Punicalagin ameliorates collagen-induced arthritis by downregulating M1 macrophage and pyroptosis via NF-κB signaling pathway

Rheumatoid arthritis(RA)is a chronic inflammatory disease that eventually leads to *** cell infiltration,severe joint breaking and systemic bone loss are the main clinical *** this study,we established a collagen-induced arthritis(CIA)model and found a large number of M1 macrophages and pyroptosis,which are important sources of proinflammatory ***(PUN)is an active substance extracted from pomegranate *** found that it inhibited joint inflammation,cartilage damage and systemic bone destruction in CIA *** effectively alleviated the high expression of inflammatory cytokines in synovial tissue in *** treatment shifted macrophages from the M1 phenotype to the M2 phenotype after stimulation with lipopolysaccharide(LPS)and interferon(IFN)-γ.The expression of inducible nitric oxide synthase(i NOS)and other proinflammatory cytokines released by M1 macrophages was decreased in the PUN treatment ***,simultaneously,the expression of markers of anti-inflammatory M2 macrophages,such as arginase(Arg)-1 and interleukin(IL)-10,was *** addition,PUN treatment attenuated pyroptosis by downregulating the expression of NLRP3 and caspase-1,thereby preventing inflammatory cell death resulting from the release of IL-1βand ***,PUN inhibited the activation of receptor activators of the nuclear factor-κB(NF-κB)signaling pathway,which contributes to M1 polarization and pyroptosis of *** concluded that PUN ameliorated pathological inflammation by inhibiting M1 phenotype polarization and pyroptosis and has great potential as a therapeutic treatment for human RA.

在西方文学批评史上,莎士比亚与《哈姆雷特》始终受到批评家的关注,并不断激发出新的诠释和想象,以致在相关的论说中,我们或许已经无法断定何为正确,却只能去描述变换不居的错误方式。“莎士比亚问题”也已经不仅是关于作家和文本的争论,而是不同语境从中延伸的理解和意义。本文以莎士比亚及其《哈姆雷特》为个案,追索最具代表性的诠释方法和批评类型,特别是当代的意识形态批评、性别身份批评和宗教观念批评,从而既为中国语境中的莎士比亚研究提供参照,也使我们更加直观地理解西方的主要批评流派。

Early detection of joint degeneration in the subtalar and talonavicular joints secondary to ankle instability in a mouse model

Background:Many patients with ankle sprain have secondary subtalar and talonavicular injuries,which have a great impact on their life and ***::This study aimed to investigate the biomechanical impact of ankle instability on the subtalar joint(STJ)and talonavicular joint(TNJ)using a mouse model to enhance the clinical understanding of early stage secondary joint ***:Twenty four 8-week old male C57BL/6 J mice were randomly allocated to three *** the calcaneofibular ligament(CFL)and anterior talofibular ligament(ATFL)of the left ankle were cut off in the CFLþATFL group to simulate severe ankle instability while only the CFL was excised in the CFL group to mimic moderate ankle *** operation was done in SHAM group to simulate the normal *** histological analysis,atomic force microscopy(AFM)–based nanoindentation tests and inspection by scanning electron microscopy(SEM)were used to assess the degree of joint degeneration in the tibiotalar joint(TTJ),STJ and TNJ of three ***:A significant increase of histological score for both CFL and CFLþATFL group was demonstrated in the TTJ,compared with the SHAM *** in the STJ and TNJ,the histological score only increased for CFLþATFL ***-based nanoindentation results demonstrated significant differences in elastic modulus between all groups in the TTJ,while for the STJ and TNJ,only the comparison between the CFLþATFL group and SHAM group was considered statistically *** SEM presented signs of cracking and disintegration within the subchondral bone(SCB)interface in the CFLþATFL group ***:The findings indicate that severe ankle instability resulted in early degeneration of the STJ and TNJ in a mouse model,which may provide referential values for clinical practice.

New frontiers in biomaterials research for tissue repair and regeneration

The field of biomaterials has recently emerged to augment or replace lost or damaged tissues and organs due to the human body's limited ability to self-heal large defects. Historically, metallic components, polymers, ceramics, and composite materials were utilized as synthetic materials along with natural materials to assist in *** novel biomaterials were developed to respond to a significant amount of new medical challenges in the past decade. Therefore, there is a need to review these newly developed biomaterials and their potential to improve tissue repair and regeneration in a variety of applications. Here, we briefly review the different strategies and attempts to use novel biomaterials, including self-assembled and macromolecular biomaterials, hydrogels, metamaterials, decellularized tissues, and biomaterials obtained via synthetic biology, used either for tissue repair and regeneration or for therapeutic use by exploiting other mechanisms of healing. All these methods aim to create functional materials, devices, systems, and/or organisms with novel and useful functions on the basis of catalogued and standardized biological building blocks. This review details the various methods and introduces the applications of these biomaterials in tissue repair and regeneration, especially for bone, nerve, and skin applications.

退行性脊柱侧凸(degenerative scoliosis,DS)是成人最常见的脊柱畸形类型,尤其在中老年人群中发病率较高。随着人口老龄化趋势的加剧,其发病率逐年上升,给社会和个人带来了沉重的经济负担。DS的病因复杂,与椎间盘退变、生物力学改变、骨质疏松和肌肉减少症等因素密切相关。诊断DS依赖临床体格检查和影像学检查,脊柱侧凸研究学会(Scoliosis Research Society,SRS)分型、Schwab分型、SRS-Schwab分型、Lenke-Silva分型、Berjano分型等分型系统为其分类和治疗提供了标准。DS的治疗包括保守治疗和手术治疗。保守治疗如物理治疗、药物治疗主要用于控制症状,治疗效果缺乏高等级的临床研究证据支持。手术治疗需要个性化规划,微创手术技术提供了创伤小、恢复快的治疗选择,但存在特定适应证和禁忌证。针对目前存在的疾病病因复杂、机制不清晰、微创手术存在局限、手术方式差异化大等问题,进一步深入DS的生物学机制研究非常必要。其目的是拓展安全、有效的微创技术应用,精细化个体化治疗策略和手术规划,以应对人口老龄化背景下DS带来的健康和经济挑战。

One-year clinical study of NeuroR egen scaffold implantation following scar resection in complete chronic spinal cord injury patients

The objective of this clinical study was to assess the safety and feasibility of the collagen scaffold, Neuro Regen scaffold, one year after scar tissue resection and implantation. Scar tissue is a physical and chemical barrier that prevents neural regeneration. However, identification of scar tissue is still a major challenge. In this study, the nerve electrophysiology method was used to distinguish scar tissue from normal neural tissue, and then different lengths of scars ranging from 0.5–4.5 cm were surgically resected in five complete chronic spinal cord injury(SCI) patients. The NeuroR egen scaffold along with autologous bone marrow mononuclear cells(BMMCs), which have been proven to promote neural regeneration and SCI recovery in animal models, were transplanted into the gap in the spinal cord following scar tissue resection. No obvious adverse effects related to scar resection or Neuro Regen scaffold transplantation were observed immediately after surgery or at the 12-month follow-up. In addition, patients showed partially autonomic nervous function improvement, and the recovery of somatosensory evoked potentials(SSEP) from the lower limbs was also detected. The results indicate that scar resection and Neuro Regen scaffold transplantation could be a promising clinical approach to treating SCI.

随着人口老龄化的进展,骨质疏松症已经成为全社会的公共健康问题。骨质疏松性椎体压缩骨折(osteoporotic vertebral compression fracture,OVCF)是由于骨质疏松导致椎体骨密度和骨质量下降,骨强度降低,脊柱受轻微外力即发生的骨折,是骨质疏松最常见的并发症。OVCF患者由于疼痛、卧床导致骨量进一步丢失,有较高的概率发生骨折不愈合或脊柱再骨折。同时,椎体骨折导致了脊柱后凸畸形,进一步影响患者心肺功能和胃肠功能,严重影响患者的生活质量。针对这一问题,我们经过长期的基础研究和临床探索,逐步形成了针对OVCF诊断治疗的系列规范[1]。随着新的理念和循证医学证据的出现,OVCF的诊疗方式也在不断革新和优化。本文在医学证据的基础上,结合我们的临床经验,对OVCF微创诊治中的热点问题进行思考和探讨。
