Mediator contributes to IgH locus VDJ rearrangements by promoting usage of most distal V segments

INTRODUCTION The immunoglobulin heavy(IgH)chain locus includes a large cluster of V,D and J segments spanning thousands of kilobases in mammals and its expression implies long-range 3D-regulation of various gene modifications.1 V(D)J recombination occurs during early B-cell ontogeny.2 It is regulated by multiple transcription factors and cis-regulatory elements,and most notably by the intronic 5′eμelement located in the JH to Cμintron.3 The intergenic control region 1(IGCR1)located between the VH and D gene clusters inhibits rearrangement of the most DH-proximal VH gene segments,while promoting usage of distal VH gene segments.4 Other downstream elements of the locus,flanking its 3′end might have more subtle roles:while the large 3′regulatory region(3′RR)shows strong internal synergies2 and behaves as a superenhancer in mature B *** deletion showed no influence on the VDJ repertoire and rather increased transcription of unrearranged V segments.3–5 Insulators binding CTCF farther downstream of the 3′RR were shown,however,to promote usage of distal VH segments.6 Altogether,there are strong indications that elements involved in long-distance regulation of transcription might also influence VDJ ***-frame splicing of VDJ exons is also needed for full Ig expression.7 Given the functional importance of gene architecture and long-range interactions for the IgH locus physiology,we explored the dependence of the VDJ repertoire on Med1,a known actor of long-range enhancer interactions.

Background:In vivo cell tracking after transplantation in regenerative medicine remains an unmet challenge and limits current understanding of the wound healing mechanism through cell-based *** study investigated tracking of human Wharton’s jelly stem cells(hWJSCs)seeded onto an acellular dermal matrix(ADM)and labeled with superparamagnetic iron oxide nanoparti-cles(SPIONs)by magnetic resonance imaging(MRI)in burn ***:The hWJSCs were characterized and assessed for growth kinetics.A total of 30 rats were enrolled in three equal *** 1 underwent scald burn injury left without treatment,the group 2 was treated by an ADM that was prepared from cosmetic surgery skin samples and the group 3 received hWJSCs labeled with SPIONs seeded onto an *** strength was evaluated before and after interventions,real time PCR assessed apoptosis,and Prussian blue staining,scanning electron microscopy(SeM)and MRI were used for the tracking of labeled ***:The hWJSCs exhibited mesenchymal stem cell *** doubling time was 40.1 *** did not show any toxic *** hWJSCs seeded onto an ADM decreased Bax and increased Bcl-2 gene *** of SPIONs within hWJSCs was confirmed by Prussian blue staining,SeM and MRI until day *** was a significant difference between the Young’s moduli of normal skin and the group receiving hWJSCs seeded onto an *** observations and SeM imaging confirmed that MRI is an accurate method to track SPION-labeled hWJSCs in ***:This study showed that SPION labeling coupled with MRI can be used to further understand the fate of stem cells after transplantation in a burn model.

Management of acute esophageal variceal bleeding by endoscopic sclerotherapy in technically difficult endoscopic band ligation cases—A population based cohort study

endoscopic band ligation is regarded as the main therapeutic option for acute esophageal variceal bleeding, while sclerotherapy may be used in the acute setting if ligation is technically difficult. The incidence of difficult-to-perform band ligation in acute esophageal variceal bleeding, as well as the outcome of patients subjected to injection sclerotherapy as an alternative treatment, has not been clearly investigated. Our aim is to study the outcome of patients subjected to injection sclerotherapy in the acute setting of esophageal variceal bleeding when endoscopic band ligation is technically difficult to perform. We included 151 patients with acute esophageal variceal bleeding originnating from medium or large sized varices. All patients were planned for eBL as the 1st treatment option (eBL group 61.6%), meanwhile, eIS using 5% ethanolamine oleate was reserved as the 2nd treatment option when eBL was technically difficult (eIS group 38.4%). The mean time to restore hemodynamic stability was significantly prolonged in the eIS group (11.5 ± 6.5 hrs versus 9.5 ± 5.0 hrs, p 0.05). Initial control of bleeding was significantly higher in the eBL group versus the eIS group (96.7% vs 84.5%, p 0.021). Re-bleeding was more among the eIS group (42.9% vs 24.2%, p 0.04). There were no significant differences as regarding mortality and duration of hospital stay. So, a considerable proportion of cases presenting with acute variceal bleeding will have difficulty in performing eBL. In these patients, sclerotherapy is not a waning procedure with an accepted success rate, without much additional complications and without deranging mortality.

AIM:To identify blood donors with occult hepatitis B virus(HBV) infection(OBI) to promote safe blood ***:Descriptive cross sectional study was conducted on 3167 blood donors negative for hepatitis B surface antigen(HBsAg),hepatitis C antibody(HCV Ab) and human immunodeficiency virus *** were subjected to the detection of alanine aminotransferase(ALT) and aspartate transaminase(AST) and screening for anti-HBV core antibodies(total) by two different techniques;[Monoliza antibodies to hepatitis B core(Anti-HBc) Plus-Bio-Rad] and(ARC-HBc total-ABBOT).Positive samples were subjected to quantitative detection of antibodies to hepatitis B surface(anti-HBs)(eTI-AB-AUK-3,Dia Sorin-Italy).Serum anti-HBs titers > 10 IU/L was considered *** HBV DNA by real time polymerase chain reaction(PCR)(QIAGeN-Germany) with 3.8 IU/mL detection limit was estimated for blood units with negative serum anti-HBs and also for 32 whose anti-HBs serum titers were > 1000 IU/***,265 recipients were included,34 of whom were followed up for 3-6 *** were investigated for ALT and AST,HBV serological markers:HBsAg(eTI-MAK-4,Dia Sorin-Italy),anti-HBc,quantitative detection of anti-HBs and ***:525/3167(16.6%) of blood units were positive for total anti-HBc,64% of those were antiHBs *** by ARCHITeCT anti-HBc assay were carried out for 498/525 anti-HBc positive samples,where 451(90.6%) confirmed *** for anti-HBc was considered confirmed only if two positive results were obtained for each sample,giving an overall prevalence of 451/3167(14.2%) for total *** DNA was quantified by real time PCR in 52/303(17.2%) of anti-HBc positive blood donors(viral load range:5 to 3.5 x 105 IU/mL) with a median of 200 IU/mL(mean:1.8 x 104 ± 5.1 x 104 IU/mL).AntiHBc was the only marker in 68.6% of *** and multivariate logistic analysis for identifying risk factors associated with anti-HBc and HBV-DNA positivity a

Review on the estimating the effective Way for Managing the Produced Water: Case Study

Water manufactured is the primary waste source in the oil and gas industry. Because of the rising amount of waste worldwide, the environmental effect of wastewater has become a primary environmental concern in recent years. The vast amounts involved have resulted in considerable costs to the industry for handling produced water. This research explains the wide variety of choices for water management. This research’s first phase was water minimization techniques, consisting of three different applications made in three different wells (Well 1, Well 2 and Well 3) and water recycling and reuse by two techniques. In Well 1, Mechanical shut-off technique was applied using through tubing bridge plug and 5 m cement dumped above it to isolate the watered out zone;as per water oil ration plot the water cut is decreased from 100% to 4% and the production is increased from 0 to 400 bcpd. In Well 2, Chemical shut-off technique using a polymer called Brightwater has been used to block channeling through high permeability intervals after PLT log detected it, and the result was brilliant, the water cut decreased from 60% to 25%, also the oil production increase from 500 to 3000 bopd. In Well 3, downhole separator installed in it using workover (unfortunately, this technique is not applied in middle east till the moment so this application is taken from an oil field in Canada)and the result was perfect, the water cut decreased from 70% to 28%, also the oil production increase from 44 to 100 bopd. This study tried to clarify and compare the most widely used water management techniques using one of the Western Desert (W.D.) (enhanced for oil recovery, constructed wetland).

The idea of application from DNA information organization for providing the next generation super-computers has been proposed by lessons learned from nature. Thus, molecular environment of reserving information was considered as succeeding generation of a mother-board project containing some subcategories with various applications. MeRI, constructed based on the binary coding system using DNA synthesizer machine, enables retrievable reading of molecular structures using DNA sequencer machine. It has high capacity for reservation of information about 1.212 × 1011 Kb/cm2 with long-term stability. It can be used in various fields of science and technology regarding its high capacity, small size and safety measures.

Analysis of extreme Precipitation events over Central Plateau of Iran

This paper describes the results of an analysis of extreme rainfall events in the central plateau of Iran. To study the extreme events, daily records of eighteen stations’ rainfalls in the region for different initial dates up to 2005 gathered from the bureau of meteorology. Then, the extreme rainfall threshold was calculated for each individual station using the statistical index of Gamble type I. Lastly, 22 mm was determined as the extreme rainfall value for the entire stations, and eventually 17 out of 169 extreme precipitation events were extracted in accordance with three factors including a) days with precipitation in not less than 50% of the stations, b) maximum rainfall is 22 mm or more in at least one of the stations, and c) mean precipitation of the basin is more than 3 mm. In the next step to analyze the synoptic features, the relevant meteorological data i.e. relative vorticity, geopotential height, sea level pressure, u and v wind components, relative humidity, vertical velocity, and precipitable water content at multiple levels of the atmosphere were examined from the NCeP/NCAR reanalysis dataset. The synoptic findings indicate that two patterns of deep trough and high ridge of the eastern Mediterranean were responsible for making the heavy precipitation events over the central plateau of Iran. The most and severest rainfall events occurred via deep tough pattern, which covered 76% of days with extreme precipitations during the examined period. Furthermore, the results suggest that the main moisture resources, which identified by HYSPLIT model’s outputs and moisture convergence/divergence zones for the rainy systems in the first pattern (deep trough) including Persian Gulf, Oman Sea, Indian Ocean, and Red Sea, while for the second pattern (high ridge) Persian Gulf and Red Sea play a significant role in feeding the storms in the central regions of Iran. With polyamide(PA)as an efficient sorbent for solid phase extraction(SPe)of Sudan dyes II,III and Red 7B from saffron and urine,their determination by HPLC was *** optimum conditions for SPe were achieved using 7 mL methanol/water(1∶9,v/v,pH 7)as the washing solvent and 3 mL tetrahydrofuran for *** clean-up and high(above 90%)recoveries were observed for all the *** optimized mobile phase composition for HPLC analysis of these compounds was methanol-water(70∶30,v/v).The SPe parameters,such as the maximum loading capacity and breakthrough volume,were also determined for each *** limits of detection(LODs),limits of quantification(LOQs),linear ranges and recoveries for the analytes were 4.6-6.6μg/L,13.0-19.8μg/L,13.0-5 000μg/L(r2>0.99)and 92.5%-113.4%,*** precisions(RSDs)of the overall analytical procedure,estimated by five replicate measurements for Sudan II,III and Red 7B in saffron and urine samples were 2.3%,1.8%and 3.6%,*** developed method is simple and successful in the application to the determination of Sudan dyes in saffron and urine samples with HPLC coupled with UV detection.
