Food nutrition and toxicology targeting on specific organs in the era of single-cell sequencing

Due to the complex natures of dietary food components,it is difficult to elucidate how the compounds affect host health.Dietary food often selectively presents its mechanism of action on different cell types,and participates in the modulation of targeted cells and their microenvironments within organs.However,the limitations of traditional in vitro assays or in vivo animal experiments cannot comprehensively examine cellular heterogeneity and the tissue-biased influences.Single-cell RNA sequencing(sc RNA-seq)has emerged as an indispensable methodology to decompose tissues into different cell types for the demonstration of transcriptional profiles of individual cells.Sc RNA-seq applications has been summarized on three typical organs(brain,liver,kidney),and two representative immune-and tumor related health problems.The everincreasing role of sc RNA-seq in dietary food research with further improvement can provide sub-cellular information and the coupling between other cellular modalities.In this review,we propose utilizing sc RNAseq to more effectively capture the subtle and complex effects of food chemicals,and how they may lead to health problems at single-cell resolution.This novel technique will be valuable to elucidate the underlying mechanism of both the health benefits of food nutrients and the detrimental consequences food toxicants at the cellular level.

Lignocellulosic biomass is a critical renewable carbon resource,but most of its utilization is inefficient,and elec-trocatalytic oxidation is a promising method of upgrading lignocellulose into value-added fuels and chemicals under mild operating conditions.Recently,efforts to enable conversion with a high efficiency and low energy con-sumption have been reported,but understanding the reaction mechanisms and realizing scaled-up applications of the electrooxidation of lignocellulosic biomass are still in their early stages.A timely overview of recently reported general reaction mechanisms,particularly the strategies developed for use in improving the reaction efficiencies,is necessary to inspire research regarding the highly efficient utilization of lignocellulose.Herein,we summa-rize the strategies developed to improve electrocatalytic performance in oxidative lignocellulose conversion.The organized summary includes strategies ranging from designing efficient electrocatalysts and adding functional co-catalysts or electrolytes to employing advanced electrolyzers.A comprehensive overview of representative examples should provide universal principles to yield insight into the reaction processes and guide the design of efficient electrocatalytic systems.Finally,the challenges and opportunities in developing the electrocatalytic oxidative upgrading of lignocellulosic biomass in the near future are proposed.

Arecoline causes cognitive impairment and potential addiction via disturbing Kenyon cells in bumblebee brain

Areca nut is the basic ingredient of betel quid， which is chewed by hundreds of millions of people in south-eastern Asia. Chewing of areca nut has been associated with oral cancers potentially due to its specific alkaloids， among which arecoline constitutes about 90% of total fraction. Being the world’s fourth most commonly used psychoactive substance， arecoline evokes stimulation， addiction， and other direct neurological effects， while its misuse correlates to neurotoxic effects. However， what might underlie its neurotoxic mechanisms has been poorly documented. The brain is encoded by a complex network of neuronal and glial cell types， and neurotoxicity of hazardous compounds present transcriptional heterogeneity. Recently， the eusocial bumblebee has been used as a model for studying brain effects， with sophisticated cognitive capability and precisely measured brain architecture. Here， we decipher cell-type-specific mental risks to arecoline using bumblebees. Arecoline induced locomotor hyperactivity and cognitive impairment. Single nucleus RNA sequencing (snRNA-seq) unearthed arecoline-induced cell-specific responses， primarily targeted on Kenyon cells (KC). Moreover， high-dose arecoline induced distinctive cell responses among KC subtypes， particularly class I large Kenyon cell (lKC)， leading to DNA damage， excitatory/inhibitory (E/I) imbalance， and calcium dyshomeostasis， which potentially resulted in cognitive impairment. Given arecoline’s popularity and growing exposure risks to humans， neurological health risks of areca nut warrant serious consideration.

Suitable lithium polysulfides diffusion and adsorption on CNTs@TiO_(2)-bronze nanosheets surface for high-performance lithium-sulfur batteries

The shuttle effect of lithium polysulfides(UPSs)in lithium-sulfur batteries(LSBs)has been hampered their commercialization.Metal oxides as separator modifications can suppress the shuttle effect.Since there is no direct electron transport between metal oxides and UPSs,absorbed UPSs should be diffused from the surface of metal oxides to the carbon matrix to go through redox reactions.If diffusivity of UPSs from metal oxides surface to carbon substrate is poor,it would hinder the redox reactions of LiPSs.Nevertheless,researchers tend to focus on the adsorption and overlook the diffusion of UPSs.Herein,same morphology and different crystal phase of TiO_(2) nanosheets grown on carbon nanotubes(CNTs@TiO_(2)-bronze and CNTs@TiO_(2)-anatase)have been designed via a simple approach.Compared with CNTs and CNTs@TiO_(2)-anatase composites,the battery with CNTs@TiO_(2)-bronze modified separator delivers higher specific capacities and stronger cycling stability,especially at high current rates(~472 mAh·g^(-1) at 2.0 C after 1,000 cycles).Adsorption tests,density functional theory calculations and electrochemical performance evaluations indicate that suitable diffusion and adsorption for LiPSs on the CNTs@TiO_(2)-B surface can effectively capture LiPSs and promote the redox reaction,leading to the superior cycling performances.

Inhaled drug-containing nanocarriers have been well applied as an important strategy in pulmonary dis-eases.Acute lung injury(ALI)is characterized by abnormal lung tension and complex pathogenesis,with high mortality because of the limitations of targeted intervention.Accordingly,in the present study,we first identified that the mechanosensitive ion channel Piezo1 participated in the ALI-associated processes induced by lipopolysaccharide(LPS)in vitro and in vivo.Then,chitosan-stabilized bovine serum albu-min nanoparticles(NCs)emulsified with the Piezo1 inhibitor GsMTx4(NC-GsMTx4)were generated and exhibited excellent biocompatibility and biological function.Through aerosol inhalation,NC-GsMTx4 im-proved lung injury and inhibited cell apoptosis in LPS-stressed ALI mice and alleviated pulmonary fibrosis during the later stage of ALI.Mechanistically,GsMTx4 could regulate inflammation and apoptosis in lung epithelial cells via NF-κB and ERK1/2 signaling.In summary,our findings provide new insights into the pathological mechanisms of Piezo1 in ALI progression,and nebulized inhalation of NC-GsMTx4 offers a prospective platform for targeting Piezo1 to treat ALI efficiently and conveniently.

Rational design and synthesis of multimetallic nanostructures(NSs)are fundamentally important for electrochemical CO_(2)reduction reaction(CO_(2)RR).Herein,a multi-step seed-mediated growth method is applied to synthesize asymmetric AuAgCu heterostructures using Au nanobipyramids as nucleation seeds,in which their composition and structures are well controlled.We find that the selectivity of C_(2)products for CO_(2)RR could be effectively regulated by tandem catalysis and electronic effect over trimetallic AuAgCu heterostructures.Particularly,the Faraday efficiency toward ethanol could reach up to 37.5%at a potential of−0.8 V versus reversible hydrogen electrode over asymmetric Au1Ag1Cu5 heterostructures with segregated domains of three constituent metals.This work provides an efficient strategy for the synthesis of multicomponent architectures to boost their promising application in CO_(2)RR.

Berberine alleviates non-alcoholic hepatic steatosis partially by promoting SIRT1 deacetylation of CPT1A in mice

Background:Berberine effectively alleviates non-alcoholic fatty liver disease(NAFLD).Nevertheless,the mechanism is incompletely comprehended.It has been reported that SIRT1 mediates lipid metabolism in liver and berberine promotes the expression of SIRT1 in hepatocytes.We hypothesized that SIRT1 mediated the effect of berberine on NAFLD.Methods:The effects of berberine on NAFLD were evaluated in C57BL/6J mice fed a high-fat diet(HFD)and in mouse primary hepatocytes and cell lines exposed to palmitate.The change of fatty acid oxidation(FAO)and the activity of CPT1A were observed in HepG2 cells.Quantitative real-time polymerase chain reaction and Western blot were employed to observe the expression of SIRT1 and lipid metabolism-related molecules.The interaction between SIRT1 and CPT1A was investigated by using co-immunoprecipitation assay in HEK293T cells.Results:Berberine treatment attenuated hepatic steatosis,reduced triglyceride(190.1611.2 lmol/g liver vs 113.667.6 lmol/g liver,P<0.001)and cholesterol(11.362.5 lmol/g liver vs 6.360.4 lmol/g liver,P<0.001)concentration in the liver,and improved lipid and glucose metabolism disorders compared with the HFD group.The expression of SIRT1 was reduced in the liver of NAFLD patients and mouse models.Berberine increased the expression of SIRT1 and promoted the protein level of CPT1A and its activity in HepG2 cells.SIRT1 overexpression mimicked the effect of berberine on reducing triglyceride levels in HepG2 cells,whereas SIRT1 knock-down attenuated the effect of berberine.Mechanistically,berberine increased the expression of SIRT1.SIRT1 deacetylated CPT1A at the Lys675 site,which suppressed its ubiquitin-dependent degradation,thereby promoting FAO and alleviating non-alcoholic liver steatosis.Conclusions:Berberine promoted SIRT1 deacetylation of CPT1A at the Lys675 site,which reduced the ubiquitin-dependent degradation of CPT1A and ameliorated non-alcoholic liver steatosis.

Synthetic conditions and ligands are the key structural defining factors of metal-organic frameworks(MOFs).Therefore,reasonable optimization of these aspects is considered to be an effective means for designing materials with novel structures and target functions.Herein,two novel Co(Ⅱ)-based MOFs,namely[Co(HL)(dibp)]_(n)(HL-8) and{[Co_(2)(L)(OH)(dibp)]·DMA}_n(HL-9)(H_(3)L=2',6'-dimethyl-[1,1'-biphenyl]-3,4',5-tricarboxylic acid;dibp=4,4'-di(1H-imidazol-1-yl)-1,1-biphenyl]),have been hydrothermally synthesized and structurally characterized.HL-8 crystallizes in the orthorhombic system (Pna2_(1)) with a grid layer structure,while HL-9 crystallizes in the monoclinic P2_(1)/n space group assembled through Co_(4)(OH)_(2)clusters with organic ligands.Remarkably,benefiting from the finite cage-like structure,HL-9 exhibited enhanced performance in carbon dioxide(CO_(2)) adsorption/catalytic transformation and excellent size selectivity during dye molecular adsorption process.

Hepatitis B virus X protein induces ALDH2 ubiquitin-dependent degradation to enhance alcoholic steatohepatitis

Background:Excessive alcohol intake with hepatitis B virus(HBV)infection accelerates chronic liver disease progression and patients with HBV infection are more susceptible to alcohol-induced liver disease.Hepatitis B virus X protein(HBx)plays a crucial role in disease pathogenesis,while its specific role in alcoholic liver disease(ALD)progression has not yet been elucidated.Here,we studied the role of HBx on the development of ALD.Methods:HBx-transgenic(HBx-Tg)mice and their wild-type littermates were exposed to chronic plus binge alcohol feeding.Primary hepatocytes,cell lines,and human samples were used to investigate the interaction between HBx and acetaldehyde dehydrogenase 2(ALDH2).Lipid profiles in mouse livers and cells were assessed by using liquid chromatography–mass spectrometry.Results:We identified that HBx significantly aggravated alcohol-induced steatohepatitis,oxidative stress,and lipid peroxidation in mice.In addition,HBx induced worse lipid profiles with high lysophospholipids generation in alcoholic steatohepatitis,as shown by using lipidomic analysis.Importantly,serum and liver acetaldehyde were markedly higher in alcoholfed HBx-Tg mice.Acetaldehyde induced lysophospholipids generation through oxidative stress in hepatocytes.Mechanistically,HBx directly bound to mitochondrial ALDH2 to induce its ubiquitin–proteasome degradation,resulting in acetaldehyde accumulation.More importantly,we also identified that patients with HBV infection reduced ALDH2 protein levels in the liver.Conclusions:Our study demonstrated that HBx-induced ubiquitin-dependent degradation of mitochondrial ALDH2 aggravates alcoholic steatohepatitis.

Pd-MnO_2 nanoparticles/TiO_2 nanotube arrays(NTAs) photo-electrodes photo-catalytic properties and their ability of degrading Rhodamine B under visible light

Pd-MnO2/TiO2 nanotube arrays（NTAs） photo-electrodes were successfully fabricated via anodization and electro deposition subsequently; the obtained Pd-MnO2/TiO2 NTAs photo electrodes were analyzed by scanning electron microscopy（SEM）, X-ray diffraction（XRD） and characterized accordingly. Moreover, the light harvesting and absorption properties were investigated via ultraviolet–visible diffuse reflectance spectrum（DRS）; photo degradation efficiency was investigated via analyzing the photo catalytic degradation of Rhodamine B under visible illumination（xenon light）. The performed analyses illustrated that Pd-MnO2 codoped particles were successfully deposited onto the surface of the TiO2 nanotube arrays;DRS results showed significant improvement in visible light absorption which was between400 and 700 nm. Finally, the photo catalytic degradation efficiency results of the designated organic pollutant（Rhodamine B） illustrated a superior photocatalytic（PC） efficiency of approximately 95% compared to the bare TiO2 NTAs, which only exhibited a photo catalytic degradation efficiency of approximately 61%, thus it indicated the significant enhancement of the light absorption properties of fabricated photo electrodes and their yield of UOH radicals.
