BACKGROUND Although the role of p53 in the evolution and prognosis of gastric cancer(GC)has been extensively examined,the exact mechanism of action is incompletely *** the last years,p53-target genes were supposed to be involved in the p53 *** of them is the tumor-suppressor gene Maspin,which codifies the protein with the same *** activity depends on its subcellular *** our knowledge,the possible role of TP53 gene in Maspin subcellular localization,in GC cells,has not yet been studied in a large number of human *** To evaluate the possible role of wild-type and mutated p53 in Maspin subcellular *** The present study included 266 consecutive patients with GC in which TP53 gene status,and mutations in exons 2 to 11,respectively,were analyzed and correlated with immunohistochemical expression of p53 and *** None of the 266 cases showed mutations in exon *** rate of TP53 mutations was 33.83%.The mutation rate was slightly higher in distally-located GCs,with a lower degree(≤5 buds/high power fields)of dyscohesivity(P<0.01).The wildtype cases had a longer survival,compared with mutant GCs,especially in patients without lymph node metastases,despite the high depth of tumor infiltration(P=0.01).The Dukes-MAC-like staging system was proved to have the most significant independent prognostic value(P<0.01).The statistical correlations proved that TP53 gene mutations in exon 7 might induce knockdown of Maspin,but wild-type p53 can partially restore nuclear Maspin expression and decrease the metastatic potential of gastric adenocarcinoma *** Downregulated Maspin might be induced by mutations in exon 7 of the TP53 gene but wild-type p53 can partially restore nuclear Maspin *** findings should be proved in experimental studies.

Ever since its discovery two decades ago,the erythro- poietin-producing hepatoma (EPH)-EPHRIN system has been shown to play multifaceted roles in human gastroenterological cancer as well as ***,amplif ication and point mutations have been found in many human cancers and many investigators have shown correlations between these up-regulationsand tumor ***,the genes in this family are considered to be potential targets of cancer *** the other hand,the down-regulation of some members as a result of epigenetic changes has also been reported in some ***,the correlation between altered expressions and clinical prognosis seems to be inconclusive.A huge amount of protein-protein interaction studies on the EPH-EPHRIN system have provided a basic scheme for signal transductions,especially bi-directional signaling involving EPH-ERPHRIN molecules at the cell *** information also provides a manipulative strategy for harnessing the actions of these *** this review,we summarize the known alterations of EPH-EPHRIN genes in human tumors of the esophagus,stomach,colorectum,liver and pancreas and present the perspective that the EPH-EPHRIN system could be a potential target of cancer therapy.

Efficacy of mindfulness-based meditation therapy on the sense of coherence and mental health of nurses

The primary goal of the study was to assess the efficacy of mindfulness-based meditation therapy on the psychological well-being and sense of coherence of nurses with respect to coping with stress. The participants were 28 nurses working in a ward for elderly patients, with 15 included in the intervention group and 13 in the control group. Nurses in the intervention group received two sessions of mindfulness-based meditation therapy, including breathing, yoga movement and meditation. Each nurse was taught the program in the first session, then exercised at home with a CD, and subsequently met with an interviewer in a second session after two weeks. Nurses in the control group did not receive any interventions. The participants completed the General Health Questionnaire (GHQ), including subdomains of physical symptoms, anxiety and sleep disturbance, interference with social activities, and depression;and the Sense of Coherence (SOC) questionnaire, which includes three subdomains of comprehensibility, manageability, and meaningfulness pre- and post-intervention. The GHQ scores of the intervention group showed a significantly greater decrease than those of the control group. The SOC scores of the intervention group showed a significantly greater increase after the intervention compared to those in the control group, with the highest score for the meaningfulness subdomain. These results indicate that mindfulness-based meditation therapy is effective for improving the psycho-logical well-being and sense of coherence of nurses, which helps them to cope with stress.

A predual of a predual of B_σ and its applications to commutators

A predual of B_σ-spaces is investigated. A predual of a predual of B_σ-spaces is also investigated,which can be used to investigate the boundedness property of the commutators. The relation between Herz spaces and local Morrey spaces is discussed. As an application of the duality results, one obtains the boundedness of the singular integral operators, the Hardy-Littlewood maximal operators and the fractional integral operators, as well as the commutators generated by the bounded mean oscillation(BMO) and the singular integral *** is new in this paper is that we do not have to depend on the specific structure of the operators. The results on the boundedness of operators are formulated in terms of B_σ-spaces and B_σ-spaces together with the detailed comparison of the ones in Herz spaces and local Morrey spaces. Another application is the nonsmooth atomic decomposition adapted to B_σ-spaces.

Shear wave elastography(SWE)is now becoming an indispensable diagnostic tool in the routine examination of liver *** particular,accuracy is required for shear wave propagation velocity measurement,which is directly related to diagnostic *** is generally accepted that the liver shear wave propagation velocity reflects the degree of fibrosis,but there are still few reports on other factors that increase the shear wave propagation *** this study,we reviewed such factors in the literature and examined their *** SWE measures propagation velocity based on the assumption that the medium has a homogeneous structure,uniform density,and is purely ***,the measurement is subject to *** other(confounding)factors that we routinely experience are primarily:(1)Conditions that appear to increase the viscous component;and(2)Conditions that appear to increase tissue ***,the former includes acute hepatitis,congested liver,biliary obstruction,etc,and the latter includes diffuse infiltration of malignant cells,various storage diseases,tissue necrosis,*** any case,it is important to evaluate SWE in the context of the entire clinical picture.

Tumor biology in estrogen receptor-positive,human epidermal growth factor receptor type 2-negative breast cancer:Mind the menopausal status

Breast cancer is not one disease,but can be categorized into four major molecular subtypes according to hormone receptor [estrogen receptor(ER) and progesterone receptor(Pg R)] and human epidermal growth factor receptor type 2(HER2) expression status. Ki67 labeling index and/or multigene assays are used to classify ERpositive,HER2-negative breast cancer into luminal A and luminal B(HER2-negative) subtypes. To date,most studies analyzing predictive or prognostic factors in ER-positive breast cancer have been performed in postmenopausal women,mainly using patients and samples in adjuvant aromatase inhibitor trials. In contrast,even the clinical roles of Pg R and Ki67 have been little analyzed so far in premenopausal women. Pg R is one of the estrogen-responsive genes,and it has been reported that plasma estradiol levels are related to expression levels of estrogen-responsive genes including PGR in ER-positive breast cancer. In this article,biological differences,especially differences in expression of Pg R and Ki67 in ER-positive breast cancer between pre- and postmenopausal women are discussed. Clinical roles of Pg R and Ki67 in ER-positive breast cancer differ between pre- and postmenopausal women. We suggest that the mechanisms of development and estrogen-dependent growth of ER-positive breast cancer might differ according to menopausal status.

Moxibustion Promotes Formation of Granulation in Wound Healing Process through Induction of Transforming Growth Factor-β in Rats

Gamete fusion is a core process of sexual reproduction and, in both plants and animals, different sex gametes fuse within species. Although most of the molecular factors involved in gamete interaction are still unknown in various sex-possessing eukaryotes, reports of such factors in algae and land plants have been increasing in the past decade. In particular, knowledge of gamete interaction in flowering plants and green algae has increased since the identification of the conserved gamete fusion factor GENERATIVE CELL SPECIFIC 1/HAPLESS 2 （GCS1/HAP2）. GCSl was first identified as a pollen generative cell-specific transmembrane protein in the lily （Lilium Iongiflorum）, and was then shown to function not only in flowering plant gamete fusion but also in various eukaryotes, including unicellular protists and metazoans. In addition, although initially restricted to Chlamydomonas, knowledge of gamete attachment in flowering plants was also acquired. This review focuses on recent progress in the study of gamete interaction in volvocine green algae and flowering plants and discusses conserved mechanisms of gamete recognition, attachment, and fusion leading to zygote formation.

Intermittent compressive force induces cell cycling and reduces apoptosis in embryoid bodies of mouse induced pluripotent stem cells

In vitro manipulation of induced pluripotent stem cells(iPSCs) by environmental factors is of great interest for three-dimensional(3D) tissue/organ induction. The effects of mechanical force depend on many factors, including force and cell type. However,information on such effects in i PSCs is lacking. The aim of this study was to identify a molecular mechanism in i PSCs responding to intermittent compressive force(ICF) by analyzing the global gene expression profile. Embryoid bodies of mouse i PSCs, attached on a tissue culture plate in 3D form, were subjected to ICF in serum-free culture medium for 24 h. Gene ontology analyses for RNA sequencing data demonstrated that genes differentially regulated by ICF were mainly associated with metabolic processes,membrane and protein binding. Topology-based analysis demonstrated that ICF induced genes in cell cycle categories and downregulated genes associated with metabolic processes. The Kyoto Encyclopedia of Genes and Genomes database revealed differentially regulated genes related to the p53 signaling pathway and cell cycle. q PCR analysis demonstrated significant upregulation of Ccnd1, Cdk6 and Ccng1. Flow cytometry showed that ICF induced cell cycle and proliferation, while reducing the number of apoptotic cells. ICF also upregulated transforming growth factor β1(Tgfb1) at both m RNA and protein levels, and pretreatment with a TGF-β inhibitor(SB431542) prior to ICF abolished ICF-induced Ccnd1 and Cdk6 expression. Taken together,these findings show that TGF-β signaling in i PSCs enhances proliferation and decreases apoptosis in response to ICF, that could give rise to an efficient protocol to manipulate i PSCs for organoid fabrication.
