Effect of nanosized NbC precipitates on hydrogen-induced cracking of high-strength low-alloy steel

We investigated the effect of nanosized NbC precipitates on hydrogen-induced cracking(HIC)of high-strength low-alloy steel by conducting slow-strain-rate tensile tests(SSRT)and performing continuous hydrogen charging and fracture *** results reveal that the HIC resistance of Nb-bearing steel is obviously superior to that of Nb-free steel,with the fractured Nb-bearing steel in the SSRT exhibiting a smaller ratio of elongation reduction(Iδ).However,as the hydrogen traps induced by NbC precipitates approach hydrogen saturation,the effect of the precipitates on the HIC resistance *** speculate that the highly dispersed nanosized NbC precipitates act as irreversible hydrogen traps that hinder the accumulation of hydrogen at potential crack nucleation *** addition,much like Nb-free steel,the Nb-bearing steel exhibits both H-solution strengthening and the resistance to HIC.

Skeletal muscle development is closely related with the amount of meat production and its quality in chickens. Natriuretic peptides(NPs) play an important role in myotube formation and fat oxidation of skeletal muscle in animals. The effect of C-type natriuretic peptide(CNP), an important member of the NPs, and its underlying molecular mechanisms in skeletal muscle are incompletely understood. Treatment of myoblasts with CNP led to enhanced proliferation/differentiation and significantly upregulated(P<0.05) m RNA expression of the CNP receptors natriuretic peptide receptor B(NPRB) and the clearance receptor C(NPRC). In cells exposed to CNP, 142 differentially expressed genes(84 up-regulation and 58 down-regulation)(P<0.05) were identified by RNA-sequencing compared with those in control cells. Sixteen genes were significantly enriched(P<0.05) in the metabolic pathway, and six of them(phospholipase C β4, phospholipase C β2, phosphoglycerate mutase 1, creatine kinase B, peroxiredoxin 6 and CD38) were closely related to skeletal muscle development and differentially expressed. In conclusion, CNP stimulated differentiation of myoblasts by upregulating expression of the NPRB and NPRC receptors and enriching key genes in the metabolic pathway.

The triggering mechanisms of debris flows were explored in the field using artificial rainfall experiments in two gullies, Dawazi Gully and Aizi Gully, in yunnan and Sichuan Provinces, China,respectively. The soils at both sites are bare, loose and cohesive gravel-dominated. The results of a direct shear test, rheological test and back-analysis using soil mass stability calculations indicate that the mechanisms responsible for triggering debris flows involved the decreases in static and dynamic resistance of the soil. The triggering processes can be divided into 7 stages: rainfall infiltration, generation of excess runoff, high pore water pressure, surface erosion, soil creep, soil slipping, debris flow triggering and debris flow increment. In addition, two critical steps are evident:(i) During the process of the soil mass changing from a static to a mobile state, its cohesion decreased sharply(e.g., the cohesion of the soil mass in Dawazi Gully decreased from 0.520 to0.090 k Pa, a decrease of 83%). This would have reduced the soil strength and the kinetic energy during slipping, eventually triggered the debris flow.(ii) When the soil mass began to slip, the velocity and the volume increment of the debris flow fluctuated as a result of the interaction of soil resistance and the sliding force. The displaced soil mass from the source area of the slope resulted in the deposition of a volume of soil more than 7-8 times greater than that in the source area.

This study was designed to analyze the effect of the mitochondrial respiratory pathways of Candida albicans （C. albicans） on the biofilm formation. The 2, 3-bis （2-methoxy- 4-nitro-5-sulfophenyl）-2H-tetrazolium-5-carboxanilide （XTT） reduction assay was used to measure the metabolic activities of biofilms formed by the C. albicans which were cultured in the presence of respiratory pathways inhibitors. The biofilms formed by the wide type （WT）, GOA1-deleted （GOA31）, GOAl-reconstituted （GOA32）, AOXla-deleted （AOX1） and AOX1b- deleted （AOX2） C. albicans strains were examined by the XTT reduction assay and fluorescence microscopy. The expression of adhesion-related genes BCR1, ALS1, ALS3, ECEI and HWP1 in the biofilms formed by the above five C. albicans strains was detected by real time polymerase chain reaction. It was found that the metabolic activity of biofilms formed by C. albicans was decreased in the presence of alternative oxidase inhibitor whereas it was increased in the presence of classical mitochondrial respiratory pathway complex Ⅲ or complex IV inhibitor. AOX1 strain produced scarce biofilms interspersed with few hyphal filaments. Moreover, no significant changes in the expression of BCR1 and ALS3 were observed in the AOX 1 strain, but the expression of ALS1 and ECE1 was down-regulated, and that of HWP1 was up-regulated. These results indicate that both AOX1 and AOX2 can promote the biofilm formation. However, AOX1a primarily plays a regulatory role in biofilm formation in the absence of inducers where the promoting effect is mainly achieved by promoting mycelial formation.

Extensive landslides were triggered by the Wenchuan earthquake(in China)on 12 May 2008,causing a tremendous amount of loose material suspended on the hillslopes,likely to be eroded and transported by *** is of great significance to study the long-term variation of the quantity and spatiotemporal distribution of loose deposits after a great earthquake in order to understand the restoration process,to assess the risk of future soil erosion,including geological hazards,and to further develop ecological governance *** this study,information about the multi-temporal loose deposits on the ranges of the Baisha River Basin,an alpine valley near the epicenter,was extracted by objectoriented remote sensing interpretation,and analysis on their spatiotemporal variation showed that the earthquake in 2008 resulted in loose deposits covering an additional area of 81.09 km2,with a volume estimated at 0.357 billion *** five years after the earthquake,the vegetation had recovered rapidly,and the extent of the deposits was significantly *** 5 to 13 years after the earthquake,the vegetation continued to recover but at a slower rate and a decreasing trend in newly formed deposits was *** total area,volume and quantity of the loose deposits gradually *** geometric mean center of the loose deposits gradually moved back towards the upstream area after the earthquake and,by 2021,the mean center was about 4 km away from its pre-earthquake *** the mean center of the loose deposits in 2008 as the center,the deposits in the downstream area shrank significantly after the earthquake,but collapses and landslides of deposits persisted in the upstream area to the west-southwest,west-northwest and northnorthwest from the mean center,indicating future source areas of new loose deposits in the basin.

Background: Peritoneal fibrosis is the primary reason that patients with end-stage renal disease (ESRD) have to cease peritoneal dialysis. Peritonitis caused by Gram-negative bacteria such as Escherichia coli (E. coli) were on the rise. We had previously shown that matrine inhibited the formation of biofilm by E. coli. However, the role of matrine on the epithelial-mesenchymal transition (EMT) in peritoneal mesothelial cells under chronic inflammatory conditions is still unknown. Methods: We cultured human peritoneal mesothelial cells (HPMCs) with lipopolysaccharide (LPS) to induce an environment that mimicked peritonitis and investigated whether matrine could inhibit LPS-induced EMT in these cells. In addition, we investigated the change in expression levels of the miR-29b and miR-129-5p. Results: We found that 10 jjig/ml of LPS induced EMT in HPMCs. Matrine inhibited LPS-induced EMT in HPMCs in a dose-dependent manner. We observed that treatment with matrine increased the expression of E-cadherin (F= 50.993, P< 0.01), and decreased the expression of alpha-smooth muscle actin (F= 32.913, P < 0.01). Furthermore, we found that LPS reduced the expression levels of miR- 29b and miR-129-5P in HPMCs, while matrine promoted the expression levels of miR-29b and miR-129-5P. Conclusions: Matrine could inhibit LPS-induced EMT in HPMCs and reverse LPS inhibited expressions of miR-29 b and miR-129- 5P in HPMCs, ultimately reduce peritoneal fibrosis. These findings provide a potential theoretical basis for using matrine in the prevention and treatment of peritoneal fibrosis.
