Incorporation of kartogenin and silk fibroin scaffolds promotes rat articular cartilage regeneration through enhancement of antioxidant functions

Treating articular cartilage defects in patients remains a challenging task due to the absence of blood vessels within the cartilage *** regenerative potential is further compromised by an imbalance between anabolism and catabolism,induced by elevated levels of reactive oxygen ***,the advent of tissue engineering introduces a promising strategy for cartilage regeneration,offering viable solutions such as mechanical support and controlled release of chondrogenic molecules or *** this study,we developed an antioxidant scaffold by incorporating natural silk fibroin(SF)and kartogenin(KGN)-loaded liposomes(SF-Lipo@KGN).The scaffold demonstrated appropriate pore size,connectivity,and water absorption and the sustained release of KGN was achieved through the encapsulation of *** vitro experiments revealed that the SF-Lipo@KGN scaffolds exhibited excellent biocompatibility,as evidenced by enhanced cell adhesion,migration,and proliferation of *** SF-Lipo@KGN scaffolds were found to stimulate cartilage matrix synthesis through the activation of the nuclear factor erythroid-2-related factor 2/heme oxygenase-1 antioxidant signaling *** vivo experiments demonstrated the effective promotion of articular cartilage regeneration by the SF-Lipo@KGN scaffolds,which enhanced extracellular matrix anabolism and restored the intrinsic redox ***,this study successfully developed biomimetic KGN-loaded scaffolds that restore cartilage redox homeostasis,indicating promising prospects for cartilage tissue engineering.

Upon the osteoporotic condition,sluggish osteogenesis,excessive bone resorption,and chronic inflammation make the osseointegration of bioinert titanium(Ti)implants with surrounding bone tissues difficult,often lead to prosthesis loosening,bone collapse,and implant *** this study,we firstly designed clickable mussel-inspired peptides(DOPA-N3)and grafted them onto the surfaces of Ti materials through robust catechol-TiO2 coordinative ***,two dibenzylcyclooctyne(DBCO)-capped bioactive peptides RGD and BMP-2 bioactive domain(BMP-2)were clicked onto the DOPA-N3-coated Ti material surfaces via bio-orthogonal *** characterized the surface morphology and biocompatibility of the Ti substrates and optimized the osteogenic capacity of Ti surfaces through adjusting the ideal ratios of BMP-2/RGD at 3:*** vitro,the dual-functionalized Ti substrates exhibited excellent promotion on adhesion and osteogenesis of mesenchymal stem cells(MSCs),and conspicuous immunopolarization-regulation to shift macrophages to alternative(M2)phenotypes and inhibit inflammation,as well as enhancement of osseointegration and mechanical stability in osteoporotic *** summary,our biomimetic surface modification strategy by bio-orthogonal reaction provided a convenient and feasible method to resolve the bioinertia and clinical complications of Ti-based implants,which was conducive to the long-term success of Ti implants,especially in the osteoporotic or inflammatory conditions.

Advancing flood susceptibility modeling using stacking ensemble machine learning: A multi-model approach

Flood susceptibility modeling is crucial for rapid flood forecasting, disaster reduction strategies, evacuation planning, and decision-making. Machine learning(ML) models have proven to be effective tools for assessing flood susceptibility. However, most previous studies have focused on individual models or comparative performance, underscoring the unique strengths and weaknesses of each model. In this study, we propose a stacking ensemble learning algorithm that harnesses the strengths of a diverse range of machine learning models. The findings reveal the following:(1) The stacking ensemble learning, using RF-XGBCB-LR model, significantly enhances flood susceptibility simulation.(2) In addition to rainfall,key flood drivers in the study area include NDVI, and impervious surfaces. Over 40% of the study area, primarily in the northeast and southeast, exhibits high flood susceptibility, with higher risks for populations compared to cropland.(3) In the northeast of the study area,heavy precipitation, low terrain, and NDVI values are key indicators contributing to high flood susceptibility, while long-duration precipitation, mountainous topography, and upper reach vegetation are the main drivers in the southeast. This study underscores the effectiveness of ML, particularly ensemble learning, in flood modeling. It identifies vulnerable areas and contributes to improved flood risk management.

Selenium-modified bone cement promotes osteoporotic bone defect repair in ovariectomized rats by restoring GPx1-mediated mitochondrial antioxidant functions

Over-accumulation of reactive oxygen species(ROS)causes mitochondrial dysfunction and impairs the osteogenic potential of bone marrow-derived mesenchymal stem cells(BMMSCs).Selenium(Se)protects BMMSCs from oxidative stress-induced damage;however,it is unknown whether Se supplementation can promote the repair of osteoporotic bone defects by rescuing the impaired osteogenic potential of osteoporotic BMMSCs(OP-BMMSCs).In vitro treatment with sodium selenite(Na_(2)SeO_(3))successfully improved the osteogenic differentiation of OP-BMMSCs,as demonstrated by increased matrix mineralization and up-regulated osteogenic genes *** importantly,Na_(2)SeO_(3) restored the impaired mitochondrial functions of OP-BMMSCs,significantly up-regulated glutathione peroxidase 1(GPx1)expression and attenuated the intracellular ROS and mitochondrial *** of Gpx1 completely abrogated the protective effects of Na_(2)SeO_(3) on mitochondrial functions of OP-BMMSCs,suggesting the important role of GPx1 in protecting OP-BMMSCs from oxidative *** further fabricated Se-modified bone cement based on silk fibroin and calcium phosphate cement(SF/CPC).After 8 weeks of implantation,Se-modified bone cement significantly promoted bone defect repair,evidenced by the increased new bone tissue formation and enhanced GPx1 expression in ovariectomized *** findings revealed that Se supplementation rescued mitochondrial functions of OP-BMMSCs through activation of the GPx1-mediated antioxidant pathway,and more importantly,supplementation with Se in SF/CPC accelerated bone regeneration in ovariectomized rats,representing a novel strategy for treating osteoporotic bone fractures or defects.

Collagen scaffolds modified with collagen-binding bFGF promotes the neural regeneration in a rat hemisected spinal cord injury model

Nerve conduit is one of strategies for spine cord injury(SCI)***,studies showed that biomaterials could guide the neurite growth and promote axon regeneration at the injury ***,the scaffold by itself was difficult to meet the need of SCI functional *** basic fibroblast growth factor(bFGF)administration significantly promotes functional recovery after organ ***,using a rat model of T9 hemisected SCI,we aimed at assessing the repair capacity of implantation of collagen scaffold(CS)modified by collagen binding bFGF(CBD-bFGF).The results showed that CS combined with CBD-bFGF treatment improved survival rates after the lateral hemisection *** CS/CBD-bFGF group showed more significant improvements in motor than the simply CS-implanted and untreated control group,when evaluated by the 21-point Basso-Beattie-Bresnahan(BBB)score and footprint *** hematoxylin and eosin(H&E)and immunohistochemical staining of neurofilament(NF)and glial fibrillary acidic protein(GFAP)demonstrated that fibers were guided to grow through the *** findings indicated that administration of CS modified with CBD-bFGF could promote spinal cord regeneration and functional recovery.

Neural differentiation of allogenic mixed-cultured rat bone mesenchymal stem cells

Many studies showed that bone mesenchymal stem cells (BMSCs) can transdifferentiate to neural cell in vitro. The purpose of this study was to investigate the mixed-culture of allogenic rat BMSCs in vitro, and to perform neural differentiation potential characterization. Rat BMSCs were isolated and cultured by plastic adherence and density gradient centrifugation respectively, and the 3rd passage cells were harvested and mixed-cultured with same concentration. The second passage cells of the mixed-cultured cells were obtained to perform Wright-Gemsa staining for morphological observation, to identify the surface marker of CD29, CD45, CD90 by flow cytometry and induced to differentiate into neural cell, then performed immunocytochemistry of Nestin, NSE and GFAP. Results showed that after Wright-Giemsa staining, the mixed-cultured cells displayed typical spindle-shape, presented layered and whirlpool-like growth;and the mixed-cultured cells were positive for CD29 and CD90, but negative for CD45 by flow cytometry. After induction, the mixed-cultured cells appeared morphological changes of neuron and glial cell, and were positive expression of Nestin, neuron specific enolase (NSE) and glial fibrillary acidic protein (GFAP). The results demonstrated that the mixed-cultured allogenic rat BMSCs can be induced into neural cell such as neuron and glial cell in vitro.

OBJECTIVE:To assess the efficacy and safety of gingerindirect moxibustion for chronic fatigue syndrome(CFS).METHODS:In this central randomized,controlled trial,290 CFS participants were recruited and randomly allocated to group A(ginger-indirect moxibustion plus acupuncture)or group B(acupuncture alone).The study consisted of a treatment period of 8 weeks with a total of 24 treatments(3 sessions per week,every other day),and a follow-up period of 12 *** outcome was measured by Fatigue Severity Scale(FSS),Psychological Health Report(SPHERE),the Self-rating depression scale(SDS)and the Hamilton anxiety scale(HAMA)at baseline,2,4,6,8,12 and 20 ***:With the treatment undergoing,the changes of FSS,SPHERE,SDS and HAMA scores in both groups increased gradually,and the effect maintained at the 12th *** groups,significantly higher score changes were seen in group A in FSS after 4 weeks treatment(11.94 vs 9.12,95%CI:0.94,4.7)and in SPHERE after 2 weeks treatment(3.7 vs 2.27,95%CI:0.56,2.31).But for SDS and HAMA,the improvement did not differ significantly between *** severe adverse events were ***:Ginger-indirect moxibustion is a safe and effective intervention to relieve fatigue and accompanying physical symptoms of CFS.

Punicalagin ameliorates collagen-induced arthritis by downregulating M1 macrophage and pyroptosis via NF-κB signaling pathway
