Investigating drivers of microbial activity and respiration in a forested bog

Northern peatlands store nearly one-third of terrestrial carbon(C)stocks while covering only 3%of the global landmass;nevertheless,the drivers of C cycling in these often-waterlogged ecosystems are different from those that control C dynamics in upland forested *** explore how multiple abiotic and biotic characteristics of bogs interact to shape microbial activity in a northern,forested bog,we added a labile C tracer(13C-labeled starch)to in situ peat mesocosms and correlated heterotrophic respiration with natural variation in several microbial predictor variables,such as enzyme activity and microbial biomass,as well as with a suite of abiotic variables and proximity to vascular plants *** found that peat moisture content was positively correlated with respiration and microbial activity,even when moisture levels exceeded total saturation,suggesting that access to organic matter substrates in drier environments may be limiting for microbial *** to black spruce trees decreased total and labile heterotrophic *** negative relationship may reflect the influence of tree evapotranspiration and peat shading effects;i.e.,microbial activity may decline as peat dries and cools near ***,we isolated the response of heterotrophic respiration to explore the variation in,and interactions among,multiple abiotic and biotic drivers that influence microbial *** approach allowed us to reveal the relative influence of individual drivers on C respiration in these globally important C sinks.

Widespread testing, case isolation and contact tracing may allow safe school reopening with continued moderate physical distancing: A modeling analysis of King County, WA data

Background:In late March 2020,a“Stay Home,Stay Healthy”order was issued in Washington State in response to the COVID-19 *** May 1,a 4-phase reopening plan *** investigated whether adjunctive prevention strategies would allow less restrictive physical distancing to avoid second epidemic waves and secure safe school ***:We developed a mathematical model,stratifying the population by age,infection status and treatment status to project SARS-CoV-2 transmission during and after the reopening *** model was parameterized with demographic and contact data from King County,WA and calibrated to confirmed cases,deaths and epidemic peak *** prevention interventions were simulated assuming different levels of pre-COVID physical interactions(pC_PI)***:the best model fit estimated~35%pC_PI under the lockdown which prevented~17,000 deaths by May *** restoring 75%pC_PI for all age groups between May 15-July 15 would have resulted in~350 daily deaths by early September ***th current testing practices to ensure low levels of daily infections and *** testing,isolation of symptomatic infections,and contact tracing permitted 60%pC_PI without significant increases in daily deaths before November and allowed opening of schools withtiviral treatment was predicted to reduce deaths significantly without lowering cases or ***:We predict that widespread testing,contact tracing and case isolation would allow relaxation of physical distancing,as well as opening of schools,without a surge in local cases and deaths.

In response to new European Union regulations,studies are underway to mitigate accumulation of toxic cadmium(Cd)in cacao(theobroma cacao,tc).this study advances such research with Cd isotope analyses of 19 genetically diverse cacao clones and yeast transformed to express cacao natural resistance-associated macrophage protein(NRAMP5)and heavy metal AtPases(HMAs).the plants were enriched in light Cd isotopes relative to the hydroponic solution withΔ^(114/110)Cd_(tot-sol)=−0.22±0.08‰.Leaves show a systematic enrichment of isotopically heavy Cd relative to total plants,in accord with closed-system isotope fractionation ofΔ^(114/110)C_(dseq-mob)=−0.13‰,by sequestering isotopically light Cd in roots/stems and mobilisation of remaining Cd to *** findings demonstrate that(i)transfer of Cd between roots and leaves is primarily unidirectional;(ii)different clones utilise similar pathways for Cd sequestration,which differ from those of other studied plants;(iii)clones differ in their efficiency of Cd *** yeast that expresses tcNRAMP5(*** natural resistance-associated macrophage gene)had isotopically lighter Cd than did *** suggests that NRAMP5 transporters constitute an important pathway for uptake of Cd by *** isotope signatures of transgenic yeast expressing HMA-family proteins suggest that they may contribute to Cd *** data are the first to record isotope fractionation induced by transporter proteins in vivo.

Immunotherapy remains more effective for hematologic tumors than for solid *** of the main challenges to immunotherapy of solid tumors is the immunosuppressive microenvironment these tumors generate,which limits the cytotoxic capabilities of immune effector cells(e.g.,cytotoxic t and natural killer cells).this microenvironment is characterized by hypoxia,nutrient starvation,accumulated waste products,and acidic ***-hijacked cells,such as fibroblasts,macrophages,and t regulatory cells,also contribute to this inhospitable microenvironment for immune cells by secreting immunosuppressive cytokines that suppress the antitumor immune response and lead to immune ***,there is a strong interest in developing new drugs and cell formulations that modulate the tumor microenvironment and reduce tumor cell immune *** systems(MPSs)are versatile tools that may accelerate the development and evaluation of these therapies,although specific examples showcasing the potential of MPSs remain *** in microtechnologies have led to the development of sophisticated microfluidic devices used to recapitulate tumor *** resulting models,also known as microphysiological systems(MPSs),are versatile tools with which to decipher the molecular mechanisms driving immune cell antitumor cytotoxicity,immune cell exhaustion,and immune cell exclusion and to evaluate new targeted ***,we review existing microphysiological platforms to study immuno-oncological applications and discuss challenges and opportunities in the field.

Correlates of subjectively and objectively measured physical activity in young adolescents

Background： Many studies examining individual-level correlates in youth utilize self-report rather than objective measures of physical activity （PA）. this utilization of self-report may result in associations that are not present when examining objectively measured PA. the present study investigates the relationship between hypothesized correlates of PA with objectively and subjectively measured PA. Methods： Participating children （n 232, 101 males, mean age - 12.3 years） provided a minimum of four monitored days of PA （via accelerometer） and completed a survey assessing moderate-to-vigorous physical activity （MVPA）, sport competence, appearance, enjoyment, and self-efficacy. Height and weight were measured and body mass index （BMI） was calculated. Results： Hierarchical regression models controlling for sex, race, and BMI Z-score showed that only sex and BMI Z-score were significant correlates of objective MVPA while only sex was a significant correlate of objective total PA. However, in a separate model examining the relationship with subjective MVPA, enjoyment of PA and self-efficacy for PA were the only significant correlates of self-reported PA. Conclusion： Measuring MVPA via self-report versus accelerometry produces considerably different results in a sample of young adolescents. Future studies should use caution when selecting outcome measures if the intent is to identify modifiable correlates of MVPA in youth.

AIM:Hydrogen sulfide (H2S) is a prominent gaseous constituent of the gastrointestinal (GI) tract with known cytotoxic properties. Endogenous concentrations of H2S are reported to range between 0.2-3.4 mmol/L in the GI tract of mice and humans. Considering such high levels we speculate that, at non-toxic concentrations, H2S may interact with chemical agents and alter the response of colonic epithelium cells to such compounds. the GI tract is a major site for the absorption of phytochemical constituents such as isothiocyanates, flavonoids, and carotenoids, with each group having a role in the prevention of human diseases such as colon cancer. the chemopreventative properties of the phytochemical agent p-phenyethyl isothiocyanate (PEItC) are well recognized. However, little is currently known about the physiological or biochemical factors present in the GI tract that may influence the biological properties of ItCs. the current study was undertaken to determine the effects of H2S on PEItC mediated apoptosis in colon cancer cells. MEtHODS: Induction of apoptosis by PEItC in human colon cancer HCt116 cells was assessed using classic apoptotic markers namely SubGl population analysis, caspase-3 like activity and nuclear fragmentation and condensation coupled with the Mtt (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrasodium bromide) viability assay and LDH leakage. RESULtS: PEItC significantly induced apoptosis in HCt116 cells as assessed by SubGl population formation, nuclear condensation, LDH leakage and caspase-3 activity after 24 h, these data being significant from control groups (Ptrast, co-treatment of cells with physiological concentrations of H2S (0.1-1 mmol/L) prevented PEItC mediated apoptosis as assessed using the parameters described. CONCLUSION: PEItC effectively induced cell death in the human adenocarcinoma cell line HCt116 in vitro through classic apoptotic mechanisms.
