Minimizing the risk of community spread of COVID-19 via institutional quarantine of high-risk travelers with serial viral RNA testing: A successful experience from Macao SAR, China

Macao,a special administrative region(SAR)of the People’s Republic of China,is located in southern China and shares the border with China's Mainland.It is the most densely populated region in the world,with a population of 667400 and a total land area of 32.9 square kilometers in 2019.Since the first case diagnosed on January 22,2020,there was a total of 45 laboratory-confirmed coronavirus disease 2019(COVID-19)cases in Macao,of which 43 patients(96%)were imported cases.To date,all patients had been discharged successfully from Centro Hospitalar Conde de São Januário,a designated hospital to manage all COVID-19 patients in Macao.Eventually,no patient died,and no local community outbreak was noted.This opinion review describes the underlying factors that could have contributed to the successful experience in Macao SAR,China,which include the following:(1)Early implementation of containment measures;(2)Large-scale quarantine using hotel rooms to reduce the risk of a local outbreak;and(3)Multidisciplinary co-operation and transparency of information to the public.Although the successful experience in Macao SAR,China,may not be generalized to other regions,it should not be unreasonable to be well prepared with sufficient logistic support to conduct timely containment and early detection of episodic cases to prevent the backsliding of COVID-19 outbreak.

Repairing Achilles tendon has emerged as a long-standing challenge in the orthopaedic surgeries.Although suture is the gold standard for re-attaching and repairing the fractured Achilles tendons in clinical surgeries,it is still subjected to numerous adverse side-effects,including chronic inflammatory,tendon tissue re-rupture,scar formation,and post-surgical peritendinous adhesion.In this work,we develop a class of hydrogel bioadhesives with tailored nanoscale phase separation for Achilles tendon repairing.To address the existing limitations of sutures,our hydrogel bioadhesives encompass three core functionalities:(i)instant and tough adhesion to Achilles tendon tissues,(ii)extraordinary long-term adhesion robustness under wet and dynamic in vivo conditions,and(iii)anti-postsurgical peritendinous adhesion.Combining our hydrogel bioadhesives with sutures,such kind of integrated approach enables a conformable yet robust biointerface with the tendon tissues,and prevents the fibroblast migration and formation of connective tissues,thus facilitating the tendon repairing.The hydrogel bioadhesives reported here open up new opportunities for the repairing of fractured Achilles tendons in diverse and complicated clinical scenarios.

Indications of pro-inflammatory cytokines in laparoscopic and open liver resection for early-stage hepatocellular carcinoma

Background:Our clinical practice of laparoscopic liver resection(LLR)had achieved better short-term and long-term benefits for patients with hepatocellular carcinoma(HCC)over open liver resection(OLR),but the underlying mechanisms are not clear.This study was to find out whether systemic inflammation plays an important role.Methods:A total of 103 patients with early-stage HCC under liver resection were enrolled(LLR group,n=53;OLR group,n=50).The expression of 9 inflammatory cytokines in patients at preoperation,postoperative day 1(POD1)and POD7 was quantified by Luminex Multiplex assay.The relationships of the cytokines and the postoperative outcomes were compared between LLR and OLR.Results:Seven of the circulating cytokines were found to be significantly upregulated on POD1 after LLR or OLR compared to their preoperative levels.Compared to OLR,the POD1 levels of granulocytemacrophage colony-stimulating factor(GM-CSF),interleukin-6(IL-6),IL-8,and monocyte chemoattractant protein-1(MCP-1)in the LLR group were significantly lower.Higher POD1 levels of these cytokines were significantly correlated with longer operative time and higher volume of blood loss during operation.The levels of these cytokines were positively associated with postoperative liver injury,and the length of hospital stay.Importantly,a high level of IL-6 at POD1 was a risk factor for HCC recurrence and poor disease-free survival after liver resection.Conclusions:Significantly lower level of GM-CSF,IL-6,IL-8,and MCP-1 after liver resection represented a milder systemic inflammation which might be an important mechanism to offer better short-term and long-term outcomes in LLR over OLR.

Adipose browning has demonstrated therapeutic potentials in several diseases.Here,by conducting transcriptomic profiling at the single-cell and single-nucleus resolution,we reconstituted the cellular atlas in mouse inguinal subcutaneous white adipose tissue(iWAT)at thermoneutrality or chronic cold condition.All major nonimmune cells within the iWAT,including adipose stem and progenitor cells(ASPCs),mature adipocytes,endothelial cells,Schwann cells,and smooth muscle cells,were recovered,allowing us to uncover an overall and detailed blueprint for transcriptomes and intercellular cross-talks and the dynamics during white adipose tissue brown remodeling.Our findings also unravel the existence of subpopulations in mature adipocytes,ASPCs,and endothelial cells,as well as new insights on their interconversion and reprogramming in response to cold.The adipocyte subpopulation competent of major histocompatibility complex class Ⅱ(MHCⅡ)antigen presentation is potentiated.Furthermore,a subcluster of ASPC with CD74 expression was identified as the precursor of this MHCⅡ^(+)adipocyte.Beige adipocytes are transdifferented from preexisting lipid generating adipocytes,which exhibit developmental trajectory from de novo differentiation of amphiregulin cells(Aregs).Two distinct immune-like endothelial subpopulations are present in iWAT and are responsive to cold.Our data reveal fundamental changes during cold-evoked adipose browning.

Prevalence and Factors Associated with Diabetes among City Hall Workers, Bohicon, Benin

Introduction: Living and working conditions are increasingly conducive to a sedentary lifestyle. Many administrative workers are exposed daily to long hours of work in a prolonged sitting position, which can favour the onset of diabetes. The aim of the study was to determine the prevalence and factors associated with diabetes among workers at a town hall in Benin in 2019. Methods: This was a descriptive cross-sectional study conducted from December 2018 to January 2019. Exhaustive recruitment was carried out. Data were collected during a face-to-face interview, using a standardized questionnaire including socio-professional factors, behavioral factors, anthropometric measurements, and blood glucose. Frequencies were calculated. Frequencies were compared using the chi-square and Fisher tests. The significance threshold was 5%. Results: A total of 85 workers were included. Their mean age was 41.55 ± 9.82 years, with a sex ratio of 6.1. = The prevalence of diabetes was estimated at 8.2% [95% CI: 3.38 - 16.23%] and 3 of the workers knew they had diabetes. A family history of diabetes was associated with diabetes (p = 0.032). Occupationally, 10.77% of permanent employees had diabetes, compared with 0.0% of non-permanent employees;12.50% of employees with at least 10 years’ seniority had diabetes, compared with 7.25% of those with less seniority;19.05% of employees who spent at least 6 hours a day sitting at work had diabetes, compared with 4.69% of those who spent less time sitting. Conclusion: Preventing chronic disease in the workplace is part of promoting workers’ health, and must take into account occupational factors such as prolonged sitting.

Chromatin modification contributes to pluripotency maintenance in embryonic stem cells(ESCs).However,the related mechanisms remain obscure.Here,we show that Npac,a"reader"of histone H3 lysine 36 trimethylation(H3K36me3),is required to maintain mouse ESC(mESC)pluripotency since knockdown of Npac causes mESC differentiation.Depletion of Npac in mouse embryonic fibroblasts(MEFs)inhibits reprogramming efficiency.Furthermore,our chromatin immunoprecipitation followed by sequencing(ChIP-seq)results of Npac reveal that Npac co-localizes with histone H3K36me3 in gene bodies of actively transcribed genes in mESCs.Interestingly,we find that Npac interacts with positive transcription elongation factor b(p-TEFb),Ser2-phosphorylated RNA PolⅡ(RNA PolⅡSer2P),and Ser5-phosphorylated RNA PolⅡ(RNA PolⅡSer5 P).Furthermore,depletion of Npac disrupts transcriptional elongation of the pluripotency genes Nanog and Rif1.Taken together,we propose that Npac is essential for the transcriptional elongation of pluripotency genes by recruiting p-TEFb and interacting with RNA PolⅡSer2P and Ser5P.

借力数字经济发展降低地方政府土地财政依赖,既是提高地方财政可持续性的必然选择,也是促进经济高质量发展的必由之路。基于2011-2021年我国277个地级市的面板数据,实证分析数字经济发展对地方政府土地财政依赖的影响及其作用机制。研究表明:数字经济发展能够显著地降低地方政府土地财政依赖,但该影响因城市地理区位和资源赋呈现异质性特征。机制分析表明,财政压力在数字经济发展影响地方政府土地财政依赖的过程中起到中介作用,经济增长压力在其中发挥负向调节作用。门槛效应检验发现,数字经济发展对地方政府土地财政依赖的影响存在门槛效应,当数字经济发展水平或数字基础设施建设水平跨过门槛值时,数字经济发展对地方政府土地财政依赖的影响显著增强。因此,要加快数字基础设施建设,为数字经济发展降低地方政府土地财政依赖奠定基础;持续释放数据要素潜能,以经济发展模式转型破解地方政府财政困局;健全地方政绩考核机制,以制度创新持续规范地方政府土地出让行为。

Role of Intrahepatic Regional Immunity in Post-Transplant Cancer Recurrence

Hepatic malignancy is a major indication for liver transplantation;however,post-tra ns plant cancer recurrence is an emerging clinical challenge affecting long-term outcomes.Pre-tra ns plant tumor biology,staging,and post-transplant immunosuppression regimens have been elucidated as risk factors for recurrent liver cancer.However,increasing evidence indicates that hepatic ischemia and reperfusion(IR)injury to allografts are crucial to providing a favorable immunologic microenvironment for cancer cell invasiveness and metastasis after liver transplantation.The association of severe graft injury in marginal grafts,such as small-for-size or fatty grafts,with lower recurrence-free survival rates in living donor liver transplantations,substantiates the correlation between hepatic IR injury and cancer recurrence.IR has been demonstrated to trigger intrahepatic immunological microenvironment remodeling,including pro-inflammatory responses exacerbating graft injury and anti-inflammatory responses promoting tissue repair.However,the role of regional immunity in post-transplant cancer recurrence is not comprehensively understood.This review describes the up-to-date evidence of the intrahepatic humoral microenvironment and regional regulatory immunological microenvironment induced by IR injury,as well as their roles in cancer recurrence after liver transplantation.A comprehensive understanding of regional immunity will provide novel precise diagnostic,therapeutic,and prognostic strategies for post-transplant cancer recurrence.

Several diverse long noncoding RNAs(lncRNAs)have been identified to be involved in hepatitis B virus(HBV)replication and oncogenesis,especially those dysregulated in HBV-related hepatocellular carcinoma(HCC).Most of these dysregulated lncRNAs are modulated by the HBV X protein.The regulatory mechanisms of some lncRNAs in HBV replication and oncogenesis have been characterized.Genetic polymorphisms of several lncRNAs affecting HBV replication or oncogenesis have also been studied.The prognosis of HCC remains poor.It is important to identify novel tumor markers for early diagnosis and find more therapeutic targets for effective treatments of HCC.Some dysregulated lncRNAs in HBV-related HCC may become biomarkers for early diagnosis and/or the therapeutic targets of HCC.This mini-review summarizes these findings briefly,focusing on recent developments.

基于文献计量的人机界面中用户体验中外对比研究

目的对比分析国内外人机界面用户体验研究的异同,总结国内学界目前的研究现状并发现问题。方法研究中使用WOS与CNKI作为数据来源,通过文献计量学的方法及综合运用CiteSpace、VOSviewer等软件将文献资料进行可视化分析。结论国内外人机界面用户体验研究领域文献产出较为一致,都在2019年及之前达到产出高峰,2019年后出现回落趋势。研究主体是与计算机科学、互联网技术等相关的理工科院校。在团队合作方面,国内跨团队的研究相对较少,存在明显的研究壁垒,需要加强研究合作的程度。在总体上呈现出“整体分散、局部集中”的特点。在关键词聚类分析上,国内外人机界面用户体验相关热点研究呈现出一定的相似性与差异性。相似性表现为国内外人机界面用户体验研究在关键词聚类上都形成四个聚类,构成研究闭环且国内外学界对可用性都有较大的关注度。而差异性方面则表现为,在与用户体验强相关的交互方式研究上,国际学界视线已脱离传统的人机界面交互方式,转而专注于虚拟交互技术,而国内目前处于初始阶段。跨学科合作不足、自身研究创新性不强、缺少自己的理论研究与模型等则是现阶段国内HCI-UE研究主的要问题。
