Multipotent stromal cells stimulate liver regeneration by influencing the macrophage polarization in rat

AIM To investigate the influence of the umbilical cordderived multipotent stromal cells(MSCs) on recovery of the liver after the subtotal resection, that is, removal of 80% of the organ mass, a renowned model of the small-for-size liver remnant syndrome. METHODS The MSCs were obtained from the intervascular tissue of umbilical cords, dissected from rat fetuses, by the explant culture technique. The vital labeling of MSCs with РКН26 was carried out on the 3 rd passage. The subtotal resection was performed on male Sprague-Dawley rats. The experimental group animals received a transplant 106 MSCs infused into the spleen. Hepatocyte proliferation was assessed by counting of either mitotic figures or Ki67-positive cells in microscopic images. MSC differentiation was assessed with antibodies to hepatocyte-specific marker cytokeratin 18(CK18), cholangiocyte-specific protein CK19, smooth muscle cell-specific protein α-SMA, the endothelial cell marker CD31, or the active fibroblast marker FAPα. Total macrophages of the liver were selectively stained in cryosections incubated with antiCD68 antibodies(1:100, Abcam), while the M2 a and M2 c macrophage populations were selectively stained with anti-CD206 antibodies. Expression of interleukin and growth factor genes was evaluated with PCR-RT.RESULTS Intrasplenic allogeneic transplantation of the umbilical cord-derived multipotent stromal cells stimulates reparative processes within the residual liver tissue after subtotal resection(removal of 80% of the organ mass), as indicated by increased rates of hepatocyte proliferation and accelerated organ mass recovery. These effects may result from paracrine influence of the transplanted cells on the resident macrophage population of the liver. The transplantation favors polarization of macrophages to M2 phenotype(the M2-polarized macrophages specifically express CD206; they are known to suppress inflammation and support tissue repair). Shock-wave impact on the knee joint affected with osteoarthritis and after arthroplasty

Degenerative diseases significantly reduce the quality of human life.Non-invasive treatments are used in the initial stages of osteoarthritis(OA).Total knee arthroplasty is used in the late stages of osteoarthritis of the knee joint.Non-invasive methods based on mechanical action are also used for the rehabilitation of a patient after arthroplasty.This paper presents numerical models of the knee joint with degenerative OA changes and arthroplasty.Using these models,a computational study was made of the influence of the intensity of shock-wave exposure on the conditioning for the regeneration of bone and cartilage tissues.Based on the modeling results,it was found that in the knee joint with degenerative OA changes,conditions for the regeneration of cartilage and meniscus tissues were fulfilled under medium and highintensity loading.Under high-intensity loading(up to 0.9 m J/mm^(2)),the stress level was significantly below the ultimate value required for fracture.At knee arthroplasty,the conditions for bone tissue regeneration around the tibia component are fulfilled only under high-intensity loading.

TNF-α gene polymorphisms: association with age-related macular degeneration in Russian population

AIM: To study polymorphisms in promotor regions of tumor necrosis factor(TNF)-α TNF-863 A/C(rs1800630), TNF-308 A/G(rs1800629), and TNF-238 A/G(rs361525) in patients with age-related macular degeneration(AMD) and associations of complex TNF-α genotypes with AMD. METHODS: One hundred and two patients(82 women, 20 men; mean age 64.2±1.2 y) with AMD and 100 healthy age-and sex-matched controls(82 women, 18 men; 60±1.4 y) were included in the study. All subjects were Caucasian, all subjects and their parents were inhabitants of Russia. Genomic DNA was obtained from EDTA-preserved blood using the standard phenol-chloroform method. Polymorphisms were detected by polymerase chain reaction followed by the restriction fragment length polymorphism method. The following TNF-α genotypes were studied: TNF-α-238 AA, GA, GG, TNF-α-308 AA, GA, GG, TNF-α-863 AA, CA, CC. RESULTS: Differences in TNF-α-863 and TNF-α-238 genotypes frequencies in patients with AMD and healthy controls were not found. The distribution of TNF-α-308 AA and TNF-α-308 GA genotypes was significantly different between the studied group and the controls [odds ratios(OR) =0.22, P=0.0287 and OR=2.91, P=0.0063, respectively]. TNF-863 CC/TNF-308 GA and TNF-308 GA/TNF-238 GG genotypes were associated with the increased risk of AMD(OR=2.48, P=0.0332 and OR=2.51, P=0.0187, respectively). Five genotypes combinations appeared to be protective. CONCLUSION: In the present study, single nucleotide polymorphisms and complex polymorphisms of one of the key inflammatory cytokines TNF-α, and a number of significant associations of these polymorphisms with AMD in Russian population have been shown. Complexanalysis of genotypes could be important in AMD risk factors detection and studying pathogenesis.

Radical Right Parties in the Political System of States： Theoretical and Practical Aspects

The article deals with the question of whether far-right parties are able to influence the political and party system in the contemporary state. Far-right parties have acquired special political features that are grounded on a broad variety of far-right ideological norms and political instruments. Far-right parties challenge the state government and policy offering the key (anti-immigration politics) to solve a variety of complicated issues. The main idea of the article is that far-right parties are flexible enough in their political rhetoric and instruments to gain electoral support. Based on different concepts and theories we should underline that far-right parties have a prominent electoral future but their place in the political arena greatly depends on existing conflicts of social and cultural nature in the nation-state.

Understanding the mechanisms of friction stir welding based on computer simulation using particles

Friction stir welding(FSW) is a novel technique for joining different materials without melting. In FSW the welded components are joined by stirring the plasticized material of the welded edges with a special rotating pin plunged into the material and moving along the joint line. From the scientific point of view,the key role of the FSW processes belongs to formation of the special plasticized conditions and activation of physical mechanisms of mixing the materials in such conditions to produce the strong homogeneous weld. But it is still a lack of complete understanding of what are these conditions and mechanisms.This paper is devoted to understanding the mechanisms of material mixing in conditions of FSW based on a computer simulation using particles. The movable cellular automaton method(MCA), which is a representative of the particle methods in mechanics of materials, was used to perform all computations.Usually, material flow including material stirring in FSW is simulated using computational fluid mechanics or smoothed particle hydrodynamics, which assume that the material is a continuum and does not take into account the material structure. MCA considers a material as an ensemble of bonded particles. Breaking of inter-particle bonds and formation of new bonds enables simulation of crack nucleation and healing, as well as mass mixing and micro-welding.The paper consists of two main parts. In the first part, the simulations in 2 D statements are performed to study the dynamics of friction stir welding of duralumin plates and influence of different welding regimes on the features of the material stirring and temperature distribution in the forming welded joints. It is shown that the ratio of the rotational speed to the advancing velocity of the tool has a dramatic effect on the joint quality. A suitable choice of these parameters combined with additional ultrasonic impact could considerably reduce the number of pores and microcracks in the weld without significant overheating of the welded materials.The secon

The oscillations in ESR spectra of Hg_(0.76)Cd_(0.24)Te implanted by Ag^+ at the X and Q-bands

The objects of the investigation were uniformly Ag~＋ doped Hg_（0.76）Cd_（0.24） Te mercury chalcogenide monocrystals obtained by ion implantation with subsequent thermal annealing over 20 days. After implantation and annealing the conductivity was inverted from n-type with carrier concentration of 10^（16） cm^（-3） to p-type with carrier concentration of ≈ 3.9 × 10^（15） cm^（-3）. The investigations of microwave absorption derivative（d P/d H） showed the existence of strong oscillations in the magnetic field for Ag：Hg_（0.76）Cd_（0.24） Te in the temperature range 4.2–12 K. The concentration and effective mass of charge carrier were determined from oscillation period and temperature dependency of oscillation amplitude. We suppose that this phenomenon is similar to the de Haas–van Alphen effect in weakly correlated electron system with imperfect nesting vector.

We report differential measurements of Ar+ ion momentum distributions from nonsequential double ionization in phase-stabilized few-cycle laser pulses.The distributions depend strongly on the carrierenvelope(CE)phase.Via control over the CE phase one is able to direct the nonsequential doubleionization dynamics.Data analysis through a classical model calculation reveals that the influence of the optical phase enters via(i)the cycle dependent electric field ionization rate,(ii)the electron recoilision time,and(iii)the accessible phase space for inelastic collisions.Our model indicates that the combination of these effects allows a look into single cycle dynamics already for few-cycle pulses.
