Oxytocin in pig seminal plasma is positively related with in vivo fertility of inseminated sows

Background:Identification of relevant in vivo biomarkers for fertility remains a challenge for the livestock *** of the small peptide hormone oxytocin(OXT),involved in male reproductive function and present in the seminal plasma(SP)of several species could be a robust *** study characterized concentrations of SPOXT in ejaculates from boars used in artificial insemination(AI)programs aiming to evaluate its relationship with sperm quality variables and in vivo fertility of their liquid-stored *** OXT concentrations(ng/mL)were measured in 169 ejaculates from 61 boars of the Duroc,Pietrain,Landrace and Large White breeds using a direct competitive immunoassay test based on AlphaLISA®***(ejaculate volume,sperm concentration,total sperm count)and sperm parameters(motility,viability,intracellular generation of reactive oxygen species,plasma membrane fluidity)were assessed at 0 h and 72 h in AI-semen samples stored at 17℃.In vivo fertility included only 18 Large White and Landrace boars whose AI-semen was used to inseminated>100 sows and evaluated both farrowing rate and litter size of 3,167 ***:The results showed that SP-OXT differed between boars and between ejaculates within boar(P<0.05)but not between breeds(Duroc,Pietrain,Landrace and Large White).Ejaculates with higher SP-OXT concentration/mL(hierarchically grouped;P<0.001)had larger volume and came from younger boars(P<0.05).Ejaculates of boars showing positive farrowing rate deviation exhibited higher(P<0.05)SP-OXT concentration/mL than those with negative farrowing rate ***:The SP concentrations of OXT are boar,ejaculate and age dependent,and positively related with ejaculate volume and farrowing rates of liquid-stored semen AI-doses.

Background Proteome characterization of the porcine endometrium and extraembryonic membranes is important to understand mother-embryo *** this study,the proteome of the endometrium and cho-rioallantoic membrane was characterized in pregnant sows(PS)during early gestation(d 18 and 24 of gestation)and in the endometrium of non-pregnant sows(NPS)during the same days using LC-MS/MS *** UniProtKB database and ClueGO were used to obtain functional Gene Ontology annotations and biological and functional networks,*** Our analysis yielded 3,254 and 3,457 proteins identified in the endometrium of PS and NPS,respectively;of these,1,753 being common while 1,501 and 1,704 were exclusive to PS and NPS,*** addition,we iden-tified 3,968 proteins in the extraembryonic membranes of *** analyses of function revealed some proteins had relevance for the immune system process and biological adhesion in endometrium while the embryonic chorion displayed abundance of proteins related to cell adhesion and cytoskeletal organization,suggesting they dominated the moment of endometrial remodeling,implantation and adhesion of the lining *** are available via Pro-teomeXchange with identifier *** This is the first in-depth proteomic characterization of the endometrium and extraembryonic mem-branes during weeks 3 to 4 of gestation;data that contribute to the molecular understanding of the dynamic environ-ment during this critical period,associated with the majority of pregnancy losses.

Men becoming fathers by injection were more often age intracytoplasmic sperm born small for gestational

Being born with nonoptimal birth characteristics decreases the chance of becoming a father. Urogenital malformations as well as metabolic syndrome are more common in men born small for gestational age （SGA） and could be contributing factors to the reduced fertility rate seen in these men. It could imply that men becoming fathers by assisted reproductive technology （ART） more often are born with low birth weight （LBW）, preterm, and/or SGA than men conceiving without treatment and also that men where intracytoplasmic sperm injection （ICSI） had to be performed more often are born with nonoptimal birth characteristics than men where conventional in vitro fertilization （IVF） successfully could be used. In this retrospective, case-control study using Swedish national registers, we compared the birth characteristics of 1206 men who have become fathers by ART with a control group consisting of age-matched men who became fathers without treatment. The differences in birth characteristics between men becoming fathers by IVF and ICSI were also assessed. For men becoming fathers by ART, OR of being born with LBW was 1.66 （95% CI = 1.17-2.36） compared with fathers who conceived without treatment. OR of being born prematurely was 1.32 （95% CI = 1.00-1.77）. Men becoming fathers via ICSI had a doubled increased likelihood of being born SGA compared with men who became fathers via IVF （OR = 2.12; 95% CI = 1.17-3.83）. In conclusion, we have found that men becoming fathers by ICSI treatments had more often been born SGA than men becoming fathers by conventional IVF.

Colloid Centrifugation of Semen: Applications in Assisted Reproduction

Colloid centrifugation can be used to separate heterogeneous populations of cells particularly semen samples, which typically contain mixtures of spermatozoa at different stage of maturity together with dead and dying spermatozoa and also non-sperm cells. Colloids can be used to enrich the population of spermatozoa that are viable and functional;these are the spermatozoa that are needed for fertilization in Assisted Reproduction. Sperm samples obtained after colloid centrifugation may be enriched for motile, morphologically normal spermatozoa with intact chromatin. Insemination of mares with stallion sperm samples prepared by colloid centrifugation resulted in a higher pregnancy rate per cycle than controls, confirming that the good sperm attributes observed in the laboratory were indicative of functional spermatozoa. The method does not simply enrich for viable spermatozoa by inhibiting dead or dying spermatozoa from passing through the colloid since emerging evidence shows that colloid centrifugation can select for spermatozoa with certain properties such as metabolic activity. Other reproductive cells, such as spermatids or progenitor cells, may also be purified by colloid centrifugation. This review describes the history of colloid centrifugation for selecting spermatozoa, compares colloid centrifugation with alternative techniques, and finally describes some of the applications of the technique within the animal breeding industry.

Flow cytometry is now a recognized methodology within animal spermatology, and has moved from being a research tool to become routine in the assessment of animal semen destined to breeding. The availability of ＇bench-top＇ flow cytometers and of newer and versatile markers for cell structure and function had allowed the instrumentation to measure more sperm parameters, from viability to reactiveness when exposed to exogenous stimuli, and to increase our capabilities to sort spermatozoa for potential fertilizing capacity, or chromosomal sex. The present review summarizes the state of the art regarding flow cytometry applied to animal andrology, albeit keeping an open comparative intent. It critically evaluates the present and future capabilities of flow cytometry for the diagnostics of potential fertility and for the development of current reproductive technologies such as sperm freezing, sperm selection and sperm sorting. The flow cytometry methods will probably further revolutionize our understanding of the sperm physiology and their functionality, and will undoubtedly extend its application in isolating many uncharacterized features of spermatozoa. However, continuous follow-up of the methods is a necessity owing to technical developments and the complexity of mapping spermatozoa.

Energy model based on solar potential and the production of electrical energy through living plants’photosynthesis

Developing a sustainable energy model is imperative considering the current trend towards decarbonizing sectors *** this purpose,Venezuela was used as a reference to propose an energy model focused on taking advantage of plant photosynthesis through microbial-vegetable fuel cells together with an agro-photovoltaic system to enhance energy and agricultural *** production from the cells was estimated using an average power density of 264 mW/m^(2) over 4%of the areas destined for crops in the entire Venezuelan region,obtaining an annual production of 19.889 GWh/*** contrast,the energy production of the agro-photovoltaic system was modelled using PVsyst software on 50%of the area used for the cells distributed throughout the states of Anzoátegui,Guárico,Monagas and Portuguesa according to their meteorological conditions,solar irradiation and agricultural activity,resulting in 3703417 GWh/*** resulting whole system proved to be able to cover>10 times the installed electricity generation capacity at a national level and,together with the tremendous scalability of the microbial fuel cells,it shows that Venezuela has a high potential for the production and distribution of clean energy.

Plant-microbial fuel cell for electrical generation through living plants: an internal resistance insight into the plant species used

Climate change is a global threat,the presence of which has encouraged the development and implementation of renewable energies,including plant–microbial fuel cells,which could generate 6629568–33147840 MWh per year due to their large-scale *** of the main challenges associated with microbial cells is internal resistance—a parameter whose magnitude is influenced by sev-eral *** the case of plant–microbial fuel cells,adding a plant positively affects the mitigation of internal ***,the species employed is expected to play an important *** the present study,the objective was to determine as a general reference the internal resistance of cells using various plant species,among which were tomato(Solanum lycopersicum),black beans(Phaseolus vulgaris),aloe vera(Aloe vera),corn(Zea mays),and moss(Dicranidae).For this purpose,an experimental procedure was carried out to compare the manual voltage measurements with a free cell concerning the voltage obtained by adding an external resistor in *** internal resistance values were determined as a function of the measured voltage,obtaining 177,179,175,324,and 233 kΩfor aloe vera,corn,tomato,black beans,and moss,*** this,it was shown that the roots of the plants represent an essen-tial addition to the internal resistance of the cell in the short *** contrast,plants with a C4 photosynthetic metabolism are more favourable,while C3 plants can also benefit from internal resistance during a more extended period of rhizodeposition.

Age-adjusted NT-proBNP could help in the early identification and follow-up of children at risk for severe multisystem inflammatory syndrome associated with COVID-19(MIS-C)

BACKGROUND Multisystem inflammatory syndrome in children(MIS-C) has emerged as a new disease associated with COVID-19 that presents in acute critically ill children with acute cardiovascular *** To determine whether the age-adjusted N-terminal pro-brain natriuretic peptide(NT-proBNP) value(Z-log-NT-proBNP) is associated with severe MIS-C and myocardial *** A retrospective study was conducted which included children with MIS-C managed at our institution between April 1,2020,and February 28,*** divided the population into groups depending on severity based on pediatric intensive care unit(PICU) *** compared Z-log-NT-proBNP values across these groups and analyzed Z-log-NT-proBNP dynamics during the onemonth *** We included 17 participants [median age 3(2-9) years] and seven(41%) required PICU ***(100%) of these cases presented very high(Z-log > 4) levels of NT-proBNP at the time of admission compared to only 5(50%) patients with non-severe MIS-C(P = 0.025).NT-proBNP was significantly correlated with highsensitive Troponin I levels(P = 0.045),Ross modified score(P = 0.003) and left ventricle ejection fraction(P = 0.021).CONCLUSION Raised NT-proBNP,specifically very high values(Z-log-NT-proBNP > 4) could help in the early identification of MIS-C patients with myocardial dysfunction requiring inotropic support and PICU admission.

Growing energy demand is a current problem with greater incidence in the *** research objective is to establish seed bases to give a leading role to plants used in anthropogenic production activities and consumption,and even plants belonging to wildlife in terms of energy *** use of living plants to obtain electrical energy through a microbial-vegetable fuel cell is proposed as a mech-anism to supply the energy demand with easy access worldwide,especially in areas far from urban *** this work,the voltage produced by microbial-vegetable fuel cells was measured using graphite electrodes and plants belonging to the species of aloe(Aloe vera),corn(Zea mays),black beans(Phaseolus vulgaris),tomato(Solanum lycopersicum)and moss(Dicranidae).They were selected for their presence in everyday life and their great commercial and agricultural value,reaching a maximum voltage of 884 mV from *** is evidence that all plants can be a source of energy to a greater or lesser extent and that their performance is given by the plant root interactions at the ***,an extensive root system can favour the accumulation of substrate after the hydrolysis of dead roots,provided that their presence does not interfere with microbial activity on the anodic *** this,using plants with low resistance to thermal stress can lead to a substrate richer in substances that can be ***,it is recommended to use plants according to their life cycle and the morphology of their roots,and plants that can grow in waterlogged conditions or without direct contact with the Sun.

Influence of metal support in-plane symmetry on the corrugation of hexagonal boron nitride and graphene monolayers

Predicting the properties of two-dimensional （2D） materials as graphene and hexagonal boron nitride （h-BN） monolayers after their growth on any given substrate is a major challenge. While the influence of the electron configuration of the atoms of the underlying surface is well-understood, the effect of substrate geometry still remains unclear. The structural properties of h-BN monolayers grown on a rectangularly packed Rh（110） surface were characterized in situ by ultrahigh vacuum scanning tunneling microscopy and were compared to those that this material exhibits when grown on substrates showing different crystallographic orientations. Although the h-BN monolayer grown on Rh（110） was dominated by a unique quasiunidimensional moir6 pattern, suggesting considerable interface interaction, the moir6 corrugation was unexpectedly smaller than those reported for strongly interacting interfaces with hexagonal-terminated substrates, owing to differences in the possible binding landscapes at interfaces with differently oriented substrates. Moreover, a rule was derived for predicting how interface corrugation and the existence and extent of subregions within moir6 supercells containing favorable sites for orbital mixing between h-BN monolayers and their supports depend on substrate symmetry. These general symmetry considerations can be applied to numerous 2D materials, including graphene, thereby enabling the prediction of how substrate choice determines the properties of these materials. Furthermore, they could also provide new routes for tuning 2D material properties and for developing nanotemplates showing different geometries for Krowing adsorbate superlattices.
