AZD1775 synergizes with SLC7A11 inhibition to promote ferroptosis

Tumor suppressor p53-mediated cell cycle arrest and DNA damage repair may exert cytoprotective effects against cancer therapies, including WEE1 inhibition. Considering that p53 activation can also lead to multiple types of cell death, the role of this tumor suppressor in WEE1 inhibitor-based therapies remains disputed. In this study, we reported that nucleolar stress-mediated p53 activation enhanced the WEE1 inhibitor AZD1775-induced ferroptosis to suppress lung cancer growth. Our findings showed that AZD1775 promoted ferroptosis by blocking cystine uptake, an action similar to that of Erastin. Meanwhile, inhibition of WEE1 by the WEE1 inhibitors or si RNAs induced compensatory upregulation of SLC7A11, which conferred resistance to ferroptosis. Mechanistically, AZD1775 prevented the enrichment of H3K9me3, a histone marker of transcriptional repression, on the SLC7A11 promoter by repressing the expression of the histone methyltransferase SETDB1, thereby enhancing NRF2-mediated SLC7A11 transcription. This finding was also validated using the H3K9me3 inhibitor BRD4770. Remarkably, we found that the nucleolar stress-inducing agent Actinomycin D(Act. D) inhibited SLC7A11 expression by activating p53, thus augmenting AZD1775-induced ferroptosis. Moreover, the combination of AZD1775 and Act. D synergistically suppressed wild-type p53-harboring lung cancer cell growth both in vitro and in vivo. Altogether, our study demonstrates that AZD1775 promotes ferroptosis by targeting cystine uptake but also mediates the adaptive activation of SLC7A11 through the WEE1-SETDB1 cascade and NRF2-induced transcription, and inhibition of SLC7A11 by Act. D boosts the anti-tumor efficacy of AZD1775 by enhancing ferroptosis in cancers with wild-type p53.

Common wheat is an important and widely cultivated food crop throughout the *** progress has been made in regard to wheat genome sequencing in the last *** from the sequencing of single chromosomes/chromosome arms whole genome sequences of common wheat and its diploid and tetraploid ancestors have been decoded along with the development of sequencing and assembling technologies. In this review, we give a brief summary on international progress in wheat genome sequencing, and mainly focus on reviewing the effort and contributions made by Chinese scientists.

Effect of sintering temperature on the microstructure and properties of foamed glass-ceramics prepared from high-titanium blast furnace slag and waste glass

Foamed glass-ceramics were prepared via a single-step sintering method using high-titanium blast furnace slag and waste glass as the main raw materials The influence of sintering temperature(900–1060℃) on the microstructure and properties of foamed glass-ceramics was studied. The results show that the crystal shape changed from grainy to rod-shaped and finally turned to multiple shapes as the sintering temperature was increased from 900 to 1060℃. With increasing sintering temperature, the average pore size of the foamed glass-ceramics increased and subsequently decreased. By contrast, the compressive strength and the bulk density decreased and subsequently increased. An excessively high temperature, however, induced the coalescence of pores and decreased the compressive strength. The optimal properties, including the highest compressive strength(16.64 MPa) among the investigated samples and a relatively low bulk density(0.83 g/cm^3), were attained in the case of the foamed glass-ceramics sintered at 1000℃.

Structural, optical, and thermal properties of MAX-phase Cr2AlB2

First-principles calculations of the structural, optical, and thermal properties of Cr2AlB2 are performed using the pseudopotential plane-wave method within the generalized gradient approximation （GGA）. Calculation of the elastic constant and phonon dispersion indicates that Cr2AlB2 is mechanically and thermodynamically stable. Analysis of the band structure and density of states indicates that Cr2AlB2 is metallic. The thermal properties under increasing temperature and pressure are investigated using the quasi-harmonic Debye model. The results show that anharmonic effects on Cr^AlB~ are important at low temperature and high pressure. The calculated equilibrium primitive cell volume is 95.91 ~3 at T = 300 K, P - 0 GPa. The ability of Cr2AlB2 to resist volume changes becomes weaker with increasing temperature and stronger with increasing pressure. Analysis of optical properties of Cr2AlB2 shows that the static dielectric function of Cr2AlB2 is 53.1, and the refractive index no is 7.3. If the incident light has a frequency exceeding 16.09 eV, which is the plasma frequency of Cr2AlB2, Cr2AlB2 changes from metallic to dielectric material.

Designer cell-self-implemented labeling of microvesicles in situ with the intracellular-synthesized quantum dots

Cell-derived microvesicles(MVs) are secreted from almost all kinds of mammalian cells into the extracellular space, and play crucial roles in intercellular communication and transporting biomolecules between cells. However, there is a great challenge for visualizing and monitoring of MVs’ bio-behaviors due to the limitations of existing labeling methods. Herein, we report the first paradigm of designer cell-self-implemented labeling of MVs secreted from living mammalian MCF-7 cells in situ using the intracellular-synthesized fluorescent quantum dots(QDs) during the formation of MVs. By elaborately coupling intracellular biochemical reactions and metabolism pathways, the MCF-7 cells can be illuminated brightly by intracellular-biosynthesized fluorescent CdSe QDs. Simultaneously, intracellular-synthesized QDs can be in situ encapsulated by the secreted MVs budding from the plasma membrane of the fluorescing cells to label the MVs with an efficiency of up to 89.9%. The whole labeling process skillfully combines designer precise cell-tuned intricate synthesis of CdSe QDs with mild in-situ labeling via cell-selfimplementation just after feeding the cell with suitable chemicals, which is structure-or function-nondestructive and much more straightforward and milder than those by chemical conjugation or indirect encapsulation with conventional fluorogenic labels.

Influence of CeO_2 addition on the preparation of foamed glass-ceramics from high-titanium blast furnace slag

Foamed glass-ceramics doped with cerium oxide（CeO_2）were successfully prepared from high-titanium blast furnace slag by one-step *** influence of CeO_2 addition（1.5wt%–3.5wt%）on the crystalline phases,microstructure,and properties of foamed glass-ceramics was *** show that CeO_2 improves the stability of the glass phase and changes the two-dimensional crystallization mechanism into three-dimensional *** analysis indicates the presence of Ca（Mg,Fe）Si_2O_6 and Ca（Ti,Mg,Al）（Si,Al）_2O_（6 ）in all sintered *** with CeO_2,Ti CeO_4 precipitates,and crystallinity increases,leading to increased thickness of pore walls and uniform *** comprehensive properties of foamed glass-ceramics are better than that of samples without CeO_*** particular,the sample added with a suitable amount of CeO_2（2.5wt%）exhibits bulk density that is similar to and compressive strength（14.9 MPa）that is more than twice of foamed glass-ceramics without CeO_2.

With the low-lying energy levels, E2 transition branching ratios and absolute transition rates of the 134Ba and 108Pd, are investigated in the neutron-proton interacting boson model (IBM2) which includes the quadrupole quadrupole interaction between like bosons and the E(5) symmetry, it shows that the IBM2 can describe the nuclei at critical point of a phase transition well.

External wall structure of green rural houses in Daqing, China, based on life cycle and ecological footprint theories

Daqing is situated in one of the severely cold resions of China. The livin8 environment in this region is extremely poor because of the harsh climate and the backward economy. The external wall is an important component of the external envelope of buildings, and it greatly contributes to the indoor thermal environment. By taking the external wall as the research object, this study summarizes the characteristics of the external wall structure and analyzes the common materials used in existing rural residences. Specifically, we combine life cycle theory and ecological footprint （EF） theory and introduce the green external wall structure, as well as its application in practice, in accordance with the local ecosystem. Results show that anecotosical residence offers a better environment and greater economic benefits than a traditional residence. The annual energy consumption, C02 consumption, and EF of the ecological residence in this study are lower than those of the traditional residence by 69.61%, 17.5 t, and 99.47%, respectively.

Analyzing the Value of Location Information for Four Taxi Dispatching Modes

Location-based service (LBS) is often quoted as one of the killer applications of mobile commerce. The LBS, especially the GPS dispatching system, is often adopted by taxi operators for fleet management, but there is no theoretical analysis on the real value of location information for dispatching services. This paper presents four types of dispatching modes and how location information is obtained and used to support dispatching services. Then the Pros and Cons of different taxi dispatching modes were compared. It is concluded that different mode fits different situations, no one overwhelms all situation, even if the GPS-based dispatching mode. The value of the GPS-based dispatching service then was further analyzed through the case study of DaZhong Company in Shanghai, China. The results show that the use of the up-to-date GPS technology radically changes the traditional approaches to taxi dispatching and enables companies to enhance customers'satisfaction.

Structural Design of a Tall Building with Underpinning Structure in Guangzhou

This paper introduces the structural design optimization of a tall building with reinforced concrete frame-core shear walls in Guangzhou *** decrease the construction cost,wide flat beam with 400 mm ×650 mm rectangular cross-section are adopted for main *** measures are used to decrease noncoincident centers of mass and *** subway tunnels cross the building underground,inclined columns,the second and third floor underground together function as the underpinning *** from traditional transform beam,the proposed underpinning structures show good performance without using large dimensional bending *** attentions should be paid to the beams connecting with the inclined column in the underpinning structures,and these beams show large tensile or compressive *** observations after completion construction indicate that the building as well as the proposed design measures work well.
