exhibited low mitochondrial activity, high reactive oxygen species(ROS) production, and high membrane lipid disorder postthaw(P he difference in GSTM3 content between GFE and PFE, together with this protein having been found to be related to poor sperm quality post-thaw, suggests that it could be used as a cryotolerance marker of boar spermatozoa. Early life investment in antlers and body growth reflects adult performance in roe deer population under supplementary feeding conditions

Recent research has challenged the idea that cervid antlers are such costly traits,supporting the assertion early-life antler investment is an honest signal of adult phenotypic ***,inferences were made based on antler measurements while growing(velvet)and thus,studies on fully-formed clean antlers are needed to avoid possible bias caused by the inter-individual variation in antler growth *** studied a semi-captive population of European roe deer inhabiting a sub-Mediterranean area(Valsemana research station)and living under optimal conditions(ad libitum food supply and veterinary care).Based on repeated measurements taken from 146 individuals,we assessed whether allocation to secondary sexual traits during early life affected body mass or antler development during ***,we evaluated which body measurements better predicted future male ***,using 488 individuals,we described age-class-specific variation in male body measurements and assessed the decline in antler size between adult and senescent stages(*** of senescence).Results agree with the assertion that there is no negative effect of a high investment in antler during early life on adult antler size or body mass,but we suggest the use of clean antlers as a more robust and reliable *** variables that better predicted body mass during adulthood were yearling body mass and body size at 6 *** score between 10 and 18 months resulted in the best indicator of adult antler ***,we support the idea that the magnitude of senescence in antler size is smaller in environments with higher resource availability during winter.
