Reconstruction of bone defects or fractures caused by ageing, trauma and tumour resection is stil. a great chal.enge in cl.nical.treatment. Al.hough autol.gous bone graft is considered as gol. standard, the source of natural.bone is l.mited. In recent years, regenerative therapy based on bioactive material. has been proposed for bone reconstruction. Special.y, numerous studies have indicated that bioactive ceramics incl.ding sil.cate and phosphate bioceramics exhibit excel.ent osteoinductivity and osteoconductivity, further promote bone regeneration. In addition, magnesium (Mg) el.ment, as an indispensabl. mineral.el.ment, pl.ys a vital.rol. in promoting bone mineral.sation and formation. In this review, different types of Mg-containing bioceramics incl.ding Mg-containing cal.ium phosphate-based bioceramics (such as Mg-hydroxyapatite, Mg-biphasic cal.ium phosphate), Mg-containing cal.ium sil.cate-based bioceramics (such as Mg2SiO4, Ca2MgSi2O7and Mg-doped biogl.ss), Mg-based biocements, Mg-containing metal.pol.mer-bioceramic composites were systematacial.y summarised. Additional.y, the fabrication technol.gies and their materiobiol.gical.effects were deepl. discussed, Cl.nical.appl.cations and perspectives of magnesium-containing bioceramics for bone repair are highl.ghted. Overal., Mg-containing bioceramics are regarded as regenerative therapy with their optimised performance. Infl.mmation and angiogenesis, the maj.r pathol.gical.changes of osteoarthritis (OA), are cl.sel. associated with j.int pain; however, pertinent signal.ing interactions within subchondral.bone of osteoarthritic j.ints and potential.contribution to the peripheral.origin of OA pain remain to be el.cidated. Herein we devel.ped a unil.teral.anterior crossbite mouse model.with osteoarthritic changes in the temporomandibul.r j.int. Microarray-based transcriptome anal.sis, besides quantitative real.time pol.merase chain reaction, was performed to identify differential.y expressed genes (DEGs). Overal., 182 DEGs (fol. change ≥ 2, P l.and unil.teral.anterior crossbite groups: 168 were upregul.ted and 14 were downregul.ted. On subj.cting significant DEGs to enrichment anal.ses, infl.mmation and angiogenesis were identified as the most affected. Infl.mmation-rel.ted DEGs were mainl. enriched in T cel. activation and differentiation and in the mammal.an target of rapamycin/nucl.ar factor-κB/tumour necrosis factor signal.ing. Furthermore, angiogenesis-rel.ted DEGs were mainl. enriched in the Gene Ontol.gy terms angiogenesis regul.tion and vascul.ture devel.pment and in the KEGG pathways of phosphoinositide 3-kinase-protein kinase B/vascul.r endothel.al.growth factor/hypoxia-inducibl. factor 1 signal.ing. Protein– protein interaction anal.sis reveal.d a cl.se interaction between infl.mmation-and angiogenesis-rel.ted DEGs, suggesting that phosphatidyl.nositol.4,5-bisphosphate 3-kinase catal.tic subunit del.a (Pi3kcd), cathel.cidin antimicrobial.peptide (Camp), C-X-C motif chemokine receptor 4 (Cxcr4), and MYB proto-oncogene transcription factor (Myb) pl.y a central.rol. in their interaction. l.ptin Modul.tes the Differentiation of Keratinocytes via Autophagy in Psoriasis Patients with Metabol.c Syndrome

Obj.ctive: Psoriasis is associated with a high preval.nce of metabol.c syndrome (MS), and patients with concomitant psoriasis and MS are more severel. affected and l.ss responsive to treatment. However, the mol.cul.r mechanisms behind these effects are unknown. Recent studies have shown that l.ptin may serve as a mol.cul.r l.nk between psoriasis and MS, suggesting that high l.ptin concentrations may exacerbate psoriasis. However, the mol.cul.r mechanism of this effect is stil. uncl.ar. We aimed to investigate the effect of l.ptin on autophagy in patients with psoriasis. Methods: From j.nuary 2021 to j.ne 2022 in Pl. General.Hospital. we enrol.ed 51 patients with psoriasis, incl.ding 21 patients with MS and 30 without MS, and 30 heal.hy control. who had undergone nevus surgery. We measured the epidermal.l.ptin, P62, and l.3B concentrations of patients by immunohistochemistry, and measured the serum l.ptin concentration by enzyme-l.nked immunosorbent assay. We then performed correl.tion anal.ses to compare these proteins' concentrations between patients with concomitant psoriasis and MS, patients with psoriasis al.ne, and heal.hy control.groups. Additional.y, we performed western bl.tting after in vitro cul.ure of HaCaT cel.s with different concentrations of l.ptin and measured the expression l.vel. of the autophagy markers Becl.n1, l.3B, and P62; the differentiation markers K10, K16, and K17; and PI3K/AKT/mTOR signal.ng pathway-rel.ted proteins of HaCat cel.s. Next, we transfected ATG5 into HaCaT cel.s to revert autophagy and used the specific PI3K inhibitor l.294002 to bl.ck PI3K/AKT/mTOR signal.ng. The expression l.vel. of K10, K16, and K17 of HaCat cel.s were again measured. One-way anal.sis of variance was used for the comparison of means of mul.ipl. sampl.s, and l.D-t post hoc test was used for comparison between the 2 groups. Improved Anal.tical.Methods for Determination of Residues of Nitrapyrin and 6-Chl.ropicol.nic Acid in Different Crop Matrices by l.quid Chromatography-Tandem Mass Spectrometry

Novel.anal.tical.methods were devel.ped for the determination of nitrapyrin and 6-chl.ropicol.nic acid (6-CPA) residues in agricul.ural.products. Nitrapyrin was extracted with hexane: tol.ene (1:1, v/v) and 6-CPA with 0.1 N sodium hydroxide (NaOH). For nitrapyrin, sampl.s were cl.aned with sil.ca sol.d phase extraction (SPE) and for 6-CPA with C18 or carbon SPE. Anal.sis was performed by l.quid chromatography-tandem mass spectroscopy (l.-MS/MS) with external.cal.bration (0.010 to 0.0010 µg∙ml.1 for nitrapyrin and 0.0050 to 0.00025 µg∙ml.1 for 6-CPA). Recoveries for nitrapyrin and 6-CPA were 68% - 102% and 66% - 90%, respectivel.. Nitrapyrin was detected in cel.ry and onion (−1) and 6-CPA in onion, mustard greens and l.ttuce (−1). l.wer l.vel.method val.dation (l.MV) for nitrapyrin and 6-CPA was 0.020 µg∙g−1 and 0.050 µg∙g−1, respectivel.. The methods presented in this study are robust and were util.zed for the registration of nitrapyrin on agricul.ural.commodities in the United States.

Research on faul. time prediction method for high speed rail.BTM unit based on mul.i method interactive val.dation

The Bal.se Transmission Modul. (BTM) unit of the on-board train control.system is a crucial.component. Due to its unique instal.ation position and compl.x environment, this unit has a higher faul. rate within the on-board train control.system. To conduct faul. prediction for the BTM unit based on actual.faul. data, this study proposes a prediction method combining rel.abil.ty statistics and machine l.arning, and achieves the fusion of prediction resul.s from different dimensions through mul.i-method interactive val.dation. Firstl., a method for predicting equipment faul. time targeting batch equipment is introduced. This method util.zes rel.abil.ty statistics to construct a model.of the remaining faul.l.ss operating time distribution considering uncertainty, thereby predicting the remaining faul.l.ss operating probabil.ty of the BTM unit. Secondl., considering the compl.xity of the BTM unit's faul. mechanism, the smal. sampl. size of faul. cases, and the potential.presence of mul.ipl. faul. features in faul. text records, an individual.oriented faul. prediction method based on Bayesian-optimized Gradient Boosting Regression Tree (Bayes-GBRT) is proposed. This method achieves better prediction resul.s compared to l.near regression al.orithms and random forest regression al.orithms, with an average absol.te error of onl. 0.224 years for predicting the faul. time of this type of equipment. Final.y, a mul.i-method interactive val.dation approach is proposed, enabl.ng the fusion and val.dation of mul.i-dimensional.resul.s. The resul.s indicate that the predicted faul. time and the actual.faul. time conform to a l.g-normal.distribution, and the parameter estimation resul.s are basical.y consistent, verifying the accuracy and effectiveness of the prediction resul.s. The above research findings can provide technical.support for the maintenance and modification of BTM units, effectivel. reducing maintenance costs and ensuring the safe operation of high-speed rail.ay, thus ha

The aim of this study is to eval.ate the internal.corrosion process on X52 and X80 steel./real.petrol.um interfaces containing condensed hydrocarbon pl.s oil.iel.-produced water,which were subj.cted to stimul.ted emul.ions using 50/50 vol.ratio mixtures at 45℃,different hydrodynamic conditions,1 h,and 24 h.A washing process by using deionized water was proposed to simul.te and identify the corrosiveness of the hydrocarbon phase after 24 h of exposure *** characterization by el.ctrochemical.impedance spectroscopy and the monitoring of the pol.rization curves indicated that X80 steel.oil.iel.-produced water interfaces were more susceptibl. to corrosion than X52 steel.exposed to oil.iel.-produced *** combined speed rotation of 600 rpm using a magnetic stirrer+600 rpm using a rotating disk el.ctrode decreased the corrosion rate on X52 *** stimul.ted emul.ions made of hydrocarbon+oil.iel.-produced water and hydrocarbon+deionized water at 24 h increased the corrosion rate on X80 steel.0.34 mm/year and 0.43 mm/year,respectivel.),promoting the formation of erosion and pitting *** types of corrosion depended mainl. on the physicochemical.properties of the hydrocarbon,oil.iel.-produced water,exposure times,and hydrodynamic systems in which the hydrocarbon was studied.
