Trypanosomosis is major drawback to profitable livestock production in sub-Sahara African, including.Nig.ria. Knowledg. of the cytokines production in the phase of natural infection may help to better diag.ose, treat and prevent bovine trypanosomosis. The purpose of the this study was to determine the levels of interferon-g.mma (IFN-γ), interleukin-10 (IL-10) and cardiac troponin–I (cTnI) in the sera of cattle naturally infected with T. brucei, T. cong.lense and T. vivax and correlate these levels with parasitaemia and PCV of the infected animals. Five milliliter of blood samples were collected via the jug.lar vein from 411 randomly selected cattle into EDTA and non-citrated bottle. PCV was determined manually using.HCT. Trypansomes were detected and characterized by microscopy and PCR, respectively. Serum levels of IFN-γ, IL-10 and cTnI were determined using.commercial ELISA kit. Data were summarized using.descriptive statistic and sig.ificance of differences determined by ANOVA. Of the 62 samples positive for trypanosomes by microscopy, 50 samples were confirmed to species level by PCR. The sera levels of IFN-γ, IL-10 and cTnI of infected cattle were hig.er than non-infected cattle. The differences were not sig.ificant (p γ, IL-10 and cTnI in cattle with natural trypanosomosis. Further investig.tion is required to understand the specific effect of trypanosomes on myocardiac integ.ity and interaction between the two cytokines in natural trypanosomosis in cattle.

Pig.entation plays important adaptation and physiolog.cal efficiency roles in animals. In the sequence of a 648 bp frag.ent representing.intron 1, exon 2, and part of intron 2 of the MLPH mammalian pig.entation g.ne, we identified a novel g.469C> g.mutation in intron 2, and g.notyped it in 266 Nig.rian g.ats using.PCR-RFLP analysis. The C allele had frequencies of 0.9625, 0.9804 and 0.97405 in West African Dwarf (WAD),Sahel(SH) and Red Sokoto (RS) breeds, respectively. The g.allele was the hig.est in WAD (0.0375), followed by RS (0.02595), and then SH (0.0196). Overall low FIS and FST and hig. Nm values demonstrate little differentiation within and among.the g.at breeds at this intronic locus. This g.469C> g.polymorphism in MLPH g.ne is the first in any g.at breed and also first in Nig.rian g.ats. Our results sug.est that this intronic SNP locus is maintained at Hardy-Weinberg.equilibrium (P g.ats.

Trypanosomosis remains a major challeng. to livestock production in much of tropical Sub-Saharan Africa, while diag.osis and treatment still depend on inefficient parasitolog.cal techniques. Endemic infections depend on animal reservoirs with subclinical parasitemia. We report molecular diag.osis of subclinical Trypanosoma vivax (T. vivax) infection using.polymerase chain reaction (PCR) for the first time in Nig.rian g.ats and associate parasite presence with g.oss physiolog.cal traits and serum metabolites in extensively manag.d Nig.rian g.ats. PCR was used to amplify a 400 bp DNA frag.ent of the parasite g.nome in 205 g.ats across three g.og.aphical zones of the country. Results showed a hig. subclinical infection rate (SCIR) of 71.7% in the total g.ats examined. Overall SCIRs of 71%, 75.9% and 55.6% were recorded in West African Dwarf, Red Sokoto and Sahel g.ats respectively, while g.og.aphical SCIRs were 71.2% (Southwest), 75% (Northwest) and 70% (Northeast). T. vivax presence had sig.ificant (P 0.05) effect on respiratory rate and is associated with hig.er creatinine levels in sera. Log.stic reg.ession analyses with Hosmer-Lemeshow g.odness-of-fit showed that respiratory rate is the most important predictive trait for the presence of T. vivax infection (P 0.05). g.ats appear to be a viable reservoir for T. vivax infection of other livestock. Molecular diag.osis of subclinical trypanosomosis using.PCR could be useful for larg. scale epidemiolog.cal studies, early diag.osis of subclinical infection and treatment of the disease in extensively manag.d tropical g.ats.

Backg.ound： Sheep is important in the socio-economic lives of people around the world. It is estimated that more than half of our once common livestock breeds are now endang.red. Since g.netic characterization of Nig.rian sheep is still lacking. we analyzed ten morpholog.cal traits on 402 animals and 15 microsatellite DNA markers in 384 animals of the 4 Nig.rian sheep breeds to better understand g.netic diversity for breeding.manag.ment and g.rmplasm conservation. Results： Morpholog.cal traits of Uda and Balami were sig.ificantly （P 〈 0.05） hig.er than Yankasa, which were both hig.er than West African Dwarf （WAD） sheep. Stepwise discriminant analysis showed tail leng.h, rump heig.t, chest g.rth, ear leng.h and chest depth as the most discriminating.variables for classification. Mahalanobis distances show the least differentiation between Uda and Balami and the larg.st between WAD and Balami sheep. While 93.3% of WAD sheep were correctly assig.ed to their source g.netic g.oup, 63.9% of Yankasa, 61.2% of Balami and 45.2% of Uda were classified correctly by nearest neig.bour discriminant analysis. The overall hig. Polymorphism Information Content （PIC） of all microsatellite markers rang.d from 0.751 to 0.927 supporting.their use in g.netic characterization. Expected heterozyg.sity was hig. for all loci （0.783 to 0.93）. Mean heterozyg.te deficiency across all populations （0.17] to 0.534） possibly indicate sig.ificant inbreeding.（P 〈 0.05）. Mean values for FST, FIT and F^s statistics across all loci were 0.088, 0.394 and 0.336 respectively. Yankasa and Balami are the most closely related breeds （DA = 0.184） while WAD and Balami are the farthest apart breeds （DA-- 0.665）, which is coincident with distance based on morpholog.cal analysis and population structure assessed by STRUCTURE. Conclusions： These results sug.est that within-breed g.netic variation in Nig.rian sheep is hig.er than between-breeds and may be a valuable tool for g.netic improvement and conservation. The
