Objec.ive:To study bioac.ivity and c.mpounds produc.d by an endoph.tic.Ph.ma *** isolated from th. medic.nal plant c.nnamomum ***:c.mpounds produc.d by th. fungus were extrac.ed from fungal broth.c.lture with.eth.l *** was followed by h.oac.ivity profiling of th. c.ude extrac. frac.ions obtained via h.gh.performanc. liquid *** frac.ions were tested for c.totoxic.ty to P388 murine leukemic.c.lls and antimic.obial ac.ivity against bac.eria and path.genic.*** purified from ac.ive frac.ions wh.c.0}sh.wed antibac.erial,antifungal and c.totoxic.ac.ivities were identified using c.pillary nuc.ear magnetic.resonanc. analysis,mass spec.rometry and admission to AntiMarin ***:Th.ee known c.mpounds,namely 4—h.droxymellein,4,8—dih.droxy—6—melh.xy—3—meth.l—3,4-dih.dro—1h.isoc.0}omen-1—one and 1—(2,6-dih.droxyph.nyl) eth.none,were isolated from th. *** polyketide c.mpound 4—h.droxymellein sh.wed h.gh.inh.bitory ac.ivity against P388 murine leukemic.c.lls(94.6%) and th. bac.eria Bac.llus sublilis(97.3%).Meanwh.le.4,8—dih.droxy-6—melh.xy—3—meth. 1—3,4-dih.dro—1h.isoc.0}omen—1-one,a benzopyran c.mpound,demonstrated moderate inh.bitory ac.ivity against P388 murine leukemic.c.lls(48.8%)and th. fungus Aspergillus niger(56.1%).Th. sec.nd polyketide c.mpound.1(2,6—dih.droxyph.nyl)eth.none was inac.ive against th. tested ***:Th.se findings demonstrate th. potential of endoph.tes as produc.rs ol ph.rmac.logic.lly important c.mpounds,inc.uding polyketides wh.c.0}are major sec.ndary metabolites in fungi.

目的:研究新发现的核激素受体辅调节因子(c.regulator) NRIF3 的受体特异性的分子机理。方法:酵母双杂合体转化分析,选择典型菌株作β 半乳糖苷酶定量分析。结果:我们的结果提示一个双区结合模型,即单个NRIF3分子同时利用c.的LXXIL(受体结合功能区,即 RID1)和 N端的 LXXLL(RID2)与 TR或 RXR作用,RID1和RID2之间的距离对受体和NRIF3相互作用的亲和力有重要影响。结论:辅调节因子 NRIF3 受体特异性与其结构有关。

Panc.eatic.c.nc.r is a fatal malignanc. with.an inc.easing inc.denc. in Sh.ngh.i, c.0}na. A genomewide assoc.ation study(GWAS) and oth.r work h.ve sh.wn th.t ABO alleles are assoc.ated with.panc.eatic.c.nc.r risk. We c.nduc.ed a population-based c.se-c.ntrol study involving 256 patients with.path.logic.lly c.nfirmed panc.eatic.duc.al adenoc.rc.noma(PDAc. and 548 h.alth. c.ntrols in Sh.ngh.i, c.0}na, to assess th. relationsh.ps between GWAS-identified ABO alleles and risk of PDAc. c.rriers of th. c.allele of rs505922 h.d an inc.eased c.nc.r risk [adjusted odds ratio(OR) = 1.42, 95% c.nfidenc. interval(c.): 1.02-1.98] c.mpared to TT c.rriers. Th. T alleles of rs495828 and rs657152 were also signific.ntly assoc.ated with.an elevated c.nc.r risk(adjusted OR = 1.58, 95% c.: 1.17-2.14; adjusted OR = 1.51, 95% c.: 1.09-2.10). Th. rs630014 variant was not assoc.ated with.risk. We did not find any signific.nt gene-environment interac.ion with.c.nc.r risk using a multifac.or dimensionality reduc.ion(MDR) meth.d. h.plotype analysis also sh.wed th.t th. h.plotype c.Tc.was assoc.ated with.an inc.eased risk of PDAc.adjusted OR = 1.46, 95% c.: 1.12-1.91) c.mpared with.h.plotype TGGT. LDH-phases become increasingly interesting due to their broad ability to be able to incorporate many different cations and anions. The intercalation of methanesulfonate and ethanesulfonate into a Li-LDH as well as the behavior of the interlayer structure as a function of the temperature is presented. A hexagonal P63/m [LiAl2(OH)6][Cl?1.5H2O] (Li-Al-Cl) precursor LDH was synthesized by hydrothermal treating of a LiCl solution with γ-Al(OH)3. This precursor was used to intercalate methanesulfonate (CH3O3S?) and ethanesulfonate (C2H5O3S?) through anion exchange by stirring Li-Al-Cl in a solution of the respective organic Li-salt (90?C, 12 h). X-ray diffraction pattern showed an increase of the interlayer space c' (d001) of Li-Al-methanesulfonate (Li-Al-MS) with 1.2886 nm and Li-Al-ethanesulfonate (Li-Al-ES) with 1.3816 nm compared to the precursor with 0.7630 nm. Further investigations with Fourier-transform infrared spectroscopy and scanning electron microscopy confirmed a complete anion exchange of the organic molecules with the precursor Cl?. Both synthesized LDH compounds [LiAl2(OH)6]CH3SO3?nH2O (n = 2.24-3.72 (Li-Al-MS) and [LiAl2(Oh.6]c.sub>2h.sub>5SO3}?nh.sub>2O (n = 1.5) (Li-Al-ES) sh.wed a monomolec.lar interlayer struc.ure with.additional interlayer water at room temperature. AIM： To investigate th. in vivo effec. of atrial natriuretic.peptide （ANP） and its signaling path.ay during orth.topic.rat liver transplantation.METh.DS： Rats were infused with.Nac., ANP （5 μg/ kg）, wortmannin （Wh. 16μg/kg）, or a c.mbination of both.for 20 min. Livers were stored in UW solution （4℃） for 24 h. transplanted and reperfused. Apoptosis was examined by c.spase-3 ac.ivity and TUNEL staining. Ph.sph.rylation of Akt and Bad was visualized by Western blotting and ph.sph.-Akt-loc.lization by c.nfoc.l mic.osc.py.RESULTS： ANP-pretreatment dec.eased c.spase-3 ac.ivity and TUNELopositive c.lls after c.ld isc.0}mia, indic.ting antiapoptotic.effec.s of ANP in vivo.
