Alterations of Gefitinib Pharmacokinetics by Co-administration of Herbal Medications in Rats

Objective： To evaluate the potential pharmacokinetic interactions of the anticancer agent gefitinib （Iressae） and the oriental medications Guipi Decoction （归脾汤, GPD, Guibi-tang in Korean） and Bawu Decoction （八物汤, BWD, Palmul-tang in Korean）. Methods： Methylceliulose （MC, control）, GPD （1,200 mg/kg）, or BWD （6,000 mg/kg） was orally administered to rats either as a single dose or multiple doses prior to gefitinib administration. To examine the effects of a single dose of the herbal medicines, gefitinib （10 mg/kg） was orally administered after 5 min or 1 h of MC or the herbal medicine pretreatments. To examine the effects of the multiple doses of the herbal medicines, gefltinib （10 mg/kg） was orally administered following 7 consecutive days of the administration of MC or each herbal medicine. The plasma concentrations of gefitinib were determined with liquid chromatography-tandem mass spectrometry assay. The plasma concentration-time profiles of gefitinib were analyzed with a noncompartmental analysis. Results： Gefitinib was rapidly absorbed and showed a mono- exponential decline with an elimination half-life of 3.7-4.1 h. The pharmacokinetics of gefitinib was not affected by GPD pretreatment. However, a significantly lower maximum plasma concentration （Crux, P〈0.05） and area under the curve （P〈0.05）, and a delayed time to reach Cmax （Tmax, P〈0.01） were observed in both single- and multiple- dose BWD-pretreated rats compared with the control rats. Conclusions： BWD and not GPD might delay and interfere with gefitinib absorption. Further evaluations of the clinical significance of these findings are needed.

The time domain entombment of bacteria by intratubular mineralization following orthograde canal obturation with mineral trioxide aggregate（MTA） was studied by scanning electron microscopy（SEM）. Single-rooted human premolars（n560） were instrumented to an apical size #50/0.06 using ProF ile and treated as follows： Group 1（n510） was filled with phosphate buffered saline（PBS）; Group 2（n510） was incubated with Enterococcus faecalis for 3 weeks, and then filled with PBS; Group 3（n520） was obturated orthograde with a paste of OrthoM TA（BioM TA, Seoul, Korea） and PBS; and Group 4（n520） was incubated with E. faecalis for 3 weeks and then obturated with OrthoM TA–PBS paste. Following their treatments, the coronal openings were sealed with PBS-soaked cotton and intermediate restorative material（IRM）, and the roots were then stored in PBS for 1, 2, 4, 8 or 16 weeks. After each incubation period, the roots were split and their dentin/MTA interfaces examined in both longitudinal and horizontal directions by SEM. There appeared to be an increase in intratubular mineralization over time in the OrthoM TA-filled roots（Groups 3 and 4）. Furthermore, there was a gradual entombment of bacteria within the dentinal tubules in the E. faecalis inoculated MTA-filled roots（Group 4）. Therefore, the orthograde obturation of root canals with OrthoM TA mixed with PBS may create a favorable environment for bacterial entombment by intratubular mineralization.

Effect of the semen extract of Cuscuta chinensis on inflammatory responses in LPS-stimulated BV-2 microglia

AIM: To investigate the anti-inflammatory activities of the semen extract of Cuscuta chinensis Lam.(Cuscutae Semen; CS) on the production of inflammatory mediators, nitric oxide(NO), prostaglandin 2(PGE2), and proinflammatory cytokines in lipopolysaccharide(LPS)-stimulated BV-2 microglia. METHOD: BV-2 cells were treated with CS extract for 30 min, and then stimulated with LPS or without for 24 h. The levels of NO, PGE2 and proinflammatory cytokines were measured by Griess assay and ELISA. The expression of inducible nitric oxide synthase(iNOS), and cyclooxygenase(COX)-2 mRNA and protein was determined by RT-PCR and Western blot, respectively. The phosphorylation of extracellular signal-regulated kinase 1/2(ERK1/2), Jun N-terminal kinase(JNK), and p38 mitogen-activated protein kinase(MAPK), and the nuclear expression of nuclear factor(NF)-κB p65 were investigated by Western blot analysis. RESULTS: CS extract significantly decreased the production of NO and PGE2 by suppressing the expression of iNOS and COX-2 in activated microglia. CS extract decreased the production of TNF-α, IL-1β, and IL-6 by down-regulating their transcription levels. In addition, CS extract suppressed the phosphorylation of ERK1/2, JNK, and p38 MAPK, and the nuclear translocation of NF-κB p65 in activated microglia. CONCLUSION: These results indicate that CS extract is capable of suppressing the inflammatory response by microglia activation, suggesting that CS extract has potential in the treatment of brain inflammation.

Mistletoes are parasite plants growing on various tree species,taking nutrients from the *** compounds of mistletoe were used in medicine and these mistletoes were commonly collected from oak species that were not well *** analyzed the nutritional value by measuring proximate and mineral components of Korean mistletoe(Viscum album ***)from various host *** was the most proximate component in both leaves and twigs followed crude protein,crude fiber,crude ash and crude *** the contents of the proximate components and minerals varied among hosts and between plant parts,leaves contained higher crude protein and ash,and more minerals except for magnesium than twigs,although no significant difference was found inmistletoe grown on Prunus *** were significant differences of proximate and mineral contents depending on the host *** grown on Chaenomeles sinensis contained higher levels of carbohydrates,crude fat,crude ash,magnesium,calcium,sodium,iron,and zinc than mistletoe grown on other *** grown on *** contained high mineral levels of potassium,copper and *** nutritional value of mistletoes were greatly influenced by the host species but among the hosts,specific nutrients accumulated in leaves more than in twigs,and vice *** on the analysis,Korean mistletoe contains relatively good quality of nutrients and has high potential for dietary supplements as well as for medicinal compounds.

Inverted cystic tubulovillous adenoma involving Brunner’s glands of duodenum

Benign neoplasia of the duodenum are very rare. Moreover, duodenal tubulovillous adenomas are more uncommon lesions. The microscopic structure of tubulovillous adenoma has frond-like projection of mucosa with branching papillary structure and generally upward growth into the lumen. We describe a 72-year-old man who showed aduodenal tubulovillous adenoma with unusual inverted cystic growth pattern. Interestingly, this tubulovillous adenomatous lesion was interrupted by gastric metaplasia in the deep portion of the cyst and was closely surrounded by Brunner’s glands. Although histogenesis of gastric metaplasia of duodenum is not fully understood, Brunner’s glands has been suggested as a precursor for gastric metaplasia. Therefore, these findings argued that this adenoma arises from Brunner’s glands through gastric metaplasia. This is the first case of inverted cystic tubulovillous adenoma involving Brunner’s glands of duodenum with gastric metaplasia.

Body-centered-cubic martensite and the role on room-temperature tensile properties in Si-added SiVCrMnFeCo high-entropy alloys

We present a new class of metastable high-entropy alloys(HEAs),triggering deformation-induced martensitic transformation(DIMT)from face-centered-cubic(FCC)to body-centered-cubic(BCC),i.e.,*** the ab-initio calculation based on 1 st order axial interaction model and combined with the Gibbs free energy calculation,the addition of Si is considered as a critical element which enables to reduce the intrinsic stacking fault energy(ISFE)in Si_xV_((9-x))Cr_(10)Mn_5 Fe_(46)Co_(30)(x=2,4,and 7 at.%)alloy *** ISFE decreases from-30.4 to-35.5 mJ/m^(2)as the Si content increases from 2 to 7 at.%,which well corresponds to the reduced phase stability of FCC against *** BCC-DIMT occurs in all the alloys via intermediate HCP martensite,and the HCP martensite provides nucleation sites of BCC ***,the transformation rate enhances as the Si content increases in an earlier deformation ra ***,the BCC-DIMT is also affected by the phase stability of FCC against BCC,and the stability is the highest at the Si content of 7 at.%.Thus,the 7Si alloy presents the moderate transformation rate in the later deformation *** to the well-controlled transformation rate and consequent strain-ha rdening rate,the 7Si alloy possesses the superior combination of strength and ductility beyond 1 GPa of tensile strength at room *** results suggest that the Si addition can be a favorable candidate in various metastable HEAs for the further property improvement.

弄清明末耶稣会传教史上译名、礼仪争论发生的主要原因,一直是后代学者们关注的重点。现有的主要研究观点将利玛窦和龙华民之间的关系一分为二地对立了起来,把龙华民视为耶稣会在中国传教失败和阻碍东西方文化交流的负面人物。如果置身于耶稣会“传教神学—天学”的历史变迁过程中,就可以发现这种认识忽视了平民路线出现的真实背景和过程。本文将重点放在龙华民对利玛窦的继承性上,力图通过对被忽视的龙华民平民灵性传教路线进行分析和考察,还原出其中隐藏的历史真相和意义。在耶稣会内,接受了利玛窦遗命的龙华民不仅继承了利玛窦的知识传教,同时还非常重视福音的纯洁性,将维持自己摸索出来的传教路线与利玛窦路线之间的均衡作为核心工作。纵观龙华民的一生,可以说他是耶稣会灵修本土化即福音的完美道德和博爱主义的典范。

The beneficial impacts of splanchnic vasoactive agents on hepatic functional recovery in massive hepatectomy porcine model

Background:Excessive portal pressure after massive hepatectomy can cause hepatic sinusoidal injury and have deleterious impacts on hepatic functional recovery,contributing to developing post-hepatectomy liver *** study aimed to assess the effects of splanchnic vasoactive agents on hepatic functional recovery and regeneration while clarifying the underlying mechanism,using a 70%hepatectomy porcine ***:Eighteen pigs undergoing 70%hepatectomy were involved in this study and divided into three groups:control(n=6),terlipressin(n=6),and octreotide(n=6).Terlipressin(0.5 mg)and octreotide(0.2 mg)were administered 3 times a day for each group with the first dose starting just before surgery until the 7th postoperative day,at which time the surviving pigs were *** the period,portal pressure,liver weight,biochemical analysis,histological injury score,and molecular markers were evaluated and compared between ***:The 7-day survival rates in the octreotide,terlipressin,and control groups were 100%,83.3%,and 66.7%,*** portal pressures decreased in both terlipressin and octreotide groups than the control group at 30 minutes,1 hour and 6 hours after *** amount of regeneration measured by liver weight to body weight ratio at the time of sacrifice in the terlipressin group was smaller than that in the control group(117%vs.129%,P=0.03).Serum aspartate aminotransferase(AST)and total bilirubin levels at 1 and 6 hours after hepatectomy and prothrombin time/international normalized ratio(PT/INR)at 6 hours after hepatectomy were significantly improved in the terlipressin and octreotide groups compared to the control *** endothelin-1(ET-1)was significantly lower in the terlipressin group than that in the control group 6 hours after hepatectomy(P<0.01).The histological injury score in the control group was significantly higher than that in the terlipressin group on the 7th postoperative day(P<0.01).Conclusions:Splanc

本文根据晚明在华耶稣会保守主义的实际历史状况、形成和发展的过程展开论述,再现了龙华民在广东韶州的传教策略。龙华民在韶州民间传教的经历为后来以儒家士大夫为主要传教对象的耶稣会利玛窦传教策略向平民阶层转换提供了经验。换言之,译名礼仪保守策略、信仰的纯粹性固守、乐观论的直接传教工作、平民归化运动等耶稣会文化传教策略,基本上是初期韶州灵性神学的平民传教经验的延伸。龙华民福音保守传教策略是从恢复教会的纯洁性和信仰完整性的角度出发,向着探索与利玛窦文化适应策略的动态均衡的方向发展,从客观上增加了信教人数。

Evaluation of non-ampullary duodenal polyps: Comparison of non-neoplastic and neoplastic lesions

AIM: To evaluate duodenal polyps, divided into nonneoplastic and neoplastic lesions. In addition, the clinical characteristics of duodenal hyperplastic polyps are determined. METHODS: We analyzed medical records of 50 114 consecutive patients submitted to for first diagnostic esophago-gastroduodenoscopy between January 2004 and December 2009. We excluded lesions on the ampulla of Vater and submucosal tumors. We studied 510 cases that were diagnosed endoscopically with duodenal polyps and enrolled a total of 221 cases that had undergone tissue biopsy. We analyzed the differences between non-neoplastic and neoplastic lesions, and determined the clinical features of duodenal hyperplastic polyps. RESULTS: Non-neoplastic lesions were found in 196 patients and neoplastic lesions in 25 patients. On uni-variate analysis, there were significant differences in shape, location, and size. Polyps more than 10 mm in diameter or polyps in the second portion had independent risk factors for being neoplastic lesions, as identified by multivariate analysis. In 23 cases of hyperplastic polyps (79.3%), they were accompanied by gastroduodenal pathology, which was possibly associated with Helicobacter pylori . CONCLUSION: Polyps of more than 10 mm or polyps in the second portion of the duodenum should be evaluated by histological examination.
