Metabolomics reveals the key role of oxygen metabolism in heat susceptibility of an alpine-dwelling ghost moth, Thitarodes xiaojinensis (Lepidoptera: Hepialidae)

Ghost moths inhabiting the alpine meadows of the Tibetan Plateau are cold? adapted stenothermal organisms that are susceptible to heat (dead within 7 days at 27 °C exposure). Exploring the metabolic basis of their heat susceptibility would extend our understanding of the thermal biology of alpine-dwelling invertebrates. Here, gas chromatography-mass spectrometry-based metabolomics was combined with physiological and transcriptional approaches to determine the metabolic mechanisms of heat susceptibility in Thitarodes xiaojinensis larvae. The metabolomics results showed that 27 °C heat stress impaired the Krebs cycle and lipolysis in T. xiaojinensis larvae, as demonstrated by the accumulation of intermediary metabolites. In addition, carbohydrate reserves were highly and exclusively consumed, and an anaerobic product, lactate, accumulated. This evidence suggested a strong reliance on glycolysis to anaerobically generate energy. The respiration rate and enzymatic activity test results indicated a deficiency in O2 metabolism;in addition, the Krebs cycle capacity was not decreased, and the metabolic flux through aerobic pathways was limited. These findings were further supported by the occurrence of hypoxia symptoms in midgut mitochondria (vacuolation and swelling) and increased transcription of hypoxia-induced factor 1-α.Overall, heat stress caused O2 limitation and depressed the overall intensity of aerobic metabolism in ghost moths, and less efficient anaerobic glycolysis was activated to sustain their energy supply. As carbohydrates were depleted, the energy supply became deficient. Our study presents a comprehensive metabolic explanation for the heat susceptibility of ghost moths and reveals the relationship between O2 metabolism and heat susceptibility in these larvae.

Two-way shape memory effect in a Ti-Zr-Nb-Ta high-temperature shape memory alloy

The two-way shape memory effect in a Ti-18.5Zr-10Nb-3.5Ta high-temperature shape memory alloy was investigated.X-ray diffraction measurem ent shows that the alloy is composed of orthorhombicα"-martensite.ωphase is not found in Ti-18.5Zr-10Nb-3.5Ta alloy due to the suppressing effect of Ta element.Theα"-martensite laths are found in the transmission electron microscope observation;after the bending deformation,there appear a lot of dislocations.The alloy exhibits a shape memory strain of 3.8%aud a high reverse martensite transformation start temperature of 464 K.The maximum two-way shape memory strain of 1.2%is obtained in the alloy with the prebending training strain of 10%.The mechanism can be ascribed to the effect of internal stress field caused by dislocations.

BACKGROUND Colorectal cavernous hemangioma is a rare vascular malformation resulting in recurrent lower gastrointestinal hemorrhage,and can be misinterpreted as colitis.Surgical resection is currently the mainstay of treatment,with an emphasis on sphincter preservation.CASE SUMMARY We present details of two young patients with a history of persistent hematochezia diagnosed with colorectal cavernous hemangioma by endoscopic ultrasound(EUS).Cavernous hemangioma was relieved by several EUS-guided lauromacrogol injections and the patients achieved favorable clinical prognosis.CONCLUSION Multiple sequential EUS-guided injections of lauromacrogol is a safe,effective,cost-efficient,and minimally invasive alternative for colorectal cavernous hemangioma.

Female moths of cotton bollworm （Lepidoptera： Noctuidae） captured by waterbasin traps baited with synthetic female sex pheromone

Cotton bollworm, Helicoverpa armigera （Hüibner）（Lepidoptera： Noctuidae）, is one of the most important pest insects in cotton fields in China. Female moths were captured by waterbasin traps with a synthetic female sex pheromone blend in cotton fields over three years. The blend contained （Z）-11-hexadecenal and （Z）-9-hexadecenal with a ratio of 97：3. Each pheromone dispenser was impregnated with 2.0 mg of pheromone blend and 0.2 mg of antioxidant dissolved with 0.1 mL of hexane, and there was a control dispenser with a similar amount of antioxidant and solvent only. Waterbasin traps were deployed in three configurations in the fields. ‘A＇ was pheromone traps only, ‘B＇ was both pheromone and control traps, ‘C＇ was control traps only. （i） In four plots of ‘A＇, the average weekly female catch was 1.5, and more females were captured by centrally located pheromone traps. （ii） In three plots of‘B＇, control traps also captured female as well as male moths, but average weekly female catches of control traps was significantly lower than that in pheromone-baited traps. （iii） There were significant linear relationships between the average weekly female catch and the corresponding layer in pheromone-baited traps in both ‘A＇ and ‘B＇ plots, and in quadratic equations in control in ‘B＇ plots. （iv） With the increase of the interval of traps, average weekly female catches per trap increased but average weekly female catches per hectare decreased. （v） Among the female moths captured by pheromone traps, 88.3% were mated female moths which each containing 1.46 spermatophores, while in control traps 86.9% of the mated female moths had 0.90 spermatophores. There was a significant difference between the average numbers of spermatophores of mated females in pheromone traps and in controls.

A cascade of in situ conversion of bicarbonate to CO_(2) and CO_(2) electroreduction in a flow cell with a Ni-N-S catalyst

Combination of CO_(2) capture using inorganic alkali with subsequently electrochemical conversion of the resultant HCO_(3)^(-)to high-value chemicals is a promising route of low cost and high efficiency.The electrochemical reduction of HCO_(3)^(-)is challenging due to the inaccessible of negatively charged molecular groups to the electrode surface.Herein,we adopt a comprehensive strategy to tackle this challenge,i.e.,cascade of in situ chemical conversion of HCO_(3)^(-)to CO_(2) and CO_(2) electrochemical reduction in a flow cell.With a tailored Ni-N-S single atom catalyst(SACs),where sulfur(S)atoms located in the second shell of Ni center,the CO_(2)electroreduction(CO_(2)ER)to CO is boosted.The experimental results and density functional theory(DFT)calculations reveal that the introduction of S increases the p electron density of N atoms near Ni atom,thereby stabilizing^(*)H over N and boosting the first proton coupled electron transfer process of CO_(2)ER,i.e.,^(*)+e^(-)+^(*)H+^(*)CO_(2)→^(*)COOH.As a result,the obtained catalyst exhibits a high faradaic efficiency(FE_(CO)~98%)and a low overpotential of 425 mV for CO production as well as a superior turnover frequency(TOF)of 47397 h^(-1),outcompeting most of the reported Ni SACs.More importantly,an extremely high FECOof 90%is achieved at 50 mA cm^(-2)in the designed membrane electrode assembly(MEA)cascade electrolyzer fed with liquid bicarbonate.This work not only highlights the significant role of the second coordination on the first coordination shell of the central metal for CO_(2)ER,but also provides an alternative and feasible strategy to realize the electrochemical conversion of HCO_(3)^(-)to high-value chemicals.

Laser manufacturing of spatial resolution approaching quantum limit

Atomic and close-to-atom scale manufacturing is a promising avenue toward single-photon emitters,single-electron transistors,single-atom memory,and quantum-bit devices for future communication,computation,and sensing applications.Laser manufacturing is outstanding to this end for ease of beam manipulation,batch production,and no requirement for photomasks.It is,however,suffering from optical diffraction limits.Herein,we report a spatial resolution improved to the quantum limit by exploiting a threshold tracing and lock-in method,whereby the two-order gap between atomic point defect complexes and optical diffraction limit is surpassed,and a feature size of<5 nm is realized.The underlying physics is that the uncertainty of local atom thermal motion dominates electron excitation,rather than the power density slope of the incident laser.We show that the colour centre yield in hexagonal boron nitride is transformed from stochastic to deterministic,and the emission from individual sites becomes polychromatic to monochromatic.As a result,single colour centres in the regular array are deterministically created with a unity yield and high positional accuracy,serving as a step forward for integrated quantum technological applications.

Macrophage senescence,manifested by the special form of durable cell cycle arrest and chronic low-grade inflammation like senescence-associated secretory phenotype,has long been consid-ered harmful.Persistent senescence of macrophages may lead to maladaptation,immune dysfunction,and finally the development of age-related diseases,infections,autoimmune diseases,and malignancies.However,it is a ubiquitous,multi-factorial,and dynamic complex phenomenon that also plays roles in remodeled processes,including wound repair and embryogenesis.In this review,we summarize some general molecular changes and several specific biomarkers during macrophage senescence,which may bring new sight to recognize senescent macrophages in different conditions.Also,we take an in-depth look at the functional changes in senescent macrophages,including metabolism,autophagy,polarization,phagocytosis,antigen presentation,and infiltration or recruitment.Furthermore,some degenerations and diseases associated with senescent macrophages as well as the mechanisms or relevant genetic regulations of senescent macrophages are integrated,not only emphasizing the possibility of regulating macrophage senescence to benefit age-associated diseases but also has an implication on the finding of potential tar-gets ordrugs clinically.

Confinement of ionomer for electrocatalytic CO2 reduction reaction via efficient mass transfer pathways

Gas diffusion electrodes（GDEs） mediate the transport of reactants, products and electrons for the electrocatalytic CO2reduction reaction（CO2RR） in membrane electrode assemblies. The random distribution of ionomer, added by the traditional physical mixing method, in the catalyst layer of GDEs affects the transport of ions and CO2. Such a phenomenon results in elevated cell voltage and decaying selectivity at high current densities. This paper describes a pre-confinement method to construct GDEs with homogeneously distributed ionomer, which enhances mass transfer locally at the active centers. The optimized GDE exhibited comparatively low cell voltages and high CO Faradaic efficiencies（FE > 90%） at a wide range of current densities. It can also operate stably for over 220 h with the cell voltage staying almost unchanged. This good performance can be preserved even with diluted CO2feeds, which is essential for pursuing a high single-pass conversion rate. This study provides a new approach to building efficient mass transfer pathways for ions and reactants in GDEs to promote the electrocatalytic CO2RR for practical applications.
