Resveratrol prevents hypoxia-induced retinal ganglion cell death related with ErbB2

AIM: To confirm the changes in proteins related with hypoxia-induced retinal cell death and to assess the effects of resveratrol(Res).METHODS: The therapeutic effect of Res was verified using an ischemic/reperfusion(I/R) model in vivo and a hypoxia modelin retinal ganglion cells(RGCs) in vitro.Death of RGCs were confirmed by TUNEL assay.Protein expression was confirmed by Western blotting and immunohistochemistry.In addition, flow cytometric analysis was used to confirm the response in the cell unit to obtain more accurate data.RESULTS: ErbB2 expression and apoptosis in the ganglion cell layer(GCL) increased after I/R injury.Treatment of Res rescued I/R-induced ganglion cell death, downregulated apoptosis and ErbB2 protein expression in the retina.In subsequent in vitro models, Res affects apoptosis by regulating the phosphorylation and expression of mouse double minute 2 homolog(MDM2), along with those of ErbB2.These results suggest that Res reverses GCL-specific apoptosis via downregulation of ErbB2 in ischemic injury.CONCLUSION: In light of Res favorable properties, it should be evaluated in the treatment of RGC death and related retinal disease characterized by ErbB2 and MDM2 expression.Therefore, Res is appropriate therapeutic agent for treating ischemic injury-related eye diseases by targeting the expression of ErbB2 and MDM2.

The fourth industrial revolution indispensably brings explosive data processing and storage;thus,a new computing paradigm based on artificial intelligence-enabling device structure is urgently required.Memristors have received considerable attention in this regard because of their ability to process and store data at the same location.However,fundamental problems with abrupt switching characteristics limit their practical application.To address this problem,we utilized the concept of metaplasticity inspired by biosystems and observed gradual switching in the peptide-based memristor at high proton conductivity.An unexpectedly high slope value>1.7 in the logI–V curve at low voltage(≤400 mV)was considered the main origin,and it might arise from the modulatory response of proton ions on the threshold of Ag ion migration in the peptide film.With the obtained gradual switching property at high proton conductivity,the device showed significantly increased accuracy of image recognition(~82.5%).We believe that such a demonstration not only contributes to the practical application of neuromorphic devices but also expands the bioinspired functional synthetic platform.

Endoscopic ultrasound-guided biliary drainage with placement of a fully covered metal stent for malignant biliary obstruction

AIM:To determine the utility of endoscopic ultrasoundguided biliary drainage(EUS-BD)with a fully covered self-expandable metal stent for managing malignant biliary stricture. METHODS:We collected data from 13 patients who presented with malignant biliary obstruction and underwent EUS-BD with a nitinol fully covered selfexpandable metal stent when endoscopic retrograde cholangiopancreatography(ERCP)fails.EUS-guided choledochoduodenostomy(EUS-CD)and EUS-guided hepaticogastrostomy(EUS-HG)was performed in 9 patients and 4 patients,respectively. RESULTS:The technical and functional success rate was 92.3%(12/13)and 91.7%(11/12),respectively. Using an intrahepatic approach(EUS-HG,n=4),there was mild peritonitis(n=1)and migration of the metal stent to the stomach(n=1).With an extrahepatic approach(EUS-CD,n=10),there was pneumoperitoneum(n=2),migration(n=2),and mild peritonitis (n=1).All patients were managed conservatively with antibiotics.During follow-up(range,1-12 mo),there was re-intervention(4/13 cases,30.7%)necessitated by stent migration(n=2)and stent occlusion(n=2). CONCLUSION:EUS-BD with a nitinol fully covered self-expandable metal stent may be a feasible and effective treatment option in patients with malignant biliary obstruction when ERCP fails.

Effect of Asiasari radix on osteoblastic differentiation of stem cells derived from gingiva

OBJECTIVE: To examine the dose-dependent impact of Asiasari Radix(A. radix) on the cell viability,differentiation and mineralization of stem cells derived from gingiva.METHODS: Stem cells that were derived from gingiva were grown in the presence of A. radix at final concentrations that ranged from 0.001 to 10 μg/m L. The morphology of the cells was viewed under an inverted microscope and the analysis of cell proliferation was performed by using Cell Counting Kit-8(CCK-8) on day 1. The alkaline phosphatase activity test was used to assess differentiation and Alizarin red S staining was used to assess mineralization of treated cells.RESULTS: The control group showed spindleshaped, fibroblast-like morphology and the shapesof the cells in 0.001, 0.01, 0.1, 1 and 10 μg/mL of A.radix were similar to that of the control group at day 1. The cultures growing in the presence of0.001 μg/m L of A. radix at day 1 showed an increase in the CCK-8 value(P 0.05). Mineralized extracellular deposits were observed after Alizarin Red S staining and the cultures grown in the presence of 0.001 μg/m L of A. radix showed the highest value for quantitative results for bound dye(P < 0.05).CONCLUSION: Within the limits of this study, A. radix influenced the proliferation of stem cells derived from the gingiva and low concentrations of A.radix might enhance osteogenic differentiation of the stem cells.

Background: Electroacupuncture(EA) may reduce the severity of acute pancreatitis(AP) and provide additional pain relief in patients with chronic pancreatitis. However, the ability of EA to relieve pain in patients with AP has not been well documented.Objective: This study was undertaken to compare the pain-relieving effects of EA and conventional treatment in patients with AP.Design, setting, participants and interventions: This study was conducted using a randomized, controlled,three-arm, parallel-group and multi-center design. Patients diagnosed with AP were randomly and equally assigned to EA1, EA2 or control groups. All participants received conventional standard-of-care therapy for AP. Local EA alone was administered in EA1, and local plus distal EA was given in EA2.Local EA included two abdominal acupoints, while distal EA included twelve peripheral acupoints. EA groups underwent one session of EA daily for 4 days(days 1–4), or until pain was resolved or discharged.Main outcome measures: The primary outcome measure was the change in the visual analogue scale(VAS;0–100) pain score between baseline and day 5.Results: Eighty-nine participants were randomized into EA1, EA2 and control groups, and 88(EA1, 30;EA2, 29;control, 29) were included in the full-analysis set. VAS score change(median [interquartile range]) on day 5 was(12.3 ± 22.5) in the EA1 group,(10.3 ± 21.5) in the EA2 group, and(8.9 ± 15.2) in the control group. There were not significant differences in the change in VAS score among treatments(P = 0.983). However, time to food intake was significantly shorter in the EA group(EA1 + EA2) than in the control group(median 2.0 days vs 3.0 days), with a hazard ratio of 0.581(P = 0.022;95% CI,0.366–0.924). No significant adverse events occurred.Conclusion: EA treatment did not significantly reduce pain after 4 days of treatment in patients with AP-associated abdominal pain but significantly reduced time to first food intake.

Characterization of three-dimensional channel reservoirs using ensemble Kalman filter assisted by principal component analysis

Ensemble-based analyses are useful to compare equiprobable scenarios of the reservoir models.However,they require a large suite of reservoir models to cover high uncertainty in heterogeneous and complex reservoir models.For stable convergence in ensemble Kalman filter(EnKF),increasing ensemble size can be one of the solutions,but it causes high computational cost in large-scale reservoir systems.In this paper,we propose a preprocessing of good initial model selection to reduce the ensemble size,and then,EnKF is utilized to predict production performances stochastically.In the model selection scheme,representative models are chosen by using principal component analysis(PCA)and clustering analysis.The dimension of initial models is reduced using PCA,and the reduced models are grouped by clustering.Then,we choose and simulate representative models from the cluster groups to compare errors of production predictions with historical observation data.One representative model with the minimum error is considered as the best model,and we use the ensemble members near the best model in the cluster plane for applying EnKF.We demonstrate the proposed scheme for two 3D models that EnKF provides reliable assimilation results with much reduced computation time.

Protective Effect of Brassica napus L.Hydrosols against Inflammation Response in RAW 264.7 Cells

Objective:To demonstrate the anti-inflammatory activity of Brassica napus L.hydrosols(BNH)in lipopolysaccharide(LPS)-stimulated RAW 264.7 cells.Methods:Composition analysis of BNH was conducted via gas chromatography-mass spectrometry after BNH were extracted.The nitric oxide(NO)production was measured using the Griess assay.Prostaglandin E2(PGE2)production was evaluated with enzyme-linked immunosorbent assay.The effects of BNH on LPS-induced pro-inflammatory enzymes including inducible nitric oxide synthase(iNOS)and cyclooxygenase-2(COX-2)were evaluated using Western blot analysis.Furthermore,phosphorylation of nuclear factor-kappa B(N F-k B)and nuclear translocation of N F-k B p65 were evaluated with Western blot analysis and immunofluorescence staining,respectively.Results:Compared with LPS-stimulated cells,BNH markedly decreased the generation of NO and PGE_(2)in LPS-stimulated RAW 264.7 cells(P<0.01 or P<0.05).Moreover,BNH inhibited protein levels of iNOS and COX-2(P<0.01).Phosphorylation of NF-k B and nuclear translocation of NF-k B p65 was significantly inhibited by BNH(P<0.01 or P<0.05).Conclusion:The anti-inflammatory activities of BNH were mediated via blockage of the N F-k B signaling pathways in LPS-stimulated RAW 264.7 cells.

Background Anethole(AN)is an organic antioxidant compound with a benzene ring and is expected to have a positive impact on early embryogenesis in mammals.However,no study has examined the effect of AN on porcine embryonic development.Therefore,we investigated the effect of AN on the development of porcine embryos and the underlying mechanism.Results We cultured porcine in vitro-fertilized embryos in medium with AN(0,0.3,0.5,and 1 mg/mL)for 6 d.AN at 0.5 mg/mL significantly increased the blastocyst formation rate,trophectoderm cell number,and cellular survival rate compared to the control.AN-supplemented embryos exhibited significantly lower reactive oxygen species levels and higher glutathione levels than the control.Moreover,AN significantly improved the quantity of mitochondria and mitochondrial membrane potential,and increased the lipid droplet,fatty acid,and ATP levels.Interestingly,the levels of proteins and genes related to the sonic hedgehog(SHH)signaling pathway were significantly increased by AN.Conclusions These results revealed that AN improved the developmental competence of porcine preimplantation embryos by activating SHH signaling against oxidative stress and could be used for large-scale production of high-quality porcine embryos.

Gastrosplenic fistula occurring in lymphoma patients: Systematic review with a new case of extranodal NK/T-cell lymphoma

AIM To provide the overall spectrum of gastrosplenic fistula(GSF) occurring in lymphomas through a systematic review including a patient at our hospital.METHODS A comprehensive literature search was performed in the MEDLINE database to identify studies of GSF occurring in lymphomas. A computerized search of our institutional database was also performed. In all cases, we analyzed the clinicopathologic/radiologic features, treatment and outcome of GSF occurring in lymphomas. RESULTS A literature search identified 25 relevant studies with 26 patients. Our institutional data search added 1 patient. Systematic review of the total 27 cases revealed that GSF occurred mainly in diffuse, large B-cell lymphoma(n = 23), but also in diffuse, histiocytic lymphoma(n = 1), Hodgkin's lymphoma(n = 2), and NK/T-cell lymphoma(n = 1, our patient). The common clinical presentations are constitutional symptoms(n = 20) and abdominal pain(n = 17), although acute gastrointestinal bleeding(n = 6) and infection symptoms due to splenic abscess(n = 3) are also noted. In all patients, computed tomography scanning was very helpful for diagnosing GSF and for evaluating the lymphoma extent. GSF could occur either post-chemotherapy(n = 10) or spontaneously(n = 17). Surgical resection has been the most common treatment. Once patients have recovered from the acute illness status after undergoing surgery, their long-term outcome has been favorable. CONCLUSION This systematic review provides an overview of GSF occurring in lymphomas, and will be helpful in making physicians aware of this rare disease entity.

Surface exsolution and local atomic structure of amorphous CeTiO_x

We made precipitated nano-ceria(~5 nm) on the surface of the catalyst by heat treatment of Cesupersaturated amorphous CeTiOxto improve the oxygen storage properties of CeO_2. The catalysts were prepared by sol-gel methods and TiO_2 nanoparticles were preferentially generated as a core material to form selective Ce-supersaturated structure on the catalyst surface. Reaction temperature and amount of doping element are optimized to induce selective crystallization of CeO_2. Cee Ce(2 nd shell)bond around 0.38 nm of Ce L3-edge extended X-ray absorption fine structure is reduced and nanostructure of precipitated ceria on the surface is observed by HREM. The catalyst is present as amorphous with precipitated nano-CeO_2 on the surface. The de-NOxefficiency of the catalyst, which has precipitated CeO_2, improves by ~50% owing to the simultaneous reactions of the nano CeO_2 and the amorphous CeTiO_x.
