Objective: To investigate the effects of asperuloside on cervical cancer based on endoplasmic reticulum(ER) stress and mitochondrial pathway. Methods: Different doses(12.5–800 μg/mL) of asperuloside were used to treat cervical cancer cell lines Hela and CaSki to calculate the half maximal inhibitory concentration(IC_(50)) of asperuloside. The cell proliferation was analyzed by clone formation assay. Cell apoptosis, intracellular reactive oxygen species(ROS) and mitochondrial membrane potential were determined by flow cytometry. The protein expressions of cleaved-caspase-3, Bcl-2, Bax, Cyt-c, cleaved-caspase-4 and glucose-regulated protein 78(GRP78) were analyzed by Western blot. And the inhibitor of ER stress, 4-phenyl butyric acid(4-PBA) was used to treat cervical cancer cells to further verify the role of ER stress in the apoptosis of cervical cancer cells induced by asperuloside. Results: Asperuloside of 325, 650, and 1300 μg/mL significantly inhibited the proliferation and promoted apoptosis of Hela and CaSki cells(P<0.01). All doses of asperuloside significantly increased intracellular ROS levels, reduced mitochondrial membrane potential, significantly reduced Bcl-2 protein expression level, and increased Bax, Cyt-c, GRP78 and cleaved-caspase-4 expressions(P<0.01). In addition, 10 mmol/L 4-PBA treatment significantly promoted cell proliferation and reduced apoptosis(P<0.05), and 650 μg/mL asperuloside could reverse 4-PBA-induced increased cell proliferation, decreased apoptosis and cleaved-caspase-3,-4 and GRP78 protein expressions(P<0.05). Conclusion: Our study revealed the role of asperuloside in cervical cancer, suggesting that asperuloside promotes apoptosis of cervical cancer cells through ER stress-mitochondrial pathway.

A new technology called in vivo glia-to-neuron conversion has emerged in recent years as a promising next generation therapy for neural regeneration and repair. This is achieved through reprogramming endogenous glial cells into neurons in the central nervous system through ectopically expressing neural transcriptional factors in glial cells. Previous studies have been focusing on glial cells in the grey matter such as the cortex and striatum, but whether glial cells in the white matter can be reprogrammed or not is unknown. To address this fundamental question, we express NeuroD1 in the astrocytes of both grey matter(cortex and striatum) and white matter(corpus callosum) to investigate the conversion efficiency, neuronal subtypes, and electrophysiological features of the converted neurons. We discover that NeuroD1 can efficiently reprogram the astrocytes in the grey matter into functional neurons, but the astrocytes in the white matter are much resistant to neuronal reprogramming. The converted neurons from cortical and striatal astrocytes are composed of both glutamatergic and GABAergic neurons, capable of firing action potentials and having spontaneous synaptic activities. In contrast, the few astrocyte-converted neurons in the white matter are rather immature with rare synaptic events. These results provide novel insights into the differential reprogramming capability between the astrocytes in the grey matter versus the white matter, and highlight the impact of regional astrocytes as well as microenvironment on the outcome of glia-toneuron conversion. Since human brain has large volume of white matter, this study will provide important guidance for future development of in vivo glia-to-neuron conversion technology into potential clinical therapies. Experimental protocols in this study were approved by the Laboratory Animal Ethics Committee of Jinan University(approval No. IACUC-20180321-03) on March 21, 2018.

Apricot has a long history of cultivation and has many varieties and types. The traditional variety identification methods are timeconsuming and labor-consuming, posing grand challenges to apricot resource management. Tool development in this regard will help researchers quickly identify variety information. This study photographed apricot fruits outdoors and indoors and constructed a dataset that can precisely classify the fruits using a U-net model (F-score:99%), which helps to obtain the fruit's size, shape, and color features. Meanwhile, a variety search engine was constructed, which can search and identify variety from the database according to the above features. Besides, a mobile and web application (ApricotView) was developed, and the construction mode can be also applied to other varieties of fruit trees.Additionally, we have collected four difficult-to-identify seed datasets and used the VGG16 model for training, with an accuracy of 97%, which provided an important basis for ApricotView. To address the difficulties in data collection bottlenecking apricot phenomics research, we developed the first apricot database platform of its kind (ApricotDIAP, http://***/) to accumulate, manage, and publicize scientific data of apricot.

The synergistic damage effect of irradiation and corrosion of reactor structural materials has been a prominent research focus.This paper provides a comprehensive review of the synergistic effects on the third-and fourth-generation fission nuclear energy structural materials used in pressurized water reactors and molten salt reactors.The competitive mechanisms of multiple influencing factors,such as the irradiation dose,corrosion type,and environmental temperature,are summarized in this paper.Conceptual approaches are proposed to alleviate the synergistic damage caused by irradiation and corrosion,thereby promoting in-depth research in the future and solving this key challenge for the structural materials used in reactors.

Objective:To explore the balance of peripheral blood T helper 17 cells/regulatory T cell(Th17/Treg)ratio and the polarization ratio of M1 and M2 macrophages in lower extremity arteriosclerosis obliterans(ASO).Methods:A rat model of lower extremity ASO was established,and blood samples from patients with lower extremity ASO before and after surgery were obtained.ELISA was used to detect interleukin 6(IL-6),IL-10,and IL-17.Real-time RCR and Western blot analyses were used to detect Foxp3,IL-6,IL-10,and IL-17 expression.Moreover,flow cytometry was applied to detect the Th17/Treg ratio and M1/M2 ratio.Results:Compared with the control group,the iliac artery wall of ASO rats showed significant hyperplasia,and the concentrations of cholesterol and triglyceride were significantly increased(Pminal angioplasty and arterial stenting),all these changes were significantly improved(P<0.05).Conclusions:The Th17/Treg and M1/M2 ratios were significantly increased in ASO,and surgery can effectively improve the balance of Th17/Treg,and reduce the ratio of M1/M2,and the expression of inflammatory factors.

BACKGROUND:The molecular mechanism of sepsis-associated acute kidney injury(SA-AKI)is unclear.We analyzed co-differentially expressed genes(co-DEGs)to elucidate the underlying mechanism and intervention targets of SA-AKI.METHODS:The microarray datasets GSE65682,GSE30718,and GSE174220 were downloaded from the Gene Expression Omnibus(GEO)database.We identified the co-DEGs and constructed a gene co-expression network to screen the hub genes.We analyzed immune correlations and disease correlations and performed functional annotation of the hub genes.We also performed single-cell and microenvironment analyses and investigated the enrichment pathways and the main transcription factors.Finally,we conducted a correlation analysis to evaluate the role of the hub genes.RESULTS:Interleukin 32(IL32)was identified as the hub gene in SA-AKI,and the main enriched signaling pathways were associated with hemopoiesis,cellular response to cytokine stimulus,inflammatory response,and regulation of kidney development.Additionally,IL32 was significantly associated with mortality in SA-AKI patients.Monocytes,macrophages,T cells,and NK cells were closely related to IL32 and were involved in the immune microenvironment in SA-AKI patients.IL32 expression increased significantly in the kidney of septic mouse.Toll-like receptor 2(TLR2)was significantly and negatively correlated with IL32.CONCLUSION:IL32 is the key gene involved in SA-AKI and is significantly associated with prognosis.TLR2 and relevant immune cells are closely related to key genes.

Applying an Improved Dung Beetle Optimizer Algorithm to Network Traffic Identification

Network traffic identification is critical for maintaining network security and further meeting various demands of network applications.However,network traffic data typically possesses high dimensionality and complexity,leading to practical problems in traffic identification data analytics.Since the original Dung Beetle Optimizer(DBO)algorithm,Grey Wolf Optimization(GWO)algorithm,Whale Optimization Algorithm(WOA),and Particle Swarm Optimization(PSO)algorithm have the shortcomings of slow convergence and easily fall into the local optimal solution,an Improved Dung Beetle Optimizer(IDBO)algorithm is proposed for network traffic identification.Firstly,the Sobol sequence is utilized to initialize the dung beetle population,laying the foundation for finding the global optimal solution.Next,an integration of levy flight and golden sine strategy is suggested to give dung beetles a greater probability of exploring unvisited areas,escaping from the local optimal solution,and converging more effectively towards a global optimal solution.Finally,an adaptive weight factor is utilized to enhance the search capabilities of the original DBO algorithm and accelerate convergence.With the improvements above,the proposed IDBO algorithm is then applied to traffic identification data analytics and feature selection,as so to find the optimal subset for K-Nearest Neighbor(KNN)classification.The simulation experiments use the CICIDS2017 dataset to verify the effectiveness of the proposed IDBO algorithm and compare it with the original DBO,GWO,WOA,and PSO algorithms.The experimental results show that,compared with other algorithms,the accuracy and recall are improved by 1.53%and 0.88%in binary classification,and the Distributed Denial of Service(DDoS)class identification is the most effective in multi-classification,with an improvement of 5.80%and 0.33%for accuracy and recall,respectively.Therefore,the proposed IDBO algorithm is effective in increasing the efficiency of traffic identification and solving the problem of the original DBO a
