Objective:To evaluate the quality of Arnebiae Radix(AR)and Dictamni Cortex(DC)and study the efficacy of herbal extracts of these two herbs on the treatment of allergic contact dermatitis(ACD).Methods:Qualitative and quantitative analysis of effective components was performed using High Performance Thin Layer Chromatography(HPTLC),High Performance Liquid Chromatography(HPLC),and HPLC-Quadrupole Time of Flight-Mass Spectrometry(HPLC-QTOF-MS).In vitro allergic ACD 3D model was established by incubating 3D reconstructed human epidermis(RHE)with skin sensitizer,potassium dichromate.A total of 65 gene expression that were associated with ACD,which included 24 antioxidant responsive element(ARE)and 41 SENS-IS genes were quantified by q *** than or equal to 10 ARE genes and 18 SENN-IS genes were induced by 1.3-fold,demonstrating the successful establishment of in vitro ACD *** extracts of AR and DC were applied on the in vitro ACD model to study the ***:Batch 3 of AR and batch 2 of DC showed presence of all active ingredients with the highest *** ingredients of the herbs were extracted using a special oil and formulated into herbal oil *** herbal oil extracts were able to down regulate the induced genes in the in-vitro ACD skin model,bringing the tissue back to homeostatic ***:The oil extracts showed the potent efficacy of using AR and DC in ACD *** combination study will be done to optimize the formulation ratio which will be developed into a topical cream.

Lithium-sulfur batteries(LSBs)have emerged as promising power sources for high-performance devices such as electric ***,the poor energy density of LSBs owing to polysulfide shuttling and passivation has limited their further market *** mitigate this challenge,two-dimensional(2D)siloxene(2DSi),a Si-based analog of graphene,is utilized as an additive for sulfur *** 2DSi is fabricated on a large scale by simple solvent extraction of calcium disilicide to form a thin-layered structure of Si planes functionalized with vertically aligned hydroxyl groups in the *** stoichiometric reaction of 2DSi with polysulfides generates a thiosulfate redox mediator,secures the intercalation pathway,and reveals Lewis acidic sites within the siloxene *** 2DSi utilizes the corresponding in-situ-formed electrocatalyst,the 2D confinement effect of the layered structure,and the surface affinity based on Lewis acid-base interaction to improve the energy density of 2DSi-based LSB *** with the commercial carbon-based current collector,2DSi-based LSB cells achieve a volumetric energy density of 612 Wh Lcell^(−1) at 1 mA cm^(−2) with minor degradation of 0.17%per cycle,which rivals those of state-of-the-art *** study presents a method for the industrial production of high-energy-dense LSBs.

The medical cost in an aging society increases substantially when elders have higher incidence of chronic diseases, disability and unable to live independently. Nowadays, the medical practice concerns not only living longer but also having a healthy lifestyle. Thus, how to simultaneously identify the association between the survival and long-term quality of life becomes an important issue. The two-stage model proposed by Tsiatis, De Gruttola and Wulfsohn(1995) and the joint model proposed by Wulfsohn and Tsiatis(1997) can be used to model a sequence of repeated continuous measures and survival jointly. However, when the repeated measurements are discrete, the generalized linear model for longitudinal data should be implemented. The paper proposes a modified two-stage model and a modified joint model for modeling survival time and the longitudinal ordinal data simultaneously. Besides the usual estimation procedure for the Cox model and generalized linear model, owing to some unobservable information in the model, some parameters in joint model have to be estimated by Monte Carlo EM algorithm and Metropolis-Hastings algorithm. Monte Carlo simulations are used to assess the performance of the proposed model based on the accuracy and precision of the estimates. A real data is used to illustrate the feasibility of the proposed model.

Aquaporin （AQP） is a water channel protein found in various subcellular membranes of both prokaryotic and eukaryotic cells. The physiological functions of AQPs have been elucidated in many organisms. However, understanding their biogenesis remains elusive, particularly regarding how they assemble into tetramers. Here, we investigated the amino acid residues involved in the tetramer formation of the Arabidopsis plasma membrane AQP AtPIP2;1 using extensive amino acid substitution mutagenesis. The mutant proteins V41A/ E44A, F51A/L52A, F87A/191A, F92A/193A, V95A/Y96A, and H216A/L217A, harboring alanine substitutions in the transmembrane （TM） helices of AtPIP2;1 polymerized into multiple oligomeric complexes with a vari- able number of subunits greater than four. Moreover, these mutant proteins failed to traffic to the plasma membrane, instead of accumulating in the endoplasmic reticulum （ER）. Structure-based modeling revealed that these residues are largely involved in interactions between TM helices within monomers. These results suggest that inter-TM interactions occurring both within and between monomers play crucial roles in tetramer formation in the AtPIP2;1 complex. Moreover, the assembly of AtPIP2;1 tetramers is critical for their trafficking from the ER to the plasma membrane, as well as water permeability.

AIM: To establish the diagnostic performance of sev-eral serological tests, individually and in combination, for diagnosing celiac disease (CD) in patients with different pretest probabilities, and to explore potential se- rological algorithms to reduce the necessity for biopsy. METHODS: We prospectively performed duodenal biopsy and serology in 679 adults who had either high risk (n = 161) or low risk (n = 518) for CD. Blood samples were tested using six assays (enzyme-linked immunosorbent assay) that detected antibodies to tissue transglutaminase (tTG) and deamidated gliadin peptide (DGP). RESULTS: CD prevalence was 39.1% in the high-risk population and 3.3% in the low-risk group. In high-risk patients, all individual assays had a high diagnostic efficacy [area under receiving operator characteristic curves (AU ROC): 0.968 to 0.999]. In contrast, assays had a lower diagnostic efficacy (AU ROC: 0.835 to 0.972) in the low-risk group. Using assay combinations, it would be possible to reach or rule out diagnosis of CD without biopsy in 92% of cases in both pretest populations. We observed that the new DGP/tTG Screen assay resulted in a surplus compared to more conventional assays in any clinical situation. CONCLUSION: The DGP/tTG Screen assay could be considered as the best initial test for CD. Combinations of two tests, including a DGP/tTG Screen, might be able to diagnose CD accurately in different clinical scenarios making biopsy avoidable in a high proportion of subjects.

Modern use of Chinese herbal formulae from Shang-Han Lun

Background The Chinese medical archive, Shang-Han Lun, is said to be written by ZHANG Zhong-jing （150-219 A.D.） This great influential work introduced the specific symptoms of six-channel disorders （Tai-Yang, Yang-Ming, Shao-Yang, Tai-Yin, Shao-Yin, and Jue-Yin） and their corresponding treatments, the combined syndromes, deterioration due to malpractice, and the concept of six-channel transitions. The concept of Shang-Han Lun is widely accepted by Chinese herbal doctors. However, no clinical data about Shang-Han symptoms are described in oriental or western medical reports. Methods The clinical prescription data of traditional Chinese medicine visits were extracted under the National Health Insurance in Taiwan. The application rate of 42 Shang-Han formulae in clinical practice was analyzed in detail with the software SPSS. Results Between 1999 and 2002, the prescription rate of Shang-Han formula was only 5.22% among a total of 528 889 576 Chinese herbal formula prescriptions. The most frequently used formula was Tai-Yang formulae （71.31%）, followed by Shao-Yang formulae （17.49%） and the most commonly prescribed individual Shang-Han formulae were Ge-Gen Tang （16.11%）, Shao-Yao-Gan-Cao Tang （12.97%）, Xiao-Qing-Long Tang （11.79%）, Ban-Xia Xie-Xin Tang （10.24%）, and Xiao-Chai-Hu Tang （9.11%）, which comprised 60.22% of the utilization rate of total Shang-Han formulae. Conclusions From the prescription patterns of Shang-Han formulae, there was no evidence of transitions among the six channels. Despite the fundamental role of Shang-Han Lun in traditional Chinese medicine, prescription of Shang-Han formulae was limited in clinical practice.

The intra-formational seal capability was characterized by multi-layered shale baffles to act as the primary or ultimate cap system, were taken into account by homogeneous and heterogeneous models, respectively. In this study, the relevant ranges of input parameter of porosity （20%-35%） and permeability （0.0001-1,000 mD）, respectively for the sandstone （storage layers） and shale （seals） have been selected, according to results of large amount of laboratory core, test from pilot 3,000 m drilling within Tai-Hsi Foreland Basin in Western Taiwan. By a comprehensive geological model, typical single well injected plume migration scenarios were stochastically simulated which covered a 20-year continuous injection （one Mt-CO2 per year） followed by a 1,000-year post-injection monitoring. The corresponding 2-D MHMD （maximum horizontal migration distances） of CO2 plume in the storage reservoir had been calculated with respect to heterogeneous reservoir models and their homogeneous counterparts. Moreover, the induced pressures at critical monitoring points above the injection zone were also evaluated. As a result, the shale baffles of intra-formational seal in a saline aquifers are proven to play a vital role, and capable of ensuring the safe carbon storage operation within a basin scale with a depth range about 2,100-2,500 mRT.
