Immunological effects of nano-enabled hyperthermia for solid tumors: opportunity and challenge

Compared to conventional hyperthermia that is limited by low selectivity and severe side effects,nano-enabled hyperthermia yields great potentials to tackle these limitations for cancer treatment.Another major advance is the observation of immunological responses associated with nano-enabled hyperthermia,which introduces a new avenue,allowing a potential paradigm shift from the acutely effective and cytotoxicity-centric response to the next-phase discovery,i.e.,long-lasting and/or systemic anti-tumor immunity.This perspective first discusses the temperature-gradient and the spatially-structured immunological landscape in solid tumors receiving nano-enabled hyperthermia.This includes the discussion about underlying mechanism such as immunogenic cell death,which initiates a profound immunological chain reaction.In order to propagate the immune activation as a viable therapeutic principle,we further discussed the tumor type-specific complexity in the immunological tumor microenvironment,including the creative design of nano-enabled combination therapy to synergize with nano-enabled hyperthermia.

Syringin is found in the root of Acanthopanax senticosus(Rupr.Maxim.)Harms and belongs to the lignin chemical compound with many biological activities.In this study,we employed commercially available starting materials and accomplished the total synthesis of syringin in 5 steps with an overall yield of 58%.Palladium-catalyzed C(O)–C bond activation and subsequent cross coupling reaction is the key to construct syringin and its natural analogues.

Submerged entry nozzle(SEN)clogging during continuous casting of Ti-stabilized ultra-pure ferritic stainless(Ti-UPFS)steels was systematically investigated via cross-sectional analysis and acid dissolution treatment.The SEN deposit profile was characterized as occurring in three major layers:(1)an eroded refractory layer;(2)an initial adhesive layer comprised an Al_(2)O_(3)-ZrO_(2) composite sub-layer and a dense Al_(2)O_(3)-based deposit sub-layer;and(3)a porous multiphase deposit layer mainly consisting of MgO·Al_(2)O_(3),CaO-Al_(2)O_(3),and CaO-TiOx.The MgO·Al_(2)O_(3)-rich inclusions did not adhere directly to the eroded refractory but were entrapped during the deposit growth.Results of inclusion characterization in the tundish revealed that the MgO·Al2O3-rich particles present in the tundish served as the primary source of clogging deposits.Furthermore,a novel cavity-induced adhesion model by circular approximation was established to explain the effects of complex inclusion characteristics and refractory material type on adhesion force.A high number of small MgO·Al_(2)O_(3) inclusions were expected to accelerate the buildup of clogging deposits.Improving the modification of MgO·Al_(2)O_(3)-rich inclusions in the size range of 2-4μm by Ca treatment was crucial to minimizing the risk of SEN clogging during the continuous casting of Ti-UPFS steels.

As well known, optimal consensus is nontrivial due to the mismatching between the whole performance optimization(which usually leads to a centralized protocol) and the given distributed communication topology [1].

In order to reveal the hotspots and trends of biological water treatment from the perspectives of scientific and technological innovation, both of the bibliometric review and patents analysis were performed in this study. The Web of Science Core Collection database and Derwent Innovation Index database recorded 30023 SCI papers and 50326 patents, respectively were analyzed via information visualization technology. The results showed that China ranked the first in both papers and patents, while the United States and Japan had advantages in papers and patents, respectively. It was concluded through literature metrology analysis that microbial population characteristics, biodegradation mechanism,toxicity analysis, nitrogen and phosphorus removal and biological treatment of micropolluted wastewater were the research hotspots of SCI papers. Activated sludge process and anaerobic-aerobic combined process were the two mainstream technologies on the basis of patent technology classification analysis. Technology evolution path of biological water treatment was also elucidated in three stages based on the citation network analysis. Furthermore, the future directions including research on the law of interaction and regulation of biological phases and pollutants and the technology innovations towards the targeted biotransformation or selective biodegradation of pollutants and resource reuse of wastewater were prospected.

Understanding the development,regeneration,and disorders of the liver is the major goal in liver biology.Current mechanistic knowledge of human livers has been largely derived from mouse models and cell lines,which fall short in recapitulating the features of human liver cells or the structures and functions of human livers.Organoids as an in vitro system hold the promise to generate organ-like tissues in a dish.Recent advances in human liver organoids also facilitate the understanding of the biology and diseases in this complex organ.Here we review the progress in human liver organoids,mainly focusing on the methods to generate liver organoids,their applications,and possible future directions.

The present study proposes a novel method based on the geometric theory for measuring the distribution of bubble swarms in the circular region of a direct-contact heat exchanger.It was determined that the mixing is uniform when the average distance between the bubble swarms in the unit circular region is approximately 0.9054,which is the standard reference value.The effect of sample size(i.e.,the number of bubbles)on mixing uniformity was investigated to determine the optimal sample size.It was verified that the metric's accuracy and stability were higher with a sample size of 155.Accordingly,it was proposed to increase the sample size by filling irregular bubbles using a segmentation method,which enabled a further accurate assessment of the mixing uniformity.The mixing uniformity of bubble swarms in the circular region and its maximum internal connection with the square region was accurately quantified.It was revealed that the relative average error increased by approximately 3.47% due to information loss.The proposed method was demonstrated to be rotationally invariant.The present study provided novel insights into evaluating mixing uniformity,which would guide enhanced heat transfer and the effective evaluation of the spatiotemporal characteristics of transient mixing in circular regions or the cross-sections of chemical transport pipelines.

In this work,we study the gradient projection method for solving a class of stochastic control problems by using a mesh free approximation ap-proach to implement spatial dimension approximation.Our main contribu-tion is to extend the existing gradient projection method to moderate high-dimensional space.The moving least square method and the general radial basis function interpolation method are introduced as showcase methods to demonstrate our computational framework,and rigorous numerical analysis is provided to prove the convergence of our meshfree approximation approach.We also present several numerical experiments to validate the theoretical re-sults of our approach and demonstrate the performance meshfree approxima-tion in solving stochastic optimal control problems.

Objective:To explore the mechanism of effects of total saponin fraction from Dioscorea Nipponica Makino(TSDN)on M1/M2 polarization of monocytes/macrophages and arachidonic acid(AA)pathway in rats with gouty arthritis(GA).Methods:Seventy-two Sprague Dawley rats were randomly divided into 4 groups(n=18 in each):normal,model,TSDN at 160 mg/kg,and celecoxib at 43.3 mg/kg.Monosodium urate crystal(MSU)was injected into the rats’ankle joints to induce an experimental GA model.Blood and tissue samples were collected on the 3rd,5th,and 8th days of drug administration.Histopathological changes in the synovium of joints were observed via hematoxylin and eosin(HE)staining.The expression levels of arachidonic acid(AA)signaling pathway were assessed via real-time polymerase chain reaction(q PCR)and Western blot.Flow cytometry was used to determine the proportion of M1 and M2 macrophages in the peripheral blood.An enzyme-linked immunosorbent assay(ELISA)was used to detect interleukine(IL)-1β,tumor necrosis factoralpha(TNF-α),IL-4,IL-10,prostaglandin E_(2)(PGE_(2)),and leukotriene B4(LTB4).Results:HE staining showed that TSDN improved the synovial tissue.q PCR and Western blot showed that on the 3rd,5th and 8th days of drug administration,TSDN reduced the m RNA and protein expressions of cyclooxygenase(COX)2,microsomal prostaglandin E synthase-1 derived eicosanoids(m PGES-1),5-lipoxygenase(5-LOX),recombinant human mothers against decapentaplegic homolog 3(Smad3),nucleotide-binding oligomerization domain-like receptor protein 3(NALP3),and inducible nitric oxide synthase(i NOS)in rats’ankle synovial tissues(P<0.01).TSDN decreased COX1 m RNA and protein expression on 3rd and 5th day of drug administration and raised it on the 8th day(both P<0.01).It lowered CD68 protein expression on days 3(P<0.01),as well as m RNA and protein expression on days 5 and 8(P<0.01).On the 3rd,5th,and 8th days of drug administration,TSDN elevated the m RNA and protein expression of Arg1 and CD163(P<0.01).Flow cytometry results showed that TSDN decreased
