Experimental Study on the Mechanical Performance of Mortise-Tenon Joints Reinforced with Replaceable Flat-Steel Jackets

The mortise-tenon joint is an important hub transmitting and distributing external loads for load-bearing components(beams,columns et al.)in the ancient-timber frame structure ***,the conventional steel hoop reinforcement methods often insert wood screws into the timber *** the reinforced joint rotates greatly,the anchoring failure of the screws will cause damage to the timber *** solve this problem,this study proposes a detachable and replaceable non-destructive flat-steel jacket reinforcement method in which horizontal flat steel is placed in the center of the joint,and the bolt is extended to the outside of the timber *** 1:3.52 scaled straight-tenon joint specimens were subjected to monotonic loading of beam ends,including three unreinforced reference joints,three joints with flat-steel jacket and three carbon fiber-reinforced plastic(CFRP)reinforced *** mechanical behaviors of the novel joints with flat-steel jacket were experimentally studied by comparing with those of the joints without strengthening and retrofitted with CFRP,based on the failure modes,the initial stiffness,the ultimate bearing capacity,and the moment-rotation relationship *** indicated that the mortise-tenon joints reinforced with flat-steel jackets maintain the original semi-rigid properties of the unreinforced mortise-tenon joints and can effectively prevent the tenon from pulling *** initial stiffness and ultimate bearing capacity were improved *** column and beam of the reinforced joints remain intact providing the reference for the practical application of joints *** mortisetenon joints reinforced with CFRP lose the semi-rigid properties of the unreinforced tenon-mortise *** joints reinforced with CFRP have the largest initial stiffness,while the unreinforced joints have the lowest *** initial stiffness of the proposed joint is in between the joints reinforced with CFRP and unreinforced joint

Dual-calibration coefficient:a more accurate protocol for simultaneous determination of superoxide and hydrogen peroxide in human Hep G2 cell extracts

We present a more accurate method for the quantification of superoxide anion （02-） and hydrogen peroxide （H202） simulta- neously in human HepG2 cell extracts. After the xanthine/xanthine oxidase system was added into cell extract which was devoid of O2＊- and H202, steady-state and in-situ produced O2＊- and H202 by xanthine/xanthine oxidase system was labeled by fluorescent probes and subsequently separated by microchip electrophoresis. Based on this method, two differential equations with the calibration coefficients were established for O2 and H202, respectively. Using the established dual-calibration coefficients, we obtained the calibrated concentrations of 02＊ and H202 that produced in human HepG2 cells, which were lower （0.66±0.03 and 0.82±0.04 lamol/L for 02＊-and H202, respectively） than that （0.85±0.03 and 0.96±0.03 gmol/L for O. and H202, respectively） obtained from statutory working curve. The proposed dual-calibration coefficient protocol takes into account both the complex matrix effect of the biological system and real time decaying of O2 ＊- and H202, providing a method with higher accuracy.

Many flaviviruses are significant human pathogens causing considerable disease burdens,including encephalitis and hemorrhagic fever,in the regions in which they are endemic.A paucity of treatments for flaviviral infections has driven interest in drug development targeting proteins essential to flavivirus replication,such as the viral *** viral replication,the flavivirus genome is translated as a single polyprotein precursor,which must be cleaved into individual proteins by a complex of the viral protease,NS3,and its cofactor,*** this cleavage is an obligate step of the viral life-cycle,the flavivirus protease is an attractive target for antiviral drug *** this review,we will survey recent drug development studies targeting the NS3 active site,as well as studies targeting an NS2B/NS3interaction site determined from flavivirus protease crystal structures.

Effects of selective cyclooxygenase-2 inhibitor on C6 glioma cell proliferation and apoptosis

BACKGROUND： Studies have shown that cyclooxygenase-2 is associated with proliferation and apoptosis of glioma cells. OBJECTIVE： To investigate the effects of selective cyclooxygenase-2 inhibitor celecoxib on proliferation and apoptosis of C6 glioma cells in vitro. DESIGN, TIME AND SETTING： A cellular, molecular, controlled study was performed at the Central Laboratory and Room of Electron Microscope, Medical School, Xi＇an Jiaotong University, China from March 2007 to March 2008. MATERIALS： C6 glioma cells during in vitro log phase were assigned to control and experimental groups. Celecoxib （Pfizer, USA）, dimethyl sulfoxide （Sigma, USA）, and MTT （Sigma, USA） were used for this study. METHODS： The control group was subdivided into blank control and dimethyl sulfoxide control groups. C6 glioma cells in the blank control and dimethyl sulfoxide control groups were incubated in Dulbecco＇s modified Eagle＇s medium supplemented with 10% calf serum and 0.3% dimethyl sulfoxide respectively. C6 glioma cells in the experimental group were separately treated with 60, 80 and 100 μmol/L celecoxib. MAIN OUTCOME MEASURES： Activity of C6 glioma cells was examined by MTT assay. C6 glioma cell cycle and apoptosis were determined by annexin V-fluorescein isothiocyanate/propidium iodide double-staining, followed by flow cytometry. Morphology and ultrastructure of C6 glioma cells were observed with an inverted microscope and a transmission electron microscope, respectively. RESULTS： Compared with the blank control group, cell density was reduced, adherence ability weakened, and irregular nuclei were visible, with the presence of chromatin condensation, margination, and some apoptotic bodies in the experimental group. Activity of C6 glioma cells was significantly decreased （P 〈 0.05）, cell number was reduced during S phase, cell number was significantly increased during G2/M phase （P 〈 0.01 ）, and the apoptotic rate was significantly increased （P 〈 0.05） in the experimental group. Silk fibroin(SF)is considered biocompatible and biodegradable for osteochondral ***,it lacks a bioactive domain for cell adhesion,proliferation and differentiation,limiting its therapeutic *** revamp SF as a biomimicking and bioactive microenvironment to regulate cell behaviours,we engineered an elastin-like polypeptide(ELP,Val-Pro-Gly-Xaa-Gly)to modify SF fibers via simple and green dehydrothermal(DHT)*** results demonstrated that the ELP successfully bound to SF,and the scaffold was reinforced by the fusion of the silk fiber intersections with ELP(S-ELP-DHT)via the DHT *** bone mesenchymal stem cells(BMSCs)and chondrocytes exhibited improved spreading and proliferation on the S-ELP-DHT *** ex vivo and in vivo experiments further demonstrated enhanced mature bone and cartilage tissue formation using the S-ELP-DHT scaffolds compared to the naked SF *** results indicated that a recombinant ELPmodified silk scaffold can mimic three-dimensional(3D)cell microenvironment,and improve bone and cartilage *** envision that our scaffolds have huge clinical potential for osteochondral repair.

Flavivirus RNA cap methyltransferase:structure,function,and inhibition

Many flaviviruses are significant human *** plus-strand RNA genome of a flavivirus contains a 5′terminal cap 1 structure(m7GpppAmG).The flavivirus encodes one methyltransferase(MTase),located at the Nterminal portion of the NS5 RNA-dependent RNA polymerase(RdRp).Here we review recent advances in our understanding of flaviviral capping machinery and the implications for drug *** NS5 MTase catalyzes both guanine N7 and ribose 2'-OH methylations during viral cap *** flavivirus MTases,from dengue,yellow fever,and West Nile virus(WNV),sequentially generate GpppA!m7GpppA!*** the existence of two distinct methylation activities,the crystal structures of flavivirus MTases showed a single binding site for S-adenosyl-L-methionine(SAM),the methyl *** finding indicates that the substrate GpppA-RNA must be repositioned to accept the N7 and 2'-O methyl groups from SAM during the sequential *** studies demonstrated that distinct RNA elements are required for the methylations of guanine N7 on the cap and of ribose 2'-OH on the first transcribed *** enzymes with different methylation defects can trans complement one another in vitro,demonstrating that separate molecules of the enzyme can independently catalyze the two cap methylations in *** the context of the infectious virus,defects in both methylations,or a defect in the N7 methylation alone,are lethal to ***,viruses defective solely in 2'-O methylation are attenuated and can protect mice from later wild-typeWNV *** results demonstrate that the N7 methylation activity is essential for the WNV life cycle and,thus,methyltransferase represents a novel and promising target for flavivirus therapy.
